从川楝子(fruitsofMeliatoosendanSieb.etZucc.)的水溶性成分中分离出两个新的苯丙三醇甙:川楝甙A(3-甲氧基-5-羟基-9-(1’-O-β-D-葡萄糖)-苏式-苯丙三醇)和川楝甙B(4-羟基-7,8-(2’,1’-O-β-D-葡萄糖)-苯丙三醇);同时首次分离出苏式-愈创木基甘油。通过波谱解析和化学方法确定了它们的结构。  相似文献

分别用浓度为25mmol/L、50mmol/L、100mmol/L和200mmol/L的NaCl、Na2SO4和Na2CO3的营养液培养小麦4d,较之不含盐的营养液,其自由基含量上升,产生速率增加,叶片质膜透性增加。不同盐的影响也不同,在低浓度时,NaCl的影响大于Na2SO4,高浓度时,NaCl影响小于Na2SO4,Na2CO3的影响最为显著。实验结果也表现出小麦叶片自由基含量和质膜透性呈现较好的相关性。因此可认为,盐胁迫促使自由基含量增加,自由基通过过氧化作用影响质膜透性,从而影响植物的生长。  相似文献

建立了纯化光合膜蛋白质复合体Cytb6f的新方法。与现有方法相比,新方法简单明了,只包括透析离心和用硫酸铵分级沉淀两个步骤,而且适于大批量制备。按此方法从菠菜叶绿体分离纯化的制剂含9.8nmolCytf·mg-1;SDSPAGE显示4条主要区带及1条低分子量区带,背景干净;Cytb6(b型血红素)∶Cytf=2∶1(mol∶mol),Chla∶Cytf=1∶1(mol∶mol);酶活性(PQ2H2→Cytc)80μmolCytc·nmolCytf-1·h-1左右;产率可达38%。  相似文献

通过外源性底物对[γ-32P]-ATP的摄入量来测定豆蔻酰佛波醇乙酯(phorbol-myristate-acetate,PMA)处理后的人脐静脉内皮细胞(humanumbilicalveinendothelialcells,HUVECs)膜蛋白激酶C(proteinkinaseC,PKC)的活性;利用间接免疫荧光标记和Western印迹方法分析蛋白激酶C活性对锚蛋白及CD44的亚细胞分布及蛋白质表达的影响。结果发现HUVECs的锚蛋白及CD44表达水平趋势与PKC活性变化相吻合;PKC活化导致CD44在细胞膜上呈聚集状,而锚蛋白则移位并聚集于CD44处;PKC抑制剂能抑制PKC活化所带来的上述作用。结果表明PKC活化通过磷酸化作用能上调锚蛋白及CD44表达,并同时导致二者发生一致性运动及共分布。  相似文献

The accuracy of protein synthesis relies on the ability of aminoacyl-tRNA synthetases (aaRSs) to discriminate among true and near cognate substrates. To date, analysis of aaRSs function, including identification of residues of aaRS participating in amino acid and tRNA discrimination, has largely relied on the steady state kinetic pyrophosphate exchange and aminoacylation assays. Pre-steady state kinetic studies investigating a more limited set of aaRS systems have also been undertaken to assess the energetic contributions of individual enzyme-substrate interactions, particularly in the adenylation half reaction. More recently, a renewed interest in the use of rapid kinetics approaches for aaRSs has led to their application to several new aaRS systems, resulting in the identification of mechanistic differences that distinguish the two structurally distinct aaRS classes. Here, we review the techniques for thermodynamic and kinetic analysis of aaRS function. Following a brief survey of methods for the preparation of materials and for steady state kinetic analysis, this review will describe pre-steady state kinetic methods employing rapid quench and stopped-flow fluorescence for analysis of the activation and aminoacyl transfer reactions. Application of these methods to any aaRS system allows the investigator to derive detailed kinetic mechanisms for the activation and aminoacyl transfer reactions, permitting issues of substrate specificity, stereochemical mechanism, and inhibitor interaction to be addressed in a rigorous and quantitative fashion.  相似文献

BackgroundStates of depression are associated with increased sensitivity to negative events. For this novel study, we have assessed the relationship between the number of depressive episodes and the dysfunctional processing of emotional facial expressions.Conclusion/SignificanceThese results provide new evidence that having more recurrent depressive episodes and serious depressive states are likely to aggravate the already abnormal processing of emotional facial expressions in patients with depression. Moreover, it further suggests that the impaired processing as indexed by N170 amplitude for positive face identification may be a potentially useful biomarker for predicting propagation of depression while N170 amplitude for negative face identification could be a potential biomarker for depression recurrence.  相似文献

It is indispensable to comprehend the mechanism that regulates plant responses to drought conditions to intensify the water use efficiency of stone fruits. The physiological, biochemical and molecular responses of drought-treated peach leaves were investigated. Results revealed that drought-treated plants manifested a significant attenuation in water potential as compared to control plants. Furthermore, sorbitol and proline contents were accumulated contrary to glucose, fructose, and sucrose that were dwindled significantly throughout the drought period. Similarly, the activities of antioxidant enzymes and expression pattern of related genes were hoisted to counter the lipid peroxidation in drought-treated plants. Moreover, reduced stomatal conductance has repressed the photosynthesis process and linked genes during drought stress. The expression level of regulatory genes (dehydration-responsive element-bindings and WRKYs) exhibited up-regulation in the drought-treated group. Overall, this study asserts that ‘Yoshihime’ peach cultivar possesses unique physiological, biochemical, and molecular responses under different spells of drought stress.  相似文献
