用人重组肿瘤坏死因子-α(Tumornecrosisfactor-α,TNF-α)和人天然α干扰素(Interferon-α-,IFN-α)在人胚胎肺纤维母细胞(HEF)和Hep-2细胞系上对常见呼吸道病毒所致细胞病变抑制进行比较观察。病毒包括不同型别的腺病毒5株,单疱病毒Ⅰ型(HSV-I)1株,鼻病毒1株,仙台病毒1株,VSV1株。结果提示TNF-α和IFN-α均具有广谱抗病毒活性。TNF-α的抑毒作用能被TNF-α申抗和IFN-β单抗完全去除,被IFN-α单抗部分去除TNF-α的抗病毒效应。TNF--α中和试验的结果提示:TNF抗病毒活性仍为IFN-β诱生所介导。  相似文献

A novel S2O3 2? luminescent sensor (Cu2+-p-CPIP) was developed and the presence of S2O3 2? caused an obvious fluorescence enhancement at 420 nm upon excitation at 330 nm, which could be distinguished with the naked eye under a UV lamp. Remarkably, the compound exhibited excellent selective and sensitive response to S2O3 2? over other common anions with a micromolar limit of detection (0.442 μM) in DMSO/H2O (v/v, 1:1) buffer. The absorbance intensity and the color of Cu2+-p -CPIP solution changed gradually with the increase of S2O3 2? concentration. The proposed method was applied to the determination of S2O3 2? in milk samples and the recoveries were 97.5–105%. The preparation of Cu2+-p -CPIP exhibited the quick, simple and facile advantages. The results showed that Cu2+-p -CPIP can be a good candidate for simple, rapid and sensitive colorimetric detection of S2O3 2? in aqueous solution.  相似文献

Oomyzus sokolowskii (Kurdjumov) is a gregarious larval-pupal parasitoid of the diamondback moth, Plutella xylostella L. Under laboratory conditions, we explored the reproductive attributes including the functional response of O. sokolowskii and the impact of mutual interference among female parasitoids on progeny production and progeny sex ratio. In the functional response experiment, groups of 1–50 late instar larvae were exposed to groups of five female parasitoids for 48 h. Both Holling's disc equation (type II response) and Williams and Martinez's model (type III response) fit significantly better than the null-model assuming density-independent parasitation efficiency. Williams and Martinez's model was only marginally better than Holling's disc equation which explained 82% of the variation in the number of parasitized hosts. According to Holling's disc equation, the estimated maximum number of hosts parasitised during 48 h was 17 per group of five parasitoids, the estimated attack efficiency of a single female parasitoid (a) was 0.0024 larvae per hour, and the estimated handling time of a single female (T h) was 14.4 h per host. In the mutual interference experiment 50 host larvae were exposed to different numbers of female parasitoids (P=5, 10, 20, 30, 40 and 50) for 48 h. With increasing numbers of conspecifics, the proportion of male progeny increased and total number of progeny per parasitoid decreased, yielding an estimated mutual interference constant (m) (±SE) of 0.95±0.14. The total number of wasps emerging from individual host pupae increased with increasing number of conspecifics. The age-specific fecundity of O. sokolowskii was investigated by providing 10 host larvae daily to individual female parasitoids. After 3 days of oviposition, 90% of the females were still alive and had achieved 76.4% of their lifetime fecundity. The proportion of female progeny decreased with female age. No relationship was found between female longevity and lifetime fecundity. The results suggest that parasitoid and host densities influence the progeny production and sex allocation strategy of O. sokolowskii. The information obtained from this study would help to develop a mass-rearing protocol for O. sokolowskii.  相似文献

Status epilepticus (SE) is a common neurological condition associated with high rates of mortality and permanent brain injury. SE usually leads to neuronal death which may be accompanied by edema, epileptogenesis and learning impairment. Aquaporin-4 (AQP4), is a transmembrane water channel protein in the neuropil of the central nervous system that has an important role in water transport in the brain; AQP4 expression is altered in many pathological conditions such as changes in the blood- brain barrier and/or astrocytic activation, including seizures. AQP4 was shown to be downregulated in the piriform cortex and the hippocampus after SE. Although it is normally expressed at a high level in the cerebellum, little is known about AQP4 levels in the cerebellum following SE. We addressed this in the present study in a mouse model of pilocarpine-induced SE. We found that AQP4 expression was reduced from 3 h to 3 days after SE, with the levels recovering on day 7. Moreover, mice in the acute post-SE stages exhibited impaired motor coordination and learning. These results indicate that cerebellar damage following SE involves changes in AQP4 expression.  相似文献

Obesity is a major global public health concern. Immune responses implicated in obesity also control certain infections. We investigated the effects of high‐fat diet‐induced obesity (DIO) on infection with the Lyme disease bacterium Borrelia burgdorferi in mice. DIO was associated with systemic suppression of neutrophil‐ and macrophage‐based innate immune responses. These included bacterial uptake and cytokine production, and systemic, progressive impairment of bacterial clearance, and increased carditis severity. B. burgdorferi‐infected mice fed normal diet also gained weight at the same rate as uninfected mice fed high‐fat diet, toll‐like receptor 4 deficiency rescued bacterial clearance defects, which greater in female than male mice, and killing of an unrelated bacterium (Escherichia coli) by bone marrow‐derived macrophages from obese, B. burgdorferi‐infected mice was also affected. Importantly, innate immune suppression increased with infection duration and depended on cooperative and synergistic interactions between DIO and B. burgdorferi infection. Thus, obesity and B. burgdorferi infection cooperatively and progressively suppressed innate immunity in mice.  相似文献

Through the analysis of human skeletal remains and mortuary practice in Yinxu, this study investigates the impact of early urbanization on the commoners during the Late Shang dynasty (ca. 1250–1046 B.C.). A total of 347 individuals examined in this study represent non-elites who were recovered from two different burial contexts (formally buried in lineage cemeteries and randomly scattered in refuse pits). Frequencies of enamel hypoplasia (childhood stress), cribra orbitalia (childhood stress and frailty) and osteoperiostitis (adult stress) were examined to assess systemic stress exposure. Our results reveal that there was no significant difference in the frequency of enamel hypoplasia between two burial groups and between sexes, suggesting these urban commoners experienced similar stresses during childhood, but significantly elevated levels of cribra orbitalia and osteoperiostitis were observed in the refuse pit female cohort. Theoretically, urbanization would have resulted in increased population density in the urban centre, declining sanitary conditions, and increased risk of resource shortage. Biologically, children would be more vulnerable to such physiological disturbance; as a result, high percentages of enamel hypoplasia (80.9% overall) and cribra orbitalia (30.3% overall) are observed in Yin commoners. Adults continued to suffer from stress, resulting in high frequencies of osteoperiostitis (40.0% total adults); in particular, in the refuse pit females who may also reflect a compound impact of gender inequality. Our data show that the non-elite urban population in the capital city of Late Shang Dynasty had experienced extensive stress exposure due to early urbanization with further social stratification only worsening the situation, and eventually contributing to collapse of the Shang Dynasty.  相似文献

在海拔2300m选择健康成年男性5人,急进抵海拔4660m,用多导监测仪分别在两地连续7h监测夜间睡眠、呼吸状态和血氧饱和度变化,进行自身对比。结果发现:(1)急进高海拔后,总睡眠时间、有效睡眠指数、Ⅲ~Ⅳ期深睡眠均较中度高原减少(p<0.01);总觉醒时间、Ⅰ~Ⅱ期浅睡眠高海拔较中度高原增多(p<0.05):(2)急进高海拔后,有3名健康人出现周期性呼吸,其中1名健康者出现周期性呼吸119次,伴有中枢性睡眠呼吸暂停,最低Sao_2为78%;(3)同海拔高度夜间睡眠时与清醒时Sao_2相比较,中度高原下降4.2%,高海拔下降11.2%(p<0.01);高海拔与中度高原夜间清醒时Sao_2相比较下降7.4%,睡眠时下降14.4%(p<0.001)。结果提示:(1)睡眠加重了高原人原有的低氧血症;(2)低氧血症导致睡眠结构的紊乱和睡眠质量的降低;(3)睡眠中出现的周期性呼吸,应视为机体的一种自我保护机制;(4)频发的周期性呼吸或睡眠呼吸暂停将影响大脑机能。  相似文献

Ethylene oxide (EtO) is an alkylating agent and a model direct-acting mutagen and carcinogen. This study has evaluated a panel of biologic markers including EtO-hemoglobin adducts (EtO-Hb), sister-chromatid exchanges (SCEs), micronuclei, chromosomal aberrations (CAs), DNA single-strand breaks (SSB) and an index of DNA repair (ratio of UDS to NA-AAF-DNA binding) in the peripheral blood cells of 34 workers at a sterilization unit of a large university hospital and 23 controls working in the university library. Comprehensive environmental histories were obtained on each subject including detailed occupational and smoking histories. Industrial hygiene data obtained prior to the study and personal monitoring during the 8 years preceding the study showed that workers were subject to low-level exposure near or below the current Occupational Safety and Health Administration (OSHA) standard of 1 ppm (TWA). Personal monitoring data obtained during 2 weeks prior to blood sampling were uniformly less than 0.3 ppm (TWA). After adjusting for smoking, EtO workplace exposure was significantly (p less than 0.001) associated with EtO-Hb (a carcinogen-protein adduct) and 2 measures of SCEs [the average number of SCEs/cell (SCE50) and the number of high frequency cells (SCEHFC)]. There was an apparent suppression of DNA repair capacity in EtO-exposed individuals as measured by the DNA repair index; i.e., the ratio of unscheduled DNA synthesis (UDS) and NA-AAF-DNA binding (p less than 0.01). No association of DNA repair index with smoking was found. Another important finding of this study is the highly significant correlation between EtO-Hb adduct levels and SCEHFC (p less than 0.01) and SCEs (p less than 0.02) which provides evidence of a direct link between a marker of biologically effective dose and markers of genotoxic response. In contrast, micronuclei, CAs and SSBs were not significantly elevated in the workers. The activity of the u-isoenzyme of glutathione-S-transferase (GT) was measured as a possible genetic marker of susceptibility and a modulator of biomarker formation. However, possibly because of confounding by age, no significant relationships were found between GT and any of the exposure-related markers by ANOVA or among other independent variables by regression. This study demonstrates significant effects of low-level EtO exposure, independent of smoking history, near or below 1 ppm on multiple biomarkers and suggests that the current OSHA standard may not be adequately protective. Previously described effects of smoking on EtO-Hb adducts, SCEs and SCEHFC were also seen in this study.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Streptococcus suis serotype 2 (S. suis 2) is an important pathogen, responsible for diverse diseases in swine and humans. To obtain a S. suis 2 strain that can be tracked in vitro and in vivo, we constructed the Egfp-HA9801 recombinant S. suis 2 strain with egfp and spc(r) genes inserted via homologous recombination. To assess the effects of the egfp and spc(r) genes in HA9801, the biochemical characteristics, growth features and virulence in Balb/C mice were compared between the recombinant and the parent HA9801 strain. We detected the EGFP expression from Egfp-HA9801 by epifluorescence microscopy. The results showed that the biochemical characterization and growth features of the Egfp-HA9801 recombinant were highly similar to that of the parent HA9801. We did not find significant differences in lethality (50% lethal dose), morbidity and mortality between the two strains. Furthermore, the bacterial counts in each various tissues of Egfp-HA9801-infected mice displayed similar dynamic compared with the HA9801-infected mice. Our results also showed that the Egfp-HA9801 cells grown at 37°C for 36 h displayed greater green fluorescence signals than the cells grown at 28°C for 36 h and 37°C for 24 h. The fluorescence in the tissue cryosections of Egfp-HA9801-injected mice was also stronger than that of the HA9801 group. Together, these results indicate that the egfp and spc(r) insertions into the Egfp-HA9801 recombinant did not significantly change the virulence when compared with HA980, and this EGFP labeled strain can be used for future S. suis 2 pathogenesis research.  相似文献
