湖北地区宫颈癌组织中人乳头瘤病毒16型E7基因的分离、克隆和序列分析

采用加端聚合酶链反应技术,从湖北地区一宫颈癌患者癌组织ＤＮＡ中分离出人乳头瘤病毒１６型（ＨＰＶ１６）Ｅ７基因,并在ｐＵＣ１８载体中克隆。经限制性核酸内切酶分析和ＤＮＡ序列分析,确认了含ＨＰＶ１６Ｅ７重组克隆质粒,命名ｐＨＰＶ１６Ｅ７─ＨＢ。ＤＮＡ序列分析表明,ＨＰＶ１６Ｅ７─ＨＢ基因全长２９４ｂｐ（与报道的标准株基因长度相同）,但其核苷酸顺序中有两处发生了Ｃ→Ｔ突变,即第４３位密码子ＣＡＡ变为ＴＡＡ,第７６位ＣＧＴ变为ＴＧＴ;前者使谷氨酰胺密码子变为终止密码,即无义奕变（ｎｏｎｓｅｎｓｅｍｕｔａｔｉｏｎ）。这种突变发生在２９４个碱基的ＤＮＡ扩增产物之中,不像是ＰＣＲ本身的错配,而很可能是湖北株与标准株之间的结构差异。     

Induction of the differentiation of human HL-60 promyelocytic leukemia cell line by succinoyl trehalose lipids

Four analogs of succinoyl trehalose lipid-3 (STL-3)with saturated even-number or odd-number carbonchains, and unsaturated or halogenated fatty acidswere examined for their ability to inhibit the growthand induce the differentiation of HL-60 humanpromyelocytic leukemia cells. The optimalconcentration of STL-3 at which such activities wererecognized was closed to the critical micelleconcentration of STL-3. Analog of STL-3 witheven-number or odd-number carbon chain and unsaturatedfatty acids strongly inhibited growth and induced thedifferentiation of HL-60 cells, as evaluated in termsof nitroblue tetrazilium-reducing activity and theappearance of the CD36 antigen. An analog of STL-3with halogenated fatty acids significantly inhibitedproliferation but only induced the differentiation ofHL-60 cells. Our results indicate that the effects ofSTL-3 and its analogs on HL-60 cells depend on thestructure of the hydrophobic moiety of STL-3.These authors contributed equally to this work     

葛洲坝枢纽下游白鲟性腺发育的初步观察

白鲟Psephurus gladius(Martens)是我国特有的稀珍鱼类,近年来对其生物学做了一些初步的研究。鉴于它的种群较小,在学术上具有较为重要的价值,所以,长江兴修水利工程以后对其资源的影响及资源保护和增殖问题,特别是聚集于坝下江段的白鲟亲鱼性腺能否发育成熟等问题,和中华鲟一样引起了水产科学工作者的普遍关注。       

Antitumor mechanism of Se-containing polysaccharide, a novel organic selenium compound

Recent studies on the inhibition of tumor growth by Se-containing polysaccharide were reviewed. Meanwhile, the possible molecular mechanisms of the inhibition of tumor cell growth through antioxidation, induction of tumor cell apoptosis, blockade of cell cycle, and enhancement of immunity by Se-containing polysaccharide were proposed. In the end, the potential application of Se-containing polysaccharide in the prevention and treatment of tumor was elucidated.     

细菌样颗粒——新型乳酸菌表面展示技术及其应用

细菌样颗粒(Bacterium-like particles,BLPs)是一种新型非遗传修饰型乳酸菌表面展示技术,外源蛋白可通过锚钩蛋白锚定于经热酸处理而得的乳酸菌肽聚糖骨架表面,形成空心表面展示颗粒。因其安全性高、表面展示密度大、黏膜递送效率高,又兼有佐剂效应,BLPs广泛应用于黏膜疫苗和黏膜佐剂的开发、病毒抗原的纯化、生物催化剂的制备等领域。本文就BLPs的构建、独特优势、目前的应用及尚需解决的问题等方面进行详细综述,以期展现BLPs新型表面展示平台的广阔应用前景。     

应用纤维素结合域标签构建谷氨酸棒状杆菌表达系统

为优化谷氨酸棒状杆菌表达系统的纯化工艺,合成里氏木霉的CBD基因,将其与谷氨酸棒状杆菌分泌表达载体pXMJ19-sp连接,构建以CBD为纯化标签的重组载体pXMJ 19-sp-CBD.在该载体中插入GFP基因并转化至谷氨酸棒状杆菌,可获得分泌表达融合蛋白GFP-CBD的重组菌.该菌经IPTG诱导后的发酵液在紫外灯下显示强烈的绿色荧光,重组蛋白的分泌表达量达200 mg/L.利用CBD标签对纤维素柱的可逆性吸附,可直接对谷氨酸棒状杆菌分泌到培养基中的重组蛋白进行纯化,从而简化工艺和降低成本,为工业化大生产奠定基础.
