We investigated the properties of the permeability transition pore (PTP) in Saccharomyces cerevisiae in agar-embedded mitochondria (AEM) and agar-embedded cells (AEC) and its role in yeast death. In AEM, ethanol-induced pore opening, as indicated by the release of calcein and mitochondrial membrane depolarization, can be inhibited by CsA, by Cpr3 deficiency, and by the antioxidant glutathione. Notably, the pore opening is inhibited, when mitochondria are preloaded by EGTA or Fluo3 to chelate matrix Ca2+, or are pretreated with 4-Br A23187 to extract matrix Ca2+, prior to agar-embedding, or when pore opening is induced in the presence of EGTA; opened pores are re-closed by sequential treatment with CsA, 4-Br A23187 plus EGTA and NADH, indicating endogenous matrix Ca2+ involvement. CsA also inhibits the pore opening with low conductance triggered by exogenous Ca2+ transport with ETH129. In AEC, the treatment of tert-butylhydroperoxide, a pro-oxidant that triggers transient pore opening in high conductance in AEM, induces yeast death, which is also dependent on CsA and Cpr3. Furthermore, AEMs from mutants lacking three ADP/ATP carrier (AAC) isoforms and with defective ATP synthase dimerization exhibit high and low conductance pore openings with CsA sensitivity, respectively. Collectively, these data show that the yeast PTP is regulated by Cpr3, endogenous matrix Ca2+, and reactive oxygen species, and that it is involved in yeast death; furthermore, ATP synthase dimers play a key role in CsA-sensitive pore formation, while AACs are dispensable. 相似文献

We have found a linear, 16 kb, double-stranded RNA (dsRNA) in symptomless Japonica rice (Oryza sativa L.) that is not found in Indica rice (Oryza sativa L.). The dsRNA was detected in every tissue and at every developmental stage, and its copy number was approximately constant (about 20 copies/cell). Double-stranded RNA was also detected in two strains of Oryza rufipogon (an ancestor of O. sativa). Hybridization experiments indicated that the dsRNA of O. rufipogon was homologous but not identical to that of O. sativa. The sequence of about 13.2 kb of the dsRNA was determined and two open reading frames (ORFs) were found. Analysis of products formed in Chlorella vulgaris 11 h cellsduring photosynthesis in air containing 3,000 ppm 14CO2 at varioustemperatures revealed that the level of 14C-starch was maximumaround 2024?C and decreased with further rise in temperatureuntil 40?C, while 14C-sucrose greatly increased at temperaturesabove about 28?C. Polar cod, Boreogadus saida, is a key species in the Arctic Ocean ecosystem. We examined the distribution pattern of B. saida and other fish larvae in relation to oceanographic parameters, including sea surface temperature and salinity (SST and SSS), the mode of temperature and salinity within the water column (F temp and F sal), and the temporal duration between the date of sea ice retreat and the date of field surveys (dSRT) in the northern Bering Sea and Chukchi Sea during the summers of 2008 and 2013. Sampling was conducted onboard the T/S Oshoro-Maru using a bongo net for 2 years. At sampling stations, the temperature and salinity were measured using conductivity-temperature-depth profiler casts. We calculated dSRT from satellite derived polar gridded sea ice concentration data. A total of 1186 individuals comprising 7 families and 16 species were collected, with B. saida (35 %) and Ammodytes hexapterus (27 %) dominating the catch in number. Based on the species composition (cluster analysis), the sampling stations were divided into four groups. Pleuronectidae dominated group A, which was characterized by relatively high temperature (SST and F temp), while B. saida dominated group B, characterized by low temperatures. A. hexapterus dominated group C and D, which had similar temperatures and salinities, but group C had a shorter dSRT than group D. The latter was also differentiated by the presence of Lumpenus sp. B. saida, which were most abundant in regions where temperatures ranged from ?2 to 0.5 °C. These results, the geographical variation of group B sampling stations, suggest there are two separate spawning areas characterized by similar fish communities and related to consistently cold bottom temperatures. Moreover, stations with a higher abundance of smaller sized larval B. saida were characterized by a short dSRT, whereas stations with a lower abundance and a larger size were characterized by a long dSRT. 相似文献

This study aimed to determine gait ability at hospital discharge in patients undergoing total knee arthroplasty (TKA) as an indicator of the risk of falling. Fifty-seven patients undergoing primary TKA for knee osteoarthritis participated in this study. Gait variability measured with accelerometers and physical function including knee range of motion (ROM), quadriceps strength, walking speed, and the Timed Up and Go (TUG) test were evaluated preoperatively and at discharge from the hospital (1 month before and 5 days after surgery). All patients were discharged directly home at 5 days after surgery. Knee flexion of ROM, quadriceps strength, walking speed, and the TUG test results were significantly worse at hospital discharge than preoperatively (p < 0.001). However, gait variability was not significantly different before and after TKA. This result indicated that patients following TKA surgery could walk at hospital discharge as stably as preoperatively regardless of the decrease in physical function, including knee ROM, quadriceps strength, and gait speed after surgery. 相似文献

ASC is a pro-apoptotic protein containing a pyrin domain (PD) and a caspase-recruitment domain (CARD). A previous study suggests that ASC interacts with Ipaf, a member of the Apaf-1/Nod1 protein family. However, the functional relevance of the interaction has not been determined. Here, we report that co-expression of ASC with Ipaf or oligomerization of ASC induces both apoptosis and NF-kappa B activation. Apoptosis induced through ASC was inhibited by a mutant form of Caspase-8 but not by that of Caspase-1. The PD of ASC physically interacted with Caspase-8 as well as with pyrin, the familial Mediterranean fever gene product. Caspase-8 deficiency rescued mouse fibroblasts from apoptosis induced by ASC oligomerization. Pyrin disrupted the interaction between ASC and Caspase-8, and inhibited both apoptosis and NF-kappa B activation induced by ASC. These findings suggest that ASC is a mediator of NF-kappa B activation and Caspase-8-dependent apoptosis in an Ipaf signaling pathway. 相似文献
