Asymmetric cell growth and division rely on polarized actin cytoskeleton remodeling events, the regulation of which is poorly understood. In budding yeast, formins stimulate the assembly of an organized network of actin cables that direct polarized secretion. Here we show that the Fer/Cip4 homology–Bin amphiphysin Rvs protein Hof1, which has known roles in cytokinesis, also functions during polarized growth by directly controlling the activities of the formin Bnr1. A mutant lacking the C-terminal half of Hof1 displays misoriented and architecturally altered cables, along with impaired secretory vesicle traffic. In vitro, Hof1 inhibits the actin nucleation and elongation activities of Bnr1 without displacing the formin from filament ends. These effects depend on the Src homology 3 domain of Hof1, the formin homology 1 (FH1) domain of Bnr1, and Hof1 dimerization, suggesting a mechanism by which Hof1 “restrains” the otherwise flexible FH1-FH2 apparatus. In vivo, loss of inhibition does not alter actin levels in cables but, instead, cable shape and functionality. Thus Hof1 tunes formins to sculpt the actin cable network.  相似文献

Fus2p is a pheromone-induced protein associated with the amphiphysin homologue Rvs161p, which is required for cell fusion during mating in Saccharomyces cerevisiae. We constructed a functional Fus2p-green fluorescent protein (GFP), which exhibits highly dynamic localization patterns in pheromone-responding cells (shmoos): diffuse nuclear, mobile cytoplasmic dots and stable cortical patches concentrated at the shmoo tip. In mitotic cells, Fus2p-GFP is nuclear but becomes cytoplasmic as cells form shmoos, dependent on the Fus3p protein kinase and high levels of pheromone signaling. The rapid cytoplasmic movement of Fus2p-GFP dots requires Rvs161p and polymerized actin and is aberrant in mutants with compromised actin organization, which suggests that the Fus2p dots are transported along actin cables, possibly in association with vesicles. Maintenance of Fus2p-GFP patches at the shmoo tip cortex is jointly dependent on actin and a membrane protein, Fus1p, which suggests that Fus1p is an anchor for Fus2p. In zygotes, Fus2p-GFP forms a dilating ring at the cell junction, returning to the nucleus at the completion of cell fusion.  相似文献

Summary We use contemporary life history theory to analyze parental decisions concerned with the defense of offspring, and with the provisioning of offspring in the presence of risk. In achieving the optimal level of parental investment, the parent faces a tradeoff between present and future reproductive success. Our models, which are based on stochastic dynamic programming, lead to predictions of the following kind: (i) offspring will be defended more vigorously as they grow older; (ii) long-lived species will accept fewer risks in caring for offspring than short-lived species; (iii) parents living in permanently riskier environments will defend their offspring more vigorously than parents in less risky environments; (iv) however, temporary increases in risk will result in temporarily less vigorous defense and provisioning of offspring. The models also show that parents and their offspring have different conceptions of the optimal level of parental investment. The flexibility of our modeling approach as a method of analyzing facultative behavior is also emphasized. Finally, we apply the methods of this paper to analyze fledging behavior of Atlantic puffins.  相似文献

Background

Yami and Ivatan islanders are Austronesian speakers from Orchid Island and the Batanes archipelago that are located between Taiwan and the Philippines. The paternal genealogies of the Yami tribe from 1962 monograph of Wei and Liu were compared with our dataset of non-recombining Y (NRY) chromosomes from the corresponding families. Then mitochondrial DNA polymorphism was also analyzed to determine the matrilineal relationships between Yami, Ivatan, and other East Asian populations.

Results

The family relationships inferred from the NRY Phylogeny suggested a low number of paternal founders and agreed with the genealogy of Wei and Liu (P < 0.01). Except for one Y short tandem repeat lineage (Y-STR), seen in two unrelated Yami families, no other Y-STR lineages were shared between villages, whereas mtDNA haplotypes were indiscriminately distributed throughout Orchid Island. The genetic affinity seen between Yami and Taiwanese aborigines or between Ivatan and the Philippine people was closer than that between Yami and Ivatan, suggesting that the Orchid islanders were colonized separately by their nearest neighbors and bred in isolation. However a northward gene flow to Orchid Island from the Philippines was suspected as Yami and Ivatan peoples both speak Western Malayo-Polynesian languages which are not spoken in Taiwan. Actually, only very little gene flow was observed between Yami and Ivatan or between Yami and the Philippines as indicated by the sharing of mtDNA haplogroup B4a1a4 and one O1a1* Y-STR lineage.

Conclusions

The NRY and mtDNA genetic information among Yami tribe peoples fitted well the patrilocal society model proposed by Wei and Liu. In this proposal, there were likely few genetic exchanges among Yami and the Philippine people. Trading activities may have contributed to the diffusion of Malayo-Polynesian languages among them. Finally, artifacts dating 4,000 YBP, found on Orchid Island and indicating association with the Out of Taiwan hypothesis might be related to a pioneering stage of settlement, as most dating estimates inferred from DNA variation in our data set ranged between 100-3,000 YBP.  相似文献

We compiled a >50‐year record of morphometrics for semipalmated sandpipers (Calidris pusilla), a shorebird species with a Nearctic breeding distribution and intercontinental migration to South America. Our data included >57,000 individuals captured 1972–2015 at five breeding locations and three major stopover sites, plus 139 museum specimens collected in earlier decades. Wing length increased by ca. 1.5 mm (>1%) prior to 1980, followed by a decrease of 3.85 mm (nearly 4%) over the subsequent 35 years. This can account for previously reported changes in metrics at a migratory stopover site from 1985 to 2006. Wing length decreased at a rate of 1,098 darwins, or 0.176 haldanes, within the ranges of other field studies of phenotypic change. Bill length, in contrast, showed no consistent change over the full period of our study. Decreased body size as a universal response of animal populations to climate warming, and several other potential mechanisms, are unable to account for the increasing and decreasing wing length pattern observed. We propose that the post‐WWII near‐extirpation of falcon populations and their post‐1973 recovery driven by the widespread use and subsequent limitation on DDT in North America selected initially for greater flight efficiency and latterly for greater agility. This predation danger hypothesis accounts for many features of the morphometric data and deserves further investigation in this and other species.  相似文献

We investigated how parental provisioning and nestling departure behaviour interact to produce prefledging mass recession in Cassin's auklets, Ptychoramphus aleuticus. Under our hypothesis, auklet parents are reluctant provisioners and should be increasingly likely to terminate or reduce provisioning of their single nestling as it matures. For the nestling, remaining in the nest presents a risk of losing mass (if the parent does not provision) but also a possibility of additional provisioning. As the nestling matures and becomes increasingly capable of independence, the benefits of remaining in the nest decrease and nestlings should be increasingly likely to fledge. Nestlings also should be more likely to fledge when the expectation of additional provisioning is low. Data on parental provisioning (based on growth increments) and nestling departure were consistent with our hypothesis. Older and heavier nestlings had smaller growth increments than younger and lighter nestlings, suggesting that parents were reluctant to provision old and heavy nestlings. Older and heavier nestlings also were increasingly likely to fledge. We hypothesized that nestlings might use the intensity of mass loss to gauge parental reluctance to provision, and accordingly, nestlings were increasingly likely to fledge after experiencing a greater amount of mass loss. We examined the effects of predation risk on the behavior ofrhinoceros auklets (Cerorhinca monocerata) breeding at PineIsland, British Columbia, in 1990. Provisioning parents in someareas of the colony risked predation by bald eagles (Haliacetusleucocephalus). Chicks in high and low predation risk areasof the colony hatched on approximately the same date, receivedsimilar amounts of food to 46 days of age, grew at the samerate, reached similar peak masses, and fledged at similar masses.However, chicks in high predation areas fledged at a youngerage than did chicks in low predation areas. These data are consistentwith the hypothesis that parents in high risk areas terminatedprovisioning several days before those in lower risk areas.Mass at fledging was inversely related to age at fledging inboth high and low risk areas. The regression line for the highrisk habitats lies below that from the low risk habitats, aspredicted by a model that examines optimal time of fledgingfrom the perspective of the parents. We conclude that risk ofpredation represents a significant cost of reproduction to somerhinoceros auklets and that individual auklets within the colonyvary their behavior according to predation risk.  相似文献

Rich habitats, intensive feeding, and large fuel deposits areassumed to improve the capability for long-distance migrationby birds but may also heighten their vulnerability or exposureto predators. Studies of habitat use by migrants have emphasizedthe importance of feeding, and relatively few studies have consideredhow migrants manage the dangers inherent in acquiring and storingfuel. Migrant western sandpipers (Calidris mauri) stop overon coastal mudflats characterized by a strong feeding–dangergradient, with both food and danger decreasing with distancefrom the shoreline. We experimentally manipulated danger byadding obstructive cover and measured sandpiper usage alongthis gradient. We compared sandpiper usage along a transectextending 100 m on either side of the obstruction with thaton matched control transects without obstructions. The droppingdensity accumulated during a low-tide period provided a sensitivemeasure of sandpiper usage. Mean usage on control transectswas 2.3 droppings/m2 and was lower by 1.5 droppings/m2 (65%)on treatment transects. Usage did not differ between controland treatment transects at the furthest distances from the obstruction,the difference increased with proximity to the obstruction,and was greater by on average 0.9 droppings/m2 on the oceanwardside (low food abundance) than on the shoreward side (high foodabundance). All these findings were predicted by danger managementtheory. Our study provides experimental evidence that migrantbirds are sensitive to danger on stopover and has implicationsfor understanding migration strategies.  相似文献
