CpG-oligonucleotides (CpG-ODNs), mimicking bacterial DNA, have recently been shown to stimulate prostate cancer invasion in
vitro via Toll-like receptor 9 (TLR9). Since cyclooxygenase 2 (COX-2), frequently overexpressed in multiple tumor types including
prostate cancer, is a causal factor for tumor development, invasion and metastasis, an interesting question is raised whether
TLR9 regulates COX-2 expression in prostate cancer cells. To address this question, herein we examined COX-2 expression in
PC-3 cells stimulated with different doses and time courses of CpG-ODNs. The regulatory role of NF-κB in TLR9-mediated COX-2
expression was also investigated. CpG-ODN was found to up-regulate the expression of COX-2 in PC-3 cells in a dose- and time-dependent
manner, but have little impact on COX-1 expression. Moreover, CpG-ODN also promoted nuclear translocation and activation of
NF-κB, which appeared to be required for COX-2 induction by CpG-ODN. Overall, TLR9 up-regulates COX-2 expression in prostate

本研究旨在建立一种基于流式细胞术的牛多细胞因子检测方法。对前期制备并筛选出的针对牛细胞因子IFN-γ、IL-2、TNF-α、IP-10和MCP-1的单克隆抗体进行荧光标记,与细胞表面分子抗体组合搭配,进行牛多细胞因子流式检测方法的建立和优化。随后利用建立的方法进行BCG体外感染牛外周血单个核细胞的细胞因子表达规律测定,并结合CFP10-ESAT6蛋白刺激剂评价上述细胞因子作为牛结核诊断标识的潜力。建立的牛多细胞因子流式检测方法可以有效测定BCG感染牛外周血T淋巴细胞的细胞因子表达,其中IFN-γ、IL-2、TNF-α在感染40h后持续上升,而IP-10和MCP-1表达水平呈现下降趋势;对于牛外周血CD4+T淋巴细胞IFN-γ、IL-2、TNF-α的联合检测能有效区分牛结核阳性和阴性样品。这一方法为牛病原菌感染、疫苗注射后细胞免疫应答水平评价以及疫病诊断提供了重要技术手段。 相似文献

Evidence is presented that in the R and P genomes (Secale cereale andAgropyron cristatum, respectively) of theTriticeae there exist closely related 350-family DNA sequences in the terminal heterochromatin. This observation is compared to the relationships between these two genomes derived from a comparison of theNor and5 S DNA loci as well as the available data on morphological characters, chromosome pairing, and isozyme studies. It is concluded that the R and P genomes are not closely related and that the common presence of very similar 350-family DNA sequences reflects the parallel amplification of this family of DNA sequences. 相似文献

Marine mammals are important models for studying convergent evolution and aquatic adaption, and thus reference genomes of marine mammals can provide evolutionary insights. Here, we present the first chromosome‐level marine mammal genome assembly based on the data generated by the BGISEQ‐500 platform, for a stranded female sperm whale (Physeter macrocephalus). Using this reference genome, we performed chromosome evolution analysis of the sperm whale, including constructing ancestral chromosomes, identifying chromosome rearrangement events and comparing with cattle chromosomes, which provides a resource for exploring marine mammal adaptation and speciation. We detected a high proportion of long interspersed nuclear elements and expanded gene families, and contraction of major histocompatibility complex region genes which were specific to sperm whale. Using comparisons with sheep and cattle, we analysed positively selected genes to identify gene pathways that may be related to adaptation to the marine environment. Further, we identified possible convergent evolution in aquatic mammals by testing for positively selected genes across three orders of marine mammals. In addition, we used publicly available resequencing data to confirm a rapid decline in global population size in the Pliocene to Pleistocene transition. This study sheds light on the chromosome evolution and genetic mechanisms underpinning sperm whale adaptations, providing valuable resources for future comparative genomics. 相似文献

半乳糖α 1,3 半乳糖抗原是引起异种器官移植超急性排斥反应 (hyperacuterejection ,HAR)的主要抗原 .α 半乳糖苷酶和α 1,2 岩藻糖转移酶基因可以以不同的方式降低半乳糖α 1,3 半乳糖抗原在内皮细胞表面的表达量 .将人α 半乳糖苷酶基因和α 1,2 岩藻糖转移酶基因单独或连接在一起导入猪血管内皮细胞PEDSV .15中 ,检测细胞表面的抗原及异种天然抗体对细胞杀伤作用 .结果表明α 半乳糖苷酶基因可以将猪血管内皮细胞表面的半乳糖α 1,3 半乳糖抗原清除 74 13%,而α 1,2 岩藻糖转移酶基因也可以清除 4 7 75 %的细胞表面异种抗原 ,但二者都不能达到完全清除的目的 .当α 半乳糖苷酶和α 1,2 岩藻糖转移酶双基因在内皮细胞内共表达时 ,则可以基本清除半乳糖α 1,3 半乳糖抗原 .抗原的减少也可以相应地减弱内皮细胞对异种天然抗体介导的杀伤作用的敏感性 ,尤其是双基因共表达时细胞基本不被杀伤 .结果表明 ,α 半乳糖苷酶基因和α 1,2 岩藻糖转移酶基因可以有效地清除血管内皮细胞表面的半乳糖α 1,3 半乳糖抗原 ,克服HAR的发生 ,为下一步进行动物实验 ,探讨克服异种移植HAR提供了技术途径 相似文献

The present study showed a wide presence of CCL28 in mouse CNS, including cerebral, cerebellum, brain stem and spinal cord. In hippocampus, the expression of CCL28 at both mRNA and protein level was clarified. The CCL28 expression was mainly localized in pyramidal cells of CA area, granular cells of dentate gyrus and some interneurons in CA area and hilus. Double-labelling immunocytochemistry revealed that most of calbindin, calretinin and parvalbumin immunopositive neurons expressed CCL28. During and after pilocarpine induced status epilepticus (SE), a down-regulated expression of CCL28 in hippocampal interneurons in the CA1 area and in the hilus of the dentate gyrus was demonstrated. The present study may, therefore provide evidence that CCL28 may have a novel role in CNS and may be involved in the loss of hippocampal interneurons, and subsequent disinhibition of pyramidal neurons. 相似文献

Background
Abnormal expression of serum TGF-β1 was found in patients with diabetic nephropathy. However, the association of TGF-β1 with the risk of diabetic nephropathy remains unknown. The present study was undertaken to investigate whether such an association exists.Methods
We searched the Chinese VIP, Wangfang, China National Knowledge Infrastructure, PubMed, Embase, and Google Scholar databases for relevant studies and extracted all eligible data. Stata12 software was used for statistical analysis.Results
Nine reports met our criteria and were used for data extraction. There were 264 patients and 227 healthy controls from qualified reports in this meta-analysis. The results suggested that serum TGF-β1 levels were significantly up-regulated in patients with diabetic nephropathy; the instrumental variable was 3.94 (95% confidence interval 3.20–4.68, p<0.01).Conclusions
Meta-analysis suggested that elevated serum TGF-β level in patients with diabetes is associated with a high risk of nephropathy. Further studies are required to validate these observations. 相似文献1000.
诺如病毒(Noroviruses,NoVs)是引起全球急性胃肠炎的常见病原.组织血型抗原(Histo-blood groups antigens,HBGAs)是NoVs黏附因子(受体),能促进病毒感染宿主细胞.NoVs主要衣壳蛋白突出(Protruding,P)区是与HBGAs结合的关键结构域.本研究构建了非流行毒株GII.26型NoVsP区的原核表达重组质粒,以谷胱甘肽巯基转移酶(Glutathione s-transferase,GST)亲和层析纯化P蛋白,人鼻病毒的3C蛋白酶去掉GST标签,通过酶联免疫吸附实验探索P蛋白与HBGAs相互作用的特点,借助同源结构模拟以及结构重叠分析其与相应糖分子之间可能存在的对接位点.结果 表明,P蛋白可与包括A型、B型、AB型、O型和非分泌型的215种唾液中的大部分发生结合,但只与19种寡糖中的H双糖结合;模拟的GII.26P单体的空间构象与GII.17类似,可通过糖结合位点的5个氨基酸与H双糖特异性结合.本研究阐明了GII.26 P蛋白与HBGAs的结合特征及潜在分子机制,为进一步揭示GII.26 NoVs可能的流行趋势及研发潜在抗病毒药物奠定一定的基础. 相似文献
