Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL)is a rare hereditary cerebrovascular disease caused by a NOTCH3 mutation.However,the underlying cellular and molecular mechanisms remain unidentified.Here,we generated non-integrative induced pluripotent stem cells(iPSCs)from fibroblasts of a CADASIL patient harboring a heterozygous NOTCH3 mutation(c.3226C>T,p.R1076C).Vascular smooth muscle cells(VSMCs)differentiated from CADASIL-specific iPSCs showed gene expression changes associated with disease phenotypes,including activation of the NOTCH and NF-kB signaling pathway,cytoskeleton disorganization,and excessive cell proliferation.In comparison,these abnormalities were not observed in vascular endothelial cells(VECs)derived from the patients iPSCs.Importantly,the abnormal upregulation of NF-kB target genes in CADASIL VSMCs was diminished by a NOTCH pathway inhibitor,providing a potential therapeutic strategy for CADASIL.Overall,using this iPSCbased disease model,our study identified clues for studying the pathogenic mechanisms of CADASIL and developing treatment strategies for this disease. 相似文献

Cell culture toxicity testing methods were modified and applied to the development of implantable glucose microsensors, and positive and negative control materials suitable for the microsensor assessment were established. The location, source and degree of the toxic effect in a multi-component biosensor was spatially visualized with cell monolayers. A freshly prepared sensor showed moderate toxicity, mainly as a result of the presence of glutaraldehyde and the residual solvents in the polymer layers. However, it was possible to reduce the toxicity by removing the leachable toxic substances through extraction in phosphate, buffer, and a non-toxic sensor was readily obtained. 相似文献

Ribonuelease activity and its isozyme pattern of mung bean cotyledon segment were determined during dedifferentiation. The results showed that the activity of ribonuelease in the dedifferentiated tissue is higher than that of control, three new bands of isozyme appear during callus formation. Addition of cycloheximide (CHM) at 0 or 24 hours inhibits either the enzyme activity or the appearance of new bands. 观察慢性缺氧对大鼠肺动脉诱导型一氧化氮合酶 (i NOS)和蛋白激酶 C (PKC)表达的影响 ,并探讨 i NOS及 PKC的相互关系在肺动脉高压形成中的作用。建立常压缺氧肺动脉高压模型。应用免疫组化染色和计算机定量图像分析技术观察各组 i NOS和 PKC在蛋白质水平表达的变化 ,并分析二者之间的关系。结果表明随着缺氧时间的延长 ,PKC的表达逐渐上升而 i NOS的表达逐渐下降 ,直线相关分析结果呈显著性负相关 (n=2 4,r=- 0 .94,P<0 .0 1) ,提示 i NOS与 PKC相互拮抗作用与缺氧性肺动脉高压的形成及发展有关 相似文献

HMG proteins are abundant chromosomal non-histone proteins. It has been suggested that the HMG proteins may play an important
role in the structure and function of chromatin. In the present study, the binding of HMG proteins (HMG1/2 and HMG14/17) to
the core DNA sequence of DNaseI hypersensitive site 2 (HS2core DNA sequence, -10681-10970 bp) in the locus control region
(LCR) of the human β-like globin gene cluster has been examined by using both thein vitro nucleosome reconstitution and the gel mobility shift assays. Here we show that HMG1/2 can bind to the naked HS2core DNA sequence,
however, HMG14/17 cannot. Using thein vitro nucleosome reconstitution we demonstrate that HMG14/17 can bind to the HS2core DNA sequence which is assembled into nucleosomes
with the core histone octamer transferred from chicken erythrocytes. In contrast, HMG1/2 cannot bind to the nucleosomes reconstitutedin vitro with the HS2core DNA sequence. These results indicate that the binding patterns between HMG proteins and the HS2core DNA
sequence which exists in different states (the naked DNA or thein vitro reconstituted nucleosomal DNA) are quite different. We speculate that HMG proteins might play a critical role in the regulation

短尾拟鲿PSEUDOBAGRUS BREVICAUDATUS(Wu)原名短(鱼危)Leiocassis brevicaudatus Wu是伍献文先生1930年命名的一个新种1。张春霖先生在《中国贴类志》中将其改称为短尾黄颖鱼PSEUDOBAGRUS BREVICAUDATUS2。湖北省水生生物研究所在《长江鱼类》一书中仍称为短尾跪2,并有较细致的形态描述。成庆泰先生等主要依据脂鳍长短和尾鳍之中央鳍条至少为最长鳍条的2/3等特征,将其归人拟偿属,称短尾拟绘4。1990-1993年,我们在进行嘉陵江绘科鱼类骨骼的比较研究中,发现短尾拟嵘骨骼的许多特征与跪属的粗唇(鱼危)Leiocassis crassilabris GUnther等相似,而与拟鳄属的细体拟偿Pseudobagrus pratti (GUnther)、切尾拟(鱼危)Pseudobagrus truncatus (Regan)和乌苏拟(鱼危)Pseudobagrus ussuriensis(Dybowski)等差异明显。
