本研究于2021年3~9月,采用目标观察和全事件记录法,对广西防城港市钦州湾八路水湿地黑翅长脚鹬(Himantopus himantopus)的繁殖习性进行全过程观察记录。黑翅长脚鹬的栖息生境主要在盐田、虾塘和鱼塘,而巢主要分布在盐田生境。共发现39巢,雌雄共同营巢,按照主要巢材将其巢分为干草巢、碎石巢、泥皮巢和牛毛毡草巢4种;巢材包括禾本科(Gramineae)和莎草科(Cyperaceae)植物以及碎石、贝壳等;巢外径为(23.3±10.7)cm,巢内径为(11.2±1.9)cm,巢深为(1.6±0.5)cm,巢高为(6.5±4.3)cm(n=39);筑巢需(3±2)d(n=6)。窝卵数2~4枚,1~2 d产1枚卵,7 d内产完满窝卵(n=6)。雌雄均参与孵卵,雄性孵卵时间比雌性长,但二者差异不显著(P> 0.05),雄性(8 550±245.9)min,雌性(7 530±263.3)min,孵卵期为(25±2)d(n=6)。育雏期(26±3)d(n=6),雌雄轮流育雏,育雏前、中期(雏鸟1~20d日龄),雌性育雏时间比雄性长,是雄性的2倍,育雏后期(雏鸟大于20 d日龄),...  相似文献

【背景】传统制作奶豆腐和酸性奶油(乌日莫)是内蒙古农牧地区最喜爱的食品,蕴含着十分丰富的乳酸菌资源,亟待开发利用。【目的】通过解析内蒙古农牧地区传统自制奶豆腐和乌日莫样品中乳酸菌多样性及分布特征,为优良菌株选育与利用提供资源和理论基础。【方法】采用稀释涂布法分离纯化乳酸菌,测定菌株16S rRNA基因序列鉴定种属关系,阐明乳酸菌系统发育、遗传分化及菌群结构。【结果】传统自制样品中共分离得到乳酸菌81株,主要归属于乳酸片球菌(Pediococcus acidilactici)、戊糖片球菌(Pediococcus pentosaceus)、短乳杆菌(Lactobacillus brevis)、瑞士乳杆菌(Lactobacillus helveticus)、副干酪乳杆菌(Lactobacillus paracasei)、食二酸乳杆菌(Lactobacillus diolivorans)、奥塔基乳杆菌(Lactobacillus otakiensis)、植物乳杆菌(Lactobacillus plantarum)、开菲尔乳杆菌(Lactobacillus kefir)、乳酸乳球菌(Lactoc...  相似文献

Summary A model antigen, rabbit immunoglobulin G, was immobilized onto polyester cloth by adsorption. The antigen cloth was reacted with sheep anti-rabbit IgG antibody. Antibody bound to the antigen cloth was nearly quantitatively eluted by saturated MgCl2, whereas a commercial antibody eluent slowly eluted only about 70 % of the antibody. Exposure of antibody to saturated MgCl2 for 30 min resulted in no loss of immunoactivity. Saturated MgCl2, therefore, is an ideal eluent in immunoaffinity purification of antibodies.  相似文献

泛素化是存在于真核生物中一种重要的翻译后修饰过程,参与调控包括蛋白质降解在内的多种生命活动。实现这一调控过程需要将一个由76个氨基酸组成的泛素蛋白共价连接到底物蛋白上。同时,泛素本身也存在多种翻译后修饰,包括泛素化、磷酸化、乙酰化等,进一步丰富了泛素的修饰类型,决定了底物蛋白不同的命运。近年来,伴随着第65位丝氨酸磷酸化泛素蛋白参与调控线粒体自噬这一突破性进展,泛素蛋白其余磷酸化位点的功能研究也获得越来越多的关注。本文根据目前已有的国内外研究和报道,总结了泛素蛋白已知的磷酸化修饰位点,梳理了泛素蛋白第12位和66位苏氨酸、第57位和65位丝氨酸等位点的磷酸化修饰对其生物物理特性带来的改变,并对相应修饰位点所涉及的生物学功能调控进行了综述。  相似文献

蜜环菌属Armillaria真菌具有较高的食药用价值。由于蜜环菌的生长发育过程较复杂,还未完全实现商业化栽培,野生资源的供应受到季节性和地域性的影响。本研究以采自东北地区蜜环菌属的3个菌株为研究对象,通过培养物的形态特征及分子标记确定菌株JG19016为奥氏蜜环菌A. ostoyae,菌株JG19017为高卢蜜环菌A. gallica,菌株JG19018为中国蜜环菌生物种C。奥氏蜜环菌JG19016最适生长温度为25 ℃,高卢蜜环菌JG19017的最适生长温度为22 ℃,中国蜜环菌生物种C JG19018则在22-25 ℃时菌丝生长速度最快;3个菌株最适pH为5-6。奥氏蜜环菌JG19016对葡萄糖和蔗糖利用率较好,高卢蜜环菌JG19017对葡萄糖利用率较好,中国蜜环菌生物种C JG19018对葡萄糖和淀粉利用率较好;蛋白胨对3个菌株促进作用最强,为最适氮源。培养基中加入VB1,对3个菌株的菌丝生长均有明显的促进作用。奥氏蜜环菌JG19016菌丝生长的最优培养基配方为:葡萄糖20 g,蛋白胨3 g,磷酸二氢钾2 g,硫酸镁1.5 g,VB1 10 mg,琼脂20 g,水1 L。在木屑基质中培养,其配方的最优碳氮比为38:1,最佳木屑粗细比为3:1以上。出菇条件探索结果显示,菌丝及菌索长满菌袋(17 mm×33 mm×5 mm丝聚乙烯袋)需要50-60 d,之后在18 ℃、60%湿度和12 h散射光的环境中,10 d左右可观察到原基产生。增加菇房湿度到90%-95%,2-3 d可观察到1-3 cm的幼子实体,7 d左右菌柄和菌盖完全分化,10 d左右观察到菌盖展开。  相似文献

Restoration of cytoplasmic male sterility (CMS) in sunflower was demonstrated to be controlled by polygenes by analysing 982 effective crosses among 109 self-crossed lines and 16 CMS lines. Two self-crossed lines and one CMS line with distinct genotypes were applied to creation of segregating populations for DNA bulks of the target gene Rfl. Bulked DNA was prepared in order to investigate single gene Rfl and its gene marker among polygenic characters at the same genetic background. Using 80 10-mer operon primers, 620 RAPD reactions were carried out between fertile and sterile DNA bulks. In about 800 loci, primary results showed that 8 were related to the restoration genes. Furthermore. 2 were confirmed as RAPD markers for gene Rfl by examining 9 maintenance and 7 restoration lines. This method is the improvement for bulked segregant analysis[1] with which markers of single gene of target can be identified rapidly among polygenic characters.  相似文献

Carotenogenic mutants ofPhycomyces, which accumulate excess β-carotene or its intermediates, always failed in zygospore development. No improvement occurred when such mutants were mated together with a helper wild type of the same mating type against the wild type of the opposite mating type. Addition of excess synthesized pheromone, trisporin B, also failed to improve the zygospore development, though the mating response was significantly activated in the early stages and abundant zygophores were formed. Exceptional acceleration of the zygospore development under these experimental conditions occurred in a regulatory albino mutant (carA), which does not accumulate excess intermediate carotenoids. Chemically- or genetically-induced ovarproduction of β-carotene or lycopene also inhibited the zygospore development. These results imply that the zygospore development ofPhycomyces is maximal when the intracellular amount of β-carotene is optimal (=wild type), and that pheromones act mainly in the early stages of mating, while other factors such as the cell-to-cell recognition system may also be involved in the later stages. Intracellular accumulation of excess β-carotene or its intermediates probably disturb such later-stage factors.  相似文献

From the analysis of restriction maps of the Amy region in eight sibling species belonging to the Drosophila melanogaster species subgroup, we herein show that the patterns of duplication of the Amy gene are almost the same in all species. This indicates that duplication occurred before speciation within this species subgroup. From the nucleotide sequence data, we show a strong within-species similarity between the duplicated loci in the Amy coding region. This is in contrast to a strong similarity in the 5' and 3' flanking regions within each locus (proximal or distal) throughout the species subgroup. This means that concerted evolution occurred only in the Amy coding region and that differentiated evolution between the duplication occurred in the flanking regions. Moreover, when comparing the species, we also found a significant excess of nonsynonymous substitutions. In particular, all the fixed substitutions specific to D. erecta were found to be nonsynonymous. We thus conclude that adaptive protein evolution occurred in the lineage of D. erecta that is a ``specialist' species for host plants and probably also occurs in the process of speciation in general.  相似文献

LINE-like retrotransposons, the so-called I elements, control the system of I-R (inducer-reactive) hybrid dysgenesis in Drosophila melanogaster. I elements are present in many Drosophila species. It has been suggested that active, complete I elements, located at different sites on the chromosomes, invaded natural populations of D. melanogaster recently (1920–1970). But old strains lacking active I elements have only defective I elements located in the chromocenter. We have cloned I elements from D. melanogaster and the melanogaster subgroup. In D. melanogaster, the nucleotide sequences of chromocentral I elements differed from those on chromosome arms by as much as 7%. All the I elements of D. mauritiana and D. sechellia are more closely related to the chromosomal I elements of D. melanogaster than to the chromocentral I elements in any species. No sequence difference was observed in the surveyed region between two chromosomal I elements isolated from D. melanogaster and one from D. simulans. These findings strongly support the idea that the defective chromocentral I elements of D. melanogaster originated before the species diverged and the chromosomal I elements were eliminated. The chromosomal I elements reinvaded natural populations of D. melanogaster recently, and were possibly introduced from D. simulans by horizontal transmission.  相似文献

A large proportion of spontaneous mutations inDrosophila melanogaster strains of laboratory origin are associated with insertions of mobile DNA elements. As a first step toward determining whether spontaneous laboratory mutations are predictive for mutational events occurring in the wild, recessivebrown (bw) eye color mutants were isolated. By inbreeding the progeny of wild-caughtDrosophila melanogaster females,bw mutations were isolated from seven separate geographic sites distributed among Japan, California, Siberia and Hungary. Among a total of 14 mutations studied, no case of transposon mutagenesis was found. At least 4 mutations are associated with small deletions in thebw gene. The remainder are inseparable from wild-typebw by Southern analysis and are presumed to be basepair changes or very small indels. Although only two spontaneousbw mutants of laboratory origin have been analyzed molecularly, one is a mobile element insertion.  相似文献
