Muscarinic activation of tracheal smooth muscle (TSM) involves a M3AChR/heterotrimeric-G protein/NPR-GC coupling mechanism. G protein activators Mastoparan (MAS) and Mastoparan-7 stimulated 4- and 10-fold the NPR-GC respectively, being insensitive to PTX and antibodies against Gαi/o subfamily. Muscarinic and MAS stimulation of NPR-GC was blocked by antibodies against C-terminal of Gαq16, whose expression was confirmed by RT-PCR. However, synthetic peptides from C-terminal of Gαq15/16 stimulated the NPR-GC. Coupling of αq16 to M3AChR is supported by MAS decreased [3H]QNB binding, being abolished after M3AChR-4-DAMP-alkylation. Anti-i3M3AChR antibodies blocked the muscarinic activation of NPR-GC, and synthetic peptide from i3M3AChR (M3P) was more potent than MAS increasing GTPγ [35S] and decreasing the [3H]QNB activities. Coupling between NPR-GC and Gαq16 was evaluated by using trypsin-solubilized-fraction from TSM membranes, which displayed a MAS-sensitive-NPR-GC activity, being immunoprecipitated with anti-Gαq16, also showing an immunoreactive heterotrimeric-G-β -subunit. These data support the existence of a novel transducing cascade, involving Gαq16β γ coupling M3AChR to NPR-GC.  相似文献

The Maldanidae annelid worms are reviewed and the phylogenetic relationships of their subgroups provided, based on Hennigian principles and maximum parsimony. Characters were coded as binary and multistate (transformation series). We used 33 terminal taxa (species), and 50 characters. Characters were treated as unordered and of equal weight, and analysis was run in TNT. Three equally most-parsimonious trees were obtained with heuristic searches, with lengths of 64 steps; CI = 0.95, and RI = 0.98. The monophyly of Maldanidae was supported with 100% of bootstrap and jackknife values. As a result of our analysis, Arenicolidae remains the sister-group of Maldanidae, and both should be referred to Maldanomorpha. Maldanidae was supported by the following synapomorphies: dorsal prostomium; prostomium keel-shaped and fused to peristomium; torus globose behind median chaetigers; median chaetigers greatly elongated; number of pre-anal segments reduced. The subfamily Bogueinae was not monophyletic; Boguea and Boguella were included within Rhodininae. Clymenura, previously included in the Clymenurinae, was included within Euclymeninae. The taxa Notoproctinae, Maldaninae, Nicomachinae and Euclymeninae were grouped in the Maldanoplaca, a new taxon. Eight further new clades have been found, but were not named.  相似文献

In Argentina, 58.2% out of the 8126 Cutaneous Leishmaniasis (CL) incident cases accumulated from 1954 to 2006 were reported in the provinces of Salta and Jujuy. The aim of this study was to develop an exploratory risk map and a potential distribution map of the vector, in order to offer recommendations for CL prevention. A total of 12 079 Phlebotominae (Diptera: Psychodidae) belonging to the species Lutzomyia neivai (Pinto), Lu. migonei (França), Lu. cortelezzii (Brèthes), Lu. shannoni (Dyar), Lu. quinquefer (Dyar) and Brumptomyia spp. (França & Parrot) were captured. Potential distribution models were created for two species, Lu. neivai (incriminated vector of Leishmania braziliensis) and Lu. migonei, associated with domestic animals in Argentina and that in turn could be involved as a link between zoonotic transmission cycles and anthropozoonotic. The Maximum Entropy Modeling System (MaxEnt) was used. The Jackknife test was performed, and the ‘rainfall of the driest month’ was the variable that best generalized the models. Accuracy was evaluated by the area under the curve (AUC) and validated by the Cohen's kappa index. This approximation provides a new analytical resource of high potential for the prevention of the disease, in order to allocate resources properly and to develop the most suitable strategies for action.  相似文献

Using Polyacrylamide gel electrophoresis, cycloheximide, incorporation of 3H-labelled amino acids and immunological methods, we have demonstrated that injection of ecdy- sterone induces de novo synthesis and release of vitellogenin in both sexes of Sarcophaga bullata. Vitellogenin concentrations were measured by the Mancini-radial immunodiffusion technique. In males a dose as low as 1 ng always makes vitellogenin appear in the haemolymph but very reproducible results are only obtained when doses varying from 10 to 250 ng were injected. In this range, the dose-response curve was linear on a semi- logarithmic scale.

In females, vitellogenin concentration remained low until a few hours after liver feeding and thereafter it rose sharply and reached its maximum about 24 h after the protein meal. 100 μg 6-hydroxydopamine HCl, injected before liver feeding in 4-day-old females, inhibited vitellogenin synthesis and yolk deposition, probably by interfering with the release of a brain hormone. This inhibitory effect on vitellogenin synthesis, but not that on yolk deposition, could be overruled by injection of ecdysterone. Juvenile hormone was ineffective on both. Females, ovariectomized on day 2 or 3, accumulated vitellogenin in their haemolymph, indicating that the continuous presence of the ovaries was not required for vitellogenin synthesis. The possible relation between the gonadotrophs hormone from the brain, vitellogenin synthesis and moulting hormone metabolism is discussed.  相似文献

Summary

RH5849 is a benzoyl hydrazine analog which has been reported to mimic several effects of the arthropod steroid hormone ecdysone to which it is chemically totally unrelated. In adult Diptera, ecdysone is the hormone that triggers vitellogenin synthesis. We report here that RH5849, upon oral ingestion, is able to induce vitellogenin synthesis in male Drosophila, Neobellieria, Phormia and Lucilia. This contrasts to data in the literature which showed that RH5849 could not mimic the pupariation-inducing effect of ecdysone in last instar fly larvae. RH5849 neither exerts a juvenile hormone mimicking effect nor behaves as an anti-juvenile hormone in both the Colorado potato beetle and Galleria.  相似文献

Summary

The size, structure, and density of a population of Lytechinus variegatus were recorded from a shallow seagrass bed (Halodule wrightii) at 40-day intervals from April 1994 to April 1995. A local mass mortality was observed in May 1994. The density declined from 0.57±0.10 to 0.06±0.05 ind.m?2, mean size declined from 37.6±0.49 to 16.2±2.14 mm and maximum size from 75 to 40 mm. Ten months after mortality, the maximum and mean sizes had returned to initialvalues, but density had not (0.10±0.03 ind.m?2). A second, smaller mass mortality occurred in April 1995. High values of gonad index were recorded before mortality (9.40±5.97) and after population recovery (11.96±5.74). Recruitment was continuous throughout the year, but reached a peak from August to October. This can he related to the patterns of upwelling in the region. Mass mortality is probably an annual event in this population that can be correlated with the occurrence of low tides during daytime in the fell. The population can recover during the summer when low tides occur during the night. The massive input of recruits prevents extinction of the population during the season of high mortality. Population recovery depends on the magnitude of recruitment.  相似文献
