The NGF content in each region of the brain of four-week-old rats was ranked in the decreasing order of cerebral cortex, hippocampus, cerebellum, midbrain/diencephalon, and pons/medulla ob-longata, and the NGF concentration, in the decreasing order of hippocampus, cerebral cortex, cerebellum, midbrain/diencephalon, and pons/medulla oblongata in both AFD and SFD groups. The NGF content and concentration in the cerebral cortex were about the same value at each age between those in the AFD and SFD groups. Those in the hippocampus were a little higher in the SFD group than in the AFD group at the ages of three and four weeks, unlike those in the other regions, where the values for the cerebellum, midbrain/diencephalon and pons/medulla oblongata tended to be somewhat higher in the AFD group than in the SFD group. The NGF concentrations in the hippocampus and cerebral cortex increased with growth: the concentration in the hippocampus at four weeks of age was about 4-fold of that at one week in the AFD group and about 5.7-fold of that at one week in the SFD group; and likewise the concentration in the cerebral cortex at four weeks of age was about 5.3-fold in the AFD group and about 7-fold in the SFD group. The NGF concentrations in the cerebellum decreased, and those in midbrain/diencephalon and pons/medulla oblongata hardly changed with growth in either AFD or SFD group. From these results NGF may have stronger implications for the neuronal growth in the hippocampus compared with those in the lower brain regions of the SFD rats.  相似文献

作者研究山西阳泉丽斑麻蜥(Eremias argus)成体的选择体温、热耐受性及食物同化和疾跑速的热依赖性。选择体温、临界低温和临界高温无显著的两性差异,分别为36·0℃、1·0℃和44·9℃。在实验温度范围内,体温显著影响食物通过时间、日摄食量、日粪尿排量、表观消化系数和同化效率。食物通过时间在26 -34℃范围内随体温升高而缩短,在更高的体温下则延长。蜥蜴在30、32、34和36℃体温下明显摄入较多的食物、排出明显较多的粪尿。34℃和36℃下的表观消化系数和同化效率大于其它更低或更高温度下的对应数值,但这两个变量未因体温变化而呈现清晰的规律性变化。疾跑速在18 -36℃范围内随体温升高而加快,在38℃体温下则减缓。36℃或附近体温最适合疾跑速。疾跑速最适体温接近蜥蜴的选择体温,表明蜥蜴疾跑速的最适温度可能与其选择体温密切相关[动物学报52 (2) : 256 -262 , 2006]。  相似文献

采用扫描电子显微镜观察人工栽培桑黄子实体的显微形态及其结构,如菌管、子实层、担孢子、锥形刚毛及离散菌丝结构等。分析结果表明:桑黄幼嫩子实体中全氨基酸含量可达11.22%,是成熟子实体(4.28%)的2.62倍,其多糖含量(1.18%)也较成熟子实体(0.64%)高许多。其多糖组分主要为甘露糖、半乳糖、葡萄糖,摩尔比分别为3.820:0.314:1.000(幼嫩子实体)和3.260:0.112:1.000(成熟子实体)。  相似文献

A novel enzyme, alpha-neoagarooligosaccharide hydrolase (EC 3.2.1.-), which hydrolyzes the alpha-1,3 linkage of neoagarooligosaccharides to yield agaropentaose (O-beta-D-galactopyranosyl(1-->4)-O-3,6-anhydro-alpha-L-galactopyranosyl (1-->3)-D-galactose], agarotriose [O-beta-D-galactopyranosyl(1-->4)-O-3,6-anhydro- alpha-L-galactopyranosyl (1-->3)-D-galactose], agarobiose [O-beta-D-galactopyranosyl(1-->4)-3,6-anhydro-L-galactose], 3,6-anhydro-L-galactose, and D-galactose was isolated from the marine bacterium Vibrio sp. strain JT0107 and characterized. This enzyme was purified 383-fold from cultured cells by using a combination of ammonium sulfate precipitation, successive anion-exchange column chromatography, gel filtration, and hydroxyapatite chromatography, gel filtration, and hydroxyapatite chromatography. The purified protein gave a single band (M(r), 42,000) on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Estimation of the M(r) by the gel filtration method gave a value of 84,000, indicating that the enzyme is dimeric. Amino acid sequence analysis revealed it to have a single N-terminal sequence that has no sequence homology to any other known agarases. The optimum temperature and pH were 30 degrees C and 7.7, respectively. The Km and maximum rate of metabolism for neoagarobiose were 5.37 mM and 92 U/mg of protein, respectively.  相似文献

观察白血病抑制因子 (LIF)受体gp190亚基完整的细胞内区和gp190胞内区C末端片段(190CT)对人白血病系HL 6 0表达CD14、CD15的影响 ,进一步了解LIF引发白血病细胞增殖抑制和分化的关系 .用基因重组技术将LIF另一亚基gp130的细胞内区换成gp190的细胞内区 ,用PCR技术扩增gp190细胞内区C末端的一个多肽的编码序列 ,构成嵌合体受体基因 130 /190及 190CT片段 ,并分别在HL 6 0细胞表达 .用免疫组化和流式细胞术检测分析在LIF的诱导下 ,HL 6 0细胞表达CD14、CD15的水平 .转染pcDNA130 /190的HL 6 0细胞 ,CD15表达量明显增高 ;转染pcDNA190CT的细胞 ,CD15的表达量降低 ;但 2组细胞的CD14表达量均较低且水平接近 .LIF可能诱导HL 6 0细胞向粒细胞而不向单核细胞分化 ,该效应是由gp190亚基细胞内区介导的 ,而gp190C末端片段可干扰LIFα受体介导的信号传导效应 .  相似文献

运用物种丰富度S、种群优势度D、群落多样性指数H'和均匀度指数E作为综合指标,分析了不同间作方式下麦田麦蚜Aphidius avenae主要寄生性和捕食性天敌群落的时间动态.结果表明,麦-油间作田天敌物种丰富度多高于单作田和麦-蒜间作田,麦-蒜间作田和单作田天敌物种丰富度基本相同;不同间作处理天敌群落优势种在调查初期有所不同,而中后期皆为燕麦蚜茧蜂;麦-油间作田天敌群落多样性指数一直高于单作田和麦-蒜间作田;调查后期(5月8日后)麦-油间作田的寄生性天敌的优势度指数显著低于单作田和麦-蒜间作田,均匀度指数则显著高于单作田和麦-蒜间作田.说明麦-油间作,麦蚜天敌物种丰富度较高,群落较稳定性,在小麦灌浆初期即可对麦蚜为害起一定控制作用;麦-蒜问作对麦蚜天敌群落的结构特点及时间动态无明显影响.  相似文献

用单细胞凝胶电泳法检测了稀土化合物氯化钇和氯化镨对人外周血淋巴细胞的DNA损伤效应。结果表明,与对照相比,3种不同浓度的氯化钇和氯化镨均可引起淋巴细胞DNA受损后DNA迁移率的显著升高,受损伤细胞的百分率与对照差异明显,提示氯化钇和氯化镨具有一定的遗传毒性。  相似文献

Plinabulin (1, NPI-2358), a potent microtubule-targeting agent derived from the natural diketopiperazine ‘phenylahistin’ with a colchicine-like tubulin depolymerization activity, is an anticancer agent undergoing Phase II clinical trials in four countries including the United States. In order to understand the precise binding mode of plinabulin with tubulin, a new bioactive biotin-tagged photoaffinity probe 4 (KPU-244-B3) was designed and synthesized. Probe 4 showed significant binding affinity to tubulin in a binding assay, and selectively bound to tubulin in an HT-1080 cell lysate without photo-irradiation. In a tubulin photoaffinity labeling study, probe 4 labeled both α- and β-tubulin subunits and these interactions were competitively inhibited by plinabulin during photo-irradiation. These results suggest that plinabulin binds in the boundary region between α- and β-tubulin near the colchicine binding site, and not inside the colchicine binding cavity.  相似文献

Combination of the DNA injection into seminiferous tubules and the subsequent in vivo electroporation (EP) has become an efficient and convenient assay system for spermatogenic-specific gene expression during spermatogenesis of mice. In this study, we made methodological modifications to enhance the transfection efficiency, and evaluated the possibility of this technique to generate transgenic offspring using green fluorescent protein (GFP) as a marker. After the in vivo gene transfer, GFP expression could be monitored easily and repeatedly on the surface of the testis of live mice under fluorescent microscopy. The serial sections of the transfected testis revealed that transient expression of GFP was extended even in the innermost region of the testis uniformly, but confined to spermatogenic cells and Sertoli cells within the seminiferous tubules. Furthermore, long-lasting GFP expression could be detected in the spermatogenic cells even 2 months after EP. Natural mating with normal adult females revealed that 65% of the transfected males maintained fertilizable ability and could generate their offspring normally. Germ-line transmission of the GFP vector to the offspring was checked under fluorescent microscopy, but no transgenic offspring has been detected up to now. These results suggest that the application of additional techniques, such as cell sorting for GFP-positive germ cells followed by nuclear transfer to the oocytes, would make this method as a novel strategy for generating transgenic animals. 实验对树干毕赤酵母(Pichia stipitis)进行了4个阶段共400 h连续恒化培养,在不同阶段以30.0 g/L葡萄糖作为基本碳源,添加30.0或15.0 g/L的木糖,通过控制温度(35±1)℃,进气量100~150mL/min,搅拌转速250~300 r/min。4个阶段共建立4个连续培养的"稳态"。对碳元素进行物料衡算发现,四个阶段碳元素回收率分别为118.0 %、105.6 %、113.5 %和94.7 %。对4个近似"稳态"的碳元素的代谢流向进行分析发现:将近50.0 %左右碳元素流向产物酒精,其次是CO2和酵母细胞;木糖醇浓度与流入底物中木糖浓度有直接关系,在相同发酵条件下流入的木糖浓度越大代谢生成木糖醇浓度也越高;实验所采用的通气条件更适合底物为30.0 g/L葡萄糖和30.0 g/L木糖混合液的连续发酵。  相似文献
