Dermatosparaxis is a recessively inherited connective-tissue disorder that results from lack of the activity of type I procollagen N-proteinase, the enzyme that removes the amino-terminal propeptides from type I procollagen. Initially identified in cattle more than 20 years ago, the disorder was subsequently characterized in sheep, cats, and dogs. Affected animals have fragile skin, lax joints, and often die prematurely because of sepsis following avulsion of portions of skin. We recently identified two children with soft, lax, and fragile skin, which, when examined by transmission electron microscopy, contained the twisted, ribbon-like collagen fibrils characteristic of dermatosparaxis. Skin extracts from one child contained collagen precursors with amino-terminal extensions. Cultured fibroblasts from both children failed to cleave the amino-terminal propeptides from the pro alpha 1(I) and pro alpha 2(I) chains in type I procollagen molecules. Extracts of normal cells cleaved to collagen, the type I procollagen synthesized by cells from both children, demonstrating that the enzyme, not the substrate, was defective. These findings distinguish dermatosparaxis from Ehlers-Danlos syndrome type VII, which results from substrate mutations that prevent proteolytic processing of type I procollagen molecules.  相似文献

The in vitro conversion of thyroxine (T4) to triiodothyronine (T3) was studied in liver homogenates from fed and fasted lean and obese Zucker rats. T3 generation was decreased in fed young (2 month) obese rats as compared to values in fed lean controls. This was not corrected by the addition of dithiothreitol (DTT), suggesting a deficiency in 5'-deiodinase activity in young obese rats. Both lean and obese 2 month old rats responded to a 2 day fast by decreasing hepatic T3 generation as is always observed in other strains of rats. The hepatic conversion rate was not decreased in older (5 month) fed obese rats when compared to age-matched lean controls. Hepatic conversion of T4 to T3 was markedly decreased in 5 month old lean Zucker rats fasted for 4 days. In contrast, a 4 day fast had no effect on the hepatic conversion rate in the 5 month old obese rats. The hepatic conversion rate was assessed in 5 month old obese rats fasted for up to 28 days and hepatic conversion still did not decrease. This paradoxical response of the 5 month old obese rat may provide a new model to further evaluate the control of hepatic T3 generation from T4.  相似文献

Rats given d-amphetamine (15 mg/kg i.p.) or apomorphine (10 mg/kg i.p.) and placed in a cold environment (4°C) developed marked hypothermia. After daily injections of either drug for seven weeks, the maximal hypothermic responses to d-amphetamine or apomorphine were reduced to 72% and 19% of those obtained initially. Subsequent injection of ET-495, a central dopamine receptor stimulant, caused rectal temperature to decrease only 72% and 49% as much as in control animals. The hypothermic response to apomorphine was also depressed in d-amphetamine-treated animals. These observations suggest that the tolerance to the hypothermic effects of both d-amphetamine and apomorphine that develops is due at least in part to alterations in the sensitivity of dopamine receptors.  相似文献

Prosolin is a major cytosolic phosphoprotein expressed prominently in rapidly proliferating human peripheral lymphocytes but produced at very low levels in resting (G0) PBL. It undergoes rapid phosphorylation upon treatment of growing cells with tumor-producing phorbol esters (TPA) and this phosphorylation event is correlated with a rapid down-regulation of DNA synthesis. In the present report we have studied various agents that, like TPA, act as partial or complete mitogens for G0 PBL and have determined their effect on phosphorylation of prosolin and on DNA synthesis in rapidly proliferating (IL-2-dependent) human PBL. Agents that activate the TCR (OKT3 and PHA), as well as agents that by-pass the receptor but activate biochemical pathways associated with TCR activation (TPA and Ca2(+)-ionophore), all produced rapid phosphorylation of prosolin and prompt down-regulation of DNA synthesis. Four phosphorylated forms of prosolin were produced, indicating activation of a complex phosphorylation pathway. Down-regulation of DNA synthesis did not lead to cell death or to permanent arrest, but was reversed after 24 to 48 h, and was not associated with any reduction in overall protein synthesis. Agents that bind to determinants closely connected to the TCR but without activating it (OKT4 and OKT8) had no effect on either prosolin phosphorylation or DNA synthesis. The results indicate that prosolin is an early target of the protein kinase activities induced by activation of the TCR in proliferating PBL, and suggest that its phosphorylation mediates the TCR signal, transmitting it into a biochemical pathway leading specifically to down-regulation of DNA synthesis. In G0 PBL, in which the negligible expression of prosolin precludes significant production of phosphorylated species, this inhibitory pathway is effectively blocked.  相似文献

The C. elegans germline is pluripotent and mitotic, similar to self-renewing mammalian tissues. Apoptosis is triggered as part of the normal oogenesis program, and is increased in response to various stresses. Here, we examined the effect of endoplasmic reticulum (ER) stress on apoptosis in the C. elegans germline. We demonstrate that pharmacological or genetic induction of ER stress enhances germline apoptosis. This process is mediated by the ER stress response sensor IRE-1, but is independent of its canonical downstream target XBP-1. We further demonstrate that ire-1-dependent apoptosis in the germline requires both CEP-1/p53 and the same canonical apoptotic genes as DNA damage-induced germline apoptosis. Strikingly, we find that activation of ire-1, specifically in the ASI neurons, but not in germ cells, is sufficient to induce apoptosis in the germline. This implies that ER stress related germline apoptosis can be determined at the organism level, and is a result of active IRE-1 signaling in neurons. Altogether, our findings uncover ire-1 as a novel cell non-autonomous regulator of germ cell apoptosis, linking ER homeostasis in sensory neurons and germ cell fate.  相似文献

The family Xeniidae (Octocorallia) constitutes an abundant benthic component on many Indo-West Pacific coral reefs and is ecologically important in the Red Sea. The genus Ovabunda Alderslade, 2001 was recently established to accommodate previous Xenia species with sclerites comprised of a mass of minute corpuscle-shaped microscleres. The aim of the present study was to examine type material of Xenia species in order to verify their generic affiliation. We present here a comprehensive account of the genus Ovabunda, using scanning electron microscopy to depict sclerite microstructure. We assign three Xenia species to the genus: O. ainex comb. n., O. gohari comb. n., and O. crenata comb. n.; and synonymize several other species of Ovabunda. We provide a key to Ovabunda species and conclude that they are mainly confined to the Red Sea, with some occurrence in the West Indian Ocean.  相似文献

Eph receptors and their ephrin ligands are important mediators of cell-cell communication. They are divided in two subclasses based on their affinities for each other and on sequence conservation. Receptor-ligand binding within each subclass is fairly promiscuous, while binding cross the subclasses happens rarely. EphA4 is an exception to this general rule, since it has long been known to bind both A- and B-class ephrin ligands but the reason for this exceptional behavior has not been worked out at molecular level. Recent structural and biochemical studies on EphA4 ligand-binding domain alone and in complex with its ligands have addressed this question. However, the published structures of EphA4/ephrin complexes differ considerably from each other and strikingly different explanations for the exceptional promiscuity of EphA4 were proposed. To address these contradictory findings, we have determined a crystal structure of the EphA4 ligand-binding domain at 2.3 Å resolution and show that the receptor has an unprecedented ability to exist in two very different, well-ordered conformations even in the unbound state. Our results suggest that the ligand promiscuity of the Ephs is directly correlated with the structural flexibility of the ligand-binding surface of the receptor.  相似文献

Rhizomelic chondrodysplasia punctata is a rare autosomal recessive disorder characterized by stippled epiphyses and rhizomelic shortening of the long bones. We report 3 subjects of rhizomelic chondrodysplasia punctata from India and thePEX7 mutations identified in them. The commonPEX7-L292X allele, whose high frequency is due to a founder effect in the northern European Caucasian population, was not identified in these patients. Instead, 2 novel alleles are described, including 64_65delGC, which was present on a singlePEX7 haplotype and could represent a common allele in the Indian population.  相似文献

A class of linearly perturbed discrete-time single species scramble competition models, like the Ricker map, is considered. Perturbations can be of both recruitment and harvesting types. Stability (bistability) is considered for models, where parameters of the map do not depend on time. For models with recruitment, the result is in accordance with Levin and May conjecture [S.A. Levin, R.M. May, A note on difference delay equations, Theor. Pop. Biol. 9 (1976) 178]: the local stability of the positive equilibrium implies its global stability. For intrinsic growth rate r-->infinity the way to chaos is broken down to get extinction of population for the depletion case and to establish a stable two-cycle period for models with immigration. The latter behaviour is also studied for models with random discrete constant perturbations of recruitment type. Extinction, persistence and existence of periodic solutions are studied for the perturbed Ricker model with time-dependent parameters.  相似文献
