Expression of a maize δ-type DNA polymerase during seed germination   总被引:1,自引:1,他引:0  
We have cloned a 1563-bp cDNA sequence from a reported 2072-bp cDNA (including a fragment of the 3' UTR region, accession number AY109773 ) corresponding to the carboxy half of maize DNA polymerase δ[ Zea mays delta-type DNA polymerase catalytic subunit (Zmpolδ); EC 2.7.7.7], and its sequence shows an identity of 95, 77 and 74% to rice, soybean and Arabidopsis enzymes, respectively, although identity is even higher if only the polδ-defining domain sequences are considered. An important difference between the monocot and dicot enzymes is the presence in the former of Zn fingers apparently required for binding to the DNA polδ holoenzyme B subunit, which is absent in the dicot enzymes. Expression of Zmpolδ and protein levels during germination remain unchanged; however, polδ shows two peaks of activity, one during early germination, perhaps related to DNA repair processes and a second peak when DNA replication is already in progress, indicating that Zmpolδ is regulated at the post-translational level. Zmpolδ has been found mainly in proliferative tissues in seeds and plantlets, although surprisingly, it is also present in seed endosperm, together with Zm proliferating cell nuclear antigen.  相似文献

BackgroundHuman mobility among residential locations can drive dengue virus (DENV) transmission dynamics. Recently, it was shown that individuals with symptomatic DENV infection exhibit significant changes in their mobility patterns, spending more time at home during illness. This change in mobility is predicted to increase the risk of acquiring infection for those living with or visiting the ill individual. It has yet to be considered, however, whether social contacts are also changing their mobility, either by socially distancing themselves from the infectious individual or increasing contact to help care for them. Social, or physical, distancing and caregiving could have diverse yet important impacts on DENV transmission dynamics; therefore, it is necessary to better understand the nature and frequency of these behaviors including their effect on mobility.Methodology and principal findingsThrough community-based febrile illness surveillance and RT-PCR infection confirmation, 67 DENV positive (DENV+) residents were identified in the city of Iquitos, Peru. Using retrospective interviews, data were collected on visitors and home-based care received during the illness. While 15% of participants lost visitors during their illness, 22% gained visitors; overall, 32% of all individuals (particularly females) received visitors while symptomatic. Caregiving was common (90%), particularly caring by housemates (91%) and caring for children (98%). Twenty-eight percent of caregivers changed their behavior enough to have their work (and, likely, mobility patterns) affected. This was significantly more likely when caring for individuals with low “health-related quality of well-being” during illness (Fisher’s Exact, p = 0.01).Conclusions/SignificanceOur study demonstrates that social contacts of individuals with dengue modify their patterns of visitation and caregiving. The observed mobility changes could impact a susceptible individual’s exposure to virus or a presymptomatic/clinically inapparent individual’s contribution to onward transmission. Accounting for changes in social contact mobility is imperative in order to get a more accurate understanding of DENV transmission.  相似文献

Background and Aims

Studies of the effects of pollination on floral scent and bee visitation remain rare, particularly in agricultural crops. To fill this gap, the hypothesis that bee visitation to flowers decreases after pollination through reduced floral volatile emissions in highbush blueberries, Vaccinium corymbosum, was tested. Other sources of variation in floral emissions and the role of floral volatiles in bee attraction were also examined.

Methods

Pollinator visitation to blueberry flowers was manipulated by bagging all flowers within a bush (pollinator excluded) or leaving them unbagged (open pollinated), and then the effect on floral volatile emissions and future bee visitation were measured. Floral volatiles were also measured from different blueberry cultivars, times of the day and flower parts, and a study was conducted to test the attraction of bees to floral volatiles.

Key Results

Open-pollinated blueberry flowers had 32 % lower volatile emissions than pollinator-excluded flowers. In particular, cinnamyl alcohol, a major component of the floral blend that is emitted exclusively from petals, was emitted in lower quantities from open-pollinated flowers. Although, no differences in cinnamyl alcohol emissions were detected among three blueberry cultivars or at different times of day, some components of the blueberry floral blend were emitted in higher amounts from certain cultivars and at mid-day. Field observations showed that more bees visited bushes with pollinator-excluded flowers. Also, more honey bees were caught in traps baited with a synthetic blueberry floral blend than in unbaited traps.

Conclusions

Greater volatile emissions may help guide bees to unpollinated flowers, and thus increase plant fitness and bee energetic return when foraging in blueberries. Furthermore, the variation in volatile emissions from blueberry flowers depending on pollination status, plant cultivar and time of day suggests an adaptive role of floral signals in increasing pollination of flowers.  相似文献

Narratives of 27 Puerto Rican and Mexican students, written first in eighth grade then again as juniors in high school, address the important question of "Who am I?" and illustrate school-sponsored silencing, with students' critiques of their educational experience ignored by both the elementary and the high school. The narratives also provide a window into the high dropout rates of Latino children, the reasons behind students' academic decisions, and interventions needed to change negative schooling processes and outcomes. Several factors have been identified as relevant in determining the abundance of non-native invasive species. Nevertheless, the relative importance of these factors will vary depending on the invaded habitat and the characteristics of the invasive species. Due to their harsh environmental conditions and remoteness, high-alpine habitats are often considered to be at low risk of plant invasion. However, an increasing number of reports have shown the presence and spread of non-native plant species in alpine habitats; thus, it is important to study which factors control the invasion process in these harsh habitats. In this study, we assessed the role of disturbance, soil characteristics, biotic resistance and seed rain in the establishment and abundance of the non-native invasive species Taraxacum officinale (dandelion) in the Andes of central Chile. By focusing on human-disturbed patches, naturally disturbed patches, and undisturbed patches, we did not find that disturbance per se, or its origin, affected the establishment and abundance of T. officinale. The abundance of this non-native invasive species was not negatively related to the diversity of native species at local scales, indicating no biotic resistance to invasion; instead, some positive relationships were found. Our results indicate that propagule pressure (assessed by the seed rain) and the abiotic soil characteristics are the main factors related to the abundance of this non-native invasive species. Hence, in contrast to what has been found for more benign habitats, disturbance and biotic resistance have little influence on the invasibility of T. officinale in this high-alpine habitat.  相似文献
