The ancestry of New World cattle was investigated through the analysis of mitochondrial and Y chromosome variation in Creoles from Argentina, Brazil, Mexico, Paraguay and the United States of America. Breeds that influenced the Creoles, such as Iberian native, British and Zebu, were also studied. Creoles showed high mtDNA diversity (H = 0.984 ± 0.003) with a total of 78 haplotypes, and the European T3 matriline was the most common (72.1%). The African T1a haplogroup was detected (14.6%), as well as the ancestral African‐derived AA matriline (11.9%), which was absent in the Iberian breeds. Genetic proximity among Creoles, Iberian and Atlantic Islands breeds was inferred through their sharing of mtDNA haplotypes. Y‐haplotype diversity in Creoles was high (H = 0.779 ± 0.019), with several Y1, Y2 and Y3 haplotypes represented. Iberian patrilines in Creoles were more difficult to infer and were reflected by the presence of H3Y1 and H6Y2. Y‐haplotypes confirmed crossbreeding with British cattle, mainly of Hereford with Pampa Chaqueño and Texas Longhorn. Male‐mediated Bos indicus introgression into Creoles was found in all populations, except Argentino1 (herd book registered) and Pampa Chaqueño. The detection of the distinct H22Y3 patriline with the INRA189‐90 allele in Caracú suggests introduction of bulls directly from West Africa. Further studies of Spanish and African breeds are necessary to elucidate the origins of Creole cattle, and determine the exact source of their African lineages.  相似文献

Synaptic transmission depends on neurotransmitter pools stored within vesicles that undergo regulated exocytosis. In the brain, the vesicular monoamine transporter-2 (VMAT2) is responsible for the loading of dopamine (DA) and other monoamines into synaptic vesicles. Prior to storage within vesicles, DA synthesis occurs at the synaptic terminal in a two-step enzymatic process. First, the rate-limiting enzyme tyrosine hydroxylase (TH) converts tyrosine to di-OH-phenylalanine. Aromatic amino acid decarboxylase (AADC) then converts di-OH-phenylalanine into DA. Here, we provide evidence that VMAT2 physically and functionally interacts with the enzymes responsible for DA synthesis. In rat striata, TH and AADC co-immunoprecipitate with VMAT2, whereas in PC 12 cells, TH co-immunoprecipitates with the closely related VMAT1 and with overexpressed VMAT2. GST pull-down assays further identified three cytosolic domains of VMAT2 involved in the interaction with TH and AADC. Furthermore, in vitro binding assays demonstrated that TH directly interacts with VMAT2. Additionally, using fractionation and immunoisolation approaches, we demonstrate that TH and AADC associate with VMAT2-containing synaptic vesicles from rat brain. These vesicles exhibited specific TH activity. Finally, the coupling between synthesis and transport of DA into vesicles was impaired in the presence of fragments involved in the VMAT2/TH/AADC interaction. Taken together, our results indicate that DA synthesis can occur at the synaptic vesicle membrane, where it is physically and functionally coupled to VMAT2-mediated transport into vesicles.  相似文献

The vegetative development of the seagrass Thalassia testudinum was assessed in three reefs (Hornos, isla Sacrificios, isla de Enmedio) of the Veracruz Reef System (Veracruz, México) which are located at increasing distances from the mainland coast. Leaf nutrient content of T. testudinum suggests that the availability of nitrogen and phosphorus increases from Hornos reef to isla Sacrificios and isla de Enmedio, that is, as the distance to the coast increases. The total biomass of this species tended to increase similarly as a result of the large increase of the biomass of rhizomes and roots but not of leaves. In contrast to previous knowledge of the response of seagrasses along gradients of nutrient availability, the rhizome and roots to shoot biomass ratio increased by a factor of two as nutrient availability increased. The density of T. testudinum shoots and their mass, the LAI and leaf productivity, and the average number of leaves produced by a T. testudinum shoot in 1 year were lowest in the reef closest to shore (Hornos). Our results show that the vegetative development of T. testudinum in Hornos reef is restrained when compared with that in isla Sacrificios and isla de Enmedio and suggest that the differences of vegetative development of this species in the Veracruz Reef System might be driven by factors other than nutrient availability.  相似文献

Twenty individuals classified as disabled due to diagnosed cerebral cysticercosis were studied to correlate the presence of neurocysticercosis with the history of having worked in contact with organic solvents as possible triggering agents of the pathology. The study was performed by direct or indirect interrogatory, review of the case histories, serologic examinations, and psychological evaluation of the intellectual performance level and psychopathological index. Of the 20 subjects, 50% had been exposed to organic solvents in their work; all had been in contact with the infecting parasite, either through frequent consumption of pork meat, or through contact with fecal mass. The titers of the haemagglutination test varied from 1:2 to 1:256, being the diagnosis value of > or = 1:16. There was also a variation as a function of the antigen used for the sera tests performed in the sampled subjects, such as complete or incomplete somatic antigen, vesicular fluid, and excretions and secretions. Other variations found, such as symptoms and the results of the psychological evaluation are depicted in tables. It is concluded that in these cases, neurocysticercosis is an invalidating disease, and the presence of organic solvents is emphasized as an important factor of the disease or as a triggering factor.  相似文献

Two recent studies have mapped QTLs associated with the level of seed glucosinolates in oilseed rape (Brassica napus L.). It was likely that the two most significant QTLs identified in each study were the same, as they were linked to RFLP alleles identified by common DNA probes. To investigate the utility of these probes in breeding programmes, they were used to study RFLPs in a range of low- and high-glucosinolate cultivars and breeding lines. It was shown that all low glucosinolate spring and winter cultivars possessed a specific RFLP fragment identified by probe wg3f7 which is linked to theGSL-1 QTL, and all high-glucosinolate cultivars possessed a specific RFLP fragment identified by probe wg7a8, which is linked to theGSL-2 QTL. Cultivar Ariana, which has intermediate levels of glucosinolates possessed the low-glucosinolate fragment atGSL-1 but the high-glucosinolate fragment atGSL-2. A similar result was found with the cvs. Martina and Bronowski which have intermediate and variable levels of glucosinolates. There were no other RFLP fragments identified by other DNA probes which were specific to either the low- or high-glucosinolate phenotypes. The use of probes wg3f7 and wg7a8 in selection of low-glucosinolate lines in breeding programmes is discussed.  相似文献
