Catalysis of collagen degradation by matrix metalloproteinase 1 (MMP-1) has been proposed to critically rely on flexibility between the catalytic (CAT) and hemopexin-like (HPX) domains. A rigorous assessment of the most readily accessed conformations in solution is required to explain the onset of substrate recognition and collagenolysis. The present study utilized paramagnetic NMR spectroscopy and small angle x-ray scattering (SAXS) to calculate the maximum occurrence (MO) of MMP-1 conformations. The MMP-1 conformations with large MO values (up to 47%) are restricted into a relatively small conformational region. All conformations with high MO values differ largely from the closed MMP-1 structures obtained by x-ray crystallography. The MO of the latter is ∼20%, which represents the upper limit for the presence of this conformation in the ensemble sampled by the protein in solution. In all the high MO conformations, the CAT and HPX domains are not in tight contact, and the residues of the HPX domain reported to be responsible for the binding to the collagen triple-helix are solvent exposed. Thus, overall analysis of the highest MO conformations indicated that MMP-1 in solution was poised to interact with collagen and then could readily proceed along the steps of collagenolysis.  相似文献

This study evaluated the relative occurrences of BK virus (BKV) and JC virus (JCV) infections in patients with chronic kidney disease (CKD). Urine samples were analysed from CKD patients and from 99 patients without CKD as a control. A total of 100 urine samples were analysed from the experimental (CKD patients) group and 99 from the control group. Following DNA extraction, polymerase chain reaction (PCR) was used to amplify a 173 bp region of the gene encoding the T antigen of the BKV and JCV. JCV and BKV infections were differentiated based on the enzymatic digestion of the amplified products using BamHI endonuclease. The results indicated that none of the patients in either group was infected with the BKV, whereas 11.1% (11/99) of the control group subjects and 4% (4/100) of the kidney patients were infected with the JCV. High levels of urea in the excreted urine, low urinary cellularity, reduced bladder washout and a delay in analysing the samples may have contributed to the low prevalence of infection. The results indicate that there is a need to increase the sensitivity of assays used to detect viruses in patients with CDK, especially given that polyomavirus infections, especially BKV, can lead to a loss of kidney function following transplantation.  相似文献

The aim of this work was to create a predictive model of the potential geographical distribution of the leaf-cutting ant Atta robusta Borgmeier 1939 (Hymenoptera: Formicidae) using ecological niche modeling. We used previous collection records available in the literature together with our database. This is the first study in Brazil using ecological niche modeling as a tool for predicting ant species distribution. Besides the already known distribution, the model generated results showing a medium to low probability of occurrence in the coastal regions of Paraná and São Paulo states, and a very high probability of occurrence in southern Bahia State, in environments popularly known as muçunungas. These environments are characterized by a lower density of trees and the dominance of herbaceous-shrub vegetation. Moreover, we discuss how geographic barriers and anthropogenic pressure in restinga environments may be the determining factors in the current distribution of this species. According to the predictive model used in this study, there is strong evidence that this species occurs only in these states and deserves special attention from government conservation agencies in the restingas where it is located.  相似文献

Heat stress is a major production constraint of sunflower worldwide. Therefore, various populations (parental, F1, F2, F3, and plant progenies) of sunflower were screened for leaf gas-exchange traits with the objectives to formulate selection criteria of heat resistance and development of heat-resistant lines. Initial screening and F2 seeds exposed to heat stress (45°C) resulted in the development of an adapted F2 population that showed leaf gas-exchange and morphological traits better than the unadapted population. Correlation coefficients of traits were partitioned into direct and indirect effects via a path analysis technique to determine the cause of their relationship with a basic parameter such as a reproductive head mass (HM). Path analysis showed a positive direct effect of leaf temperature (Tleaf) (0.32) on HM and also an indirect effect (0.77) of the transpiration rate (E) on HM. Moreover, Tleaf showed high heritability estimates. Tleaf was used to select superior plants within the F2 population. This selection brought about an improvement in the net photosynthetic rate (P N) and E as it was indicated from progeny performance and realized heritability. Progenies selected on the basis of Tleaf also showed an increase in achene yield and heat resistance over unselected F3 progenies and a commercial hybrid.  相似文献

Recent studies have indicated a causal link between high dietary cholesterol intake and brain oxidative stress. In particular, we have previously shown a positive correlation between elevated plasma cholesterol levels, cortico-cerebral oxidative stress and mitochondrial dysfunction in low density lipoprotein receptor knockout (LDLr?/?) mice, a mouse model of familial hypercholesterolemia. Here we show that the organoselenium compound diphenyl diselenide (PhSe)2 (1 mg/kg; o.g., once a day for 30 days) significantly blunted the cortico-cerebral oxidative stress and mitochondrial dysfunction induced by a hypercholesterolemic diet in LDLr?/? mice. (PhSe)2 effectively prevented the inhibition of complex I and II activities, significantly increased the reduced glutathione (GSH) content and reduced lipoperoxidation in the cerebral cortex of hypercholesterolemic LDLr?/? mice. Overall, (PhSe)2 may be a promising molecule to protect against hypercholesterolemia-induced effects on the central nervous system, in addition to its already demonstrated antiatherogenic effects.  相似文献

Progress in biotechnological research over the last two decades has provided greater scope for the improvement of crops, forest trees and other important plant species. Plant propagation using synthetic seeds has opened new vistas in the field of agriculture. Synseed technology is a highly promising tool for the management of transgenic and seedless plant species, polyploid plants with elite traits and plant lines that are difficult to propagate through conventional propagation methods. Delivery of synseeds also alleviates issues like undertaking several passages for scaling up in vitro cultures as well as acclimatization to ex vitro conditions. Optimization of synchronized propagule development followed by automation of the whole process (sorting, harvesting, encapsulation and conversion) can enhance the pace of synseed production. Cryopreservation of encapsulated germplasm has now been increasingly used as an ex vitro conservation tool with the possible minimization of adverse effects of cryoprotectants and post-preservation damages. Through synseed technology, germplasm exchange between countries could be accelerated as a result of reduced plant quarantine requirements because of the aseptic condition of the plant material.  相似文献

Haemoglobin I from Lucina pectinata is a monomeric protein consisting of 142 amino acids. Its active site contains a peculiar arrangement of phenylalanine residues (PheB10, PheCD1 and PheE11) and a distal Gln at position E7. Active site mutations at positions B10, E7 and E11 were performed in deoxy haemoglobin I (HbI), followed by 10 ns molecular dynamic simulations. The results showed that the mutations induced changes in domains far from the active site producing more flexible structures than the native HbI. Distance analyses revealed that the heme pocket amino acids at positions E7 and B10 are extremely sensitive to any heme pocket residue mutation. The high flexibility observed by the E7 position suggests an important role in the ligand binding kinetics in ferrous HbI, while both positions play a major role in the ligand stabilisation processes. Furthermore, our results showed that E11Phe plays a pivotal role in protein stability.  相似文献

The Reactive Nitrogen and Oxygen Species (the so-called RNOS), which are well-known radicals formed in the mitochondria under nitro-oxidative cell stress, are responsible for nitration of tyrosines in a wide variety of proteins and, in particular, in cytochrome c (Cc). Only three out of the five tyrosine residues of human Cc, namely those at positions 67, 74 and 97, have been detected in vivo as nitrotyrosines. However, nitration of the two other tyrosines, namely those at positions 46 and 48, has never been detected in vivo despite they are both well-exposed to solvent. Here we investigate the changes in heme coordination and alkaline transition, along with the peroxidase activity and in cell degradation of Cc mutants in which all their tyrosine residues - with the only exception of that at position 46 or 48 - are replaced by phenylalanines. In Jurkat cell extracts devoid of proteases inhibitors, only the high-spin iron nitrated forms of these monotyrosine mutants are degraded. Altogether the resulting data suggest that nitration of tyrosines 46 and 48 makes Cc easily degradable upon turning the heme iron state to high-spin.  相似文献

This case study examines the pollen dispersal distance, pollen dispersal patterns and intra‐family genetic structure for isolated trees in pastures of the bat‐pollinated Neotropical tree species Hymenaea stigonocarpa using six microsatellite loci and parentage analysis. The sampling included 28 grouped trees (referred to as the population) and six isolated trees in pastureland along a highway in Mato Grosso do Sul State, Brazil. From the population, we sampled 137 seeds from 12 seed‐trees, and from the isolated trees, we sampled 34 seeds from two seed‐trees. The results showed that pollen was dispersed over long distances (reaching 7353 m) and therefore the spatially isolated trees were not reproductively isolated. The pollen immigration rate in the population was also high (31%). Isolated trees presented a higher selfing rate (s=26%) than trees in the population (s=12%), suggesting that the spatial isolation of the trees increased selfing. However, selfing was responsible for only 30 percent of the inbreeding in offspring and mating among relatives was 70 percent. In the population, excluding selfing, ca 72 percent of the pollen was dispersed over distances <1000 m (average: 860 m). For the two isolated seed‐trees, excluding selfing, the average pollen dispersal distance was 5229 m. The results demonstrate that although pollen can be dispersed over long distances for H. stigonocarpa isolated trees, a high percentage of pollen comes from the same tree (selfing) and mating was correlated. Consequently, seeds must be collected from a large number of seed‐trees for conservation purposes.  相似文献
