Abstract Pseudomonas aeruginosa PAO1 used a broad range of alkanesulfonic acids as sole sulfur source for growth, with molar growth yields of 2.2 to 2.9 kg protein per mol sulfur. 4-Phenylbutane-1-sulfonate was desulfonated in vivo to yield 4-pheny 1-1-butyric acid quantitatively as the sole product, suggesting that the desulfonation mechanism is the same as when alkanesulfonates serve as a carbon source for growth. This contrasts with aromatic sulfonate utilization in other organisms, where different desulfonation reactions are used to provide carbon and sulfur. Desulfonation of alkanesulfonates to provide sulfur was repressed by sulfate or thiocyanate, and derepressed in their absence. The alkanesulfonatase system is hence controlled as part of the sulfate starvation-induced stimulon. 相似文献

The aim of the current work was to evaluate the effect of preschoolers' television (TV) watching time on the prevalence of obesity even after controlling for their total energy intake and their physical activity status. A representative sample of 2,374 Greek children aged 1–5 years was examined (“Growth, Exercise and Nutrition Epidemiological Study in preSchoolers”, GENESIS study). Children's TV watching time on a usual weekday and at a usual weekend was recorded. The overall mean of children's TV viewing time was 1.32 h/day. The majority of participants (74.0%) spent <2 h/day watching TV whereas only 3.1% spent >4 h/day in front of a TV set. Overall, 65.2% of participants were normal weight, 17.2% were overweight, and the rest 17.6% were obese. The prevalence of obesity was significantly higher among those with TV viewing time ≥2 h/day (21.7%) compared to those watching TV <2 h/day (16.1%, P = 0.003). TV viewing time remained significantly associated with the likelihood of being obese even after controlling for potential confounders (i.e., socio demographic and other characteristics and physical activity status) only among children aged 3–5 years. However, further adjusting for children's total energy intake revealed that the association between the TV viewing time and the probability of being obese was no longer statistically significant. On the other hand, physical activity status continued to be an independent factor of being obese. The current findings support the hypothesis that the effect of TV viewing time on childhood obesity is independent of physical activity status and may be attributed to the increased total energy intake during TV watching. 相似文献

After continued etiolation of oat ( Avena sativa L. cv. Arnold) seedlings without greening at any stage, the volumes of both etioplasts and prolamellar bodies were found to increase to a maximum around the 9th day, declining thereafter. Throughout this process a proportion of the early lamellar components is reconverted later into prolamellar body material to account in part for this crystalline accumulation. Starch grains and stroma-centres arc only prominent in early etioplasts. The pro-lamellar bodies of such etioplasts are more amorphous in form resembling more closely the semi-crystalline bodies of dark-grown algae or light-grown cereal seedlings. The high levels of total reducing sugars and fructans after ca 7 days of continued etiolation declined rapidly afterwards, although sucrose content was maximal on the ninth day. It is thought that these carbohydrate reserves were partly used to synthesize sterols (mainly desmethyl sterols) and carotenoids (mainly xanthophylls) found in the high concentrations on around the 9th and 10th days of continued etiolation. High levels of the steroidal saponins in etiolated primary leaves after this time declined to less than half due entirely to the disappearance of avenacoside B, which was partially converted into avenacoside A. Only traces of desgluco-avenacosides A plus B were found in the late stages of continued etiolation. These results demonstrate that etiolated tissue changes constantly with time. Addition of high concentrations of compatible co-solutes such as sugars, sugar alcohols and polyols has recently been shown to lead to marked increases in the thermal stability of oxygen-evolution in chloroplasts (Williams et al. (1992) Biochim. Biophys. Acta 1099, 137-144). In this paper, a similar stabilisation is demonstrated for oxygen-evolving PS II core preparations. The presence of such co-solutes appears, however, to have no ability to stabilise PS II reaction-centre preparations against heat-induced changes in their absorption spectrum. Nor do they protect electron transport from artificial electron donors in PS II core preparations lacking the extrinsic 33 kDa polypeptide of the oxygen-evolution system. Measurements performed on core preparations retaining the 33 kDa polypeptide but lacking the 17 kDa and 23 kDa polypeptides indicate that the co-solutes protect PS-II-mediated electron transport by stabilising the binding of the 33 kDa polypeptide to the core complexes. These findings are discussed in terms of an extension of the general principles underlying the Hofmeister effect observed for soluble proteins to the stabilisation of photosynthetic membrane preparations. 相似文献

Summary Low temperatures induce the accumulation of soluble sugars in plant cells. An attempt is made to identify the primary site
of action of low temperatures and the sequence of physiological and biochemical events leading to sugar accumulation. The
integration of all the available information points to a central role of increased intracellular calcium ion concentration
generated by the inhibition of ATPases concerned with its homeostasis. A positive role of a cold-induced dysfunction of mitochondrial
and chloroplast electron flow is also proposed. The biological significance of an explanation of this physiological response
to a stress factor relies on its categorization into the major family of those employing alternative energy-producing pathways
under stress conditions. More specifically, a connection of sugar mobilization to the needs of the cold-stressed cell to employ
fermentative energy-producing mechanisms is made. From this ankle of view, new ways of genetically modifying the ability of

The activity levels of the enzyme poly(A)polymerase and the levels of protein synthesis primed by endogenous messenger RNA (mRNA) as well as polyuridylic acid, poly(U) directed polyphenylalanine synthesis, were determined in lymphocytic extracts from 17 patients with chronic lymphocytic leukemia of the B cell type. The enzyme activity values have not been found to correlate with the poly(U)-protein synthesis, whereas a positive linear correlation has been established between the activity levels of poly(A)polymerase and the endogenous mRNA-primed protein synthesis (r = 0.735, p less than 0.01). Photoacoustic microscopy (PAM) has great potential for visualization of the microvasculature with high spatial resolution and contrast. Early detection and differentiation of newly developed blood vessels named choroidal neovascularization (CNV) from normal vasculature remains a challenge in ophthalmology. Exogenous contrast agents can assist with improving PAM sensitivity, leading to differentiation of CNV. Here, an FDA-approved indocyanine green (ICG) was utilized as a PAM contrast agent. ICG was conjugated with RGD peptides, allowing the ICG to bind to the integrin expressed in CNV. Molecular PAM imaging showed that ICG-RGD can target CNV for up to 5 days post intravenous administration in living rabbits with a model of CNV. The PAM image sensitivity and image contrast were significantly enhanced by 15-fold at 24 h post-injection. Overall, the presented approach demonstrates the possibility of targeted ICG to be employed in PAM molecular imaging, allowing more precise evaluation of neovascularization. 相似文献
