Bh (black at hatch) is a mutation of Japanese quails which causes darkening or lightening of the plumage in heterozygotes or homozygotes, respectively. We chemically analyzed melanin pigments in feather germs of Bh mutant embryos and in feathers of adult animals. Dark brown dorsal feathers of wild-type adult animals had white barrings, but heterozygous ones lacked clear barrings. The feathers of wild-type and heterozygote animals contained both eumelanins and pheomelanins, the latter being more pheomelanic. On the dorsal skin of 10-day old wild-type embryos, longitudinal stripes from black and yellow rows of feather germs developed; two or three longitudinal rows of black feather germs and then two or three rows of yellow feather germs next to the short central feather germs. Heterozygous embryos appeared black in plumage pigmentation, due to the presence of‘gray’feather germs in rows of dorsal feather germs that corresponded to yellow rows in wild-type embryos. Homozygous dorsal feather germs did not develop the black and yellow longitudinal stripes, but were brown. Chemical analysis showed that embryos of each genotype contained both eumelanins and pheomelanins in the feather germs; however, the eumelanin content in ho-mozygous feather germs was very low. These results suggest that the Bh mutation causes pheomelanic changes in feathers of quails.  相似文献

The vertical distribution of dissolved greenhouse gases (CH4,CO2 and N2O), NO ,and 13C of CO2 in Lake Biwa during a stagnantperiod was precisely determined. CO2 as well as NO was accumulated in the hypolimnion, whereas NO and CH4concen\-trations were generally higher in theepilimnion than in the hypolimnion. In August, NO andCH4 were ephemerally accumulated at the thermocline. Theconcentration of CH4 always exceeded equilibrium with respectto air/water exchange. N2O was rather uniformly distributed inboth time and space, and remained near equilibrium with respect toair/water exchange. All of these observations are similar to otherstratified, oligotrophic lakes, in which the hypolimnia were welloxygenated. The 13C of CO2 became morenegative with increasing depth, and showed a strong negativecorrelation with apparent oxygen utilization. From the data, the 13Cvalue of organic matter decomposed into CO2 inthe hypolimnion was calculated by isotope mass-balance, and found tobe in a similar range to 13C of phytoplankton and benthic algaeand distinctively higher than 13C of both terrestrial andsedimentary organic matters. This suggests that autochthonous organicmatter was the major source of CO2.  相似文献

Melanocytes synthesise two types of melanin: the brown-black eumelanin and the red-yellow phaeomelanin. In mice, the relative proportions of these two melanins are regulated by α-MSH, which preferentially increases the synthesis of eumelanin and by the Agouti protein (AP), the expression of which correlates with the growth of yellow phaeomelanin-containing hair. It has been proposed that AP acts by antagonizing the action of α-MSH at the MCI receptor, although it has been suggested that it may also act independently of α-MSH. In the present study we show that AP inhibits melanogenesis in B16F1 melanoma cells in the presence and absence of α-MSH and also causes dose-related decreases in the synthesis of both eumelanin and phaeomelanin. In the presence of α-MSH AP had a greater effect on eumelanin production and this is consistent with an antagonistic action at the MCI receptor. In the absence of α-MSH however, AP produced similar reductions in the synthesis of both melanins. These changes were not seen in B16G4F cells which lack the MCI receptor, suggesting that even in the absence of α-MSH AP acts at the MCI receptor. How this action is mediated at the intracellular level is not yet clear, although it appears to be associated with a decrease in tyrosinase activity.  相似文献

Pluripotent, feeder-dependent teratocarcinoma cell lines were cultured without a feeder layer in a medium containing 10 −;4 M β-mercaptoethanol (β-medium) and compared for the development of early markers with cells cultured with a feeder layer. The cells cultured in β-medium lost the PNA (peanut agglutinin) receptor typically at low density. This change was accompanied with enhanced secretion of plasminogen activator and the loss of sensitivity to anti-F9 serum, indicating the stem cell differentiation. In contrast, the cells cultured on a feeder layer did not show any marker changes, thus indicating the lack of differentiation. These results indicate that while the presence of feeder cells inhibits cell differentiation, cultivation in β–medium permits the differentiation of pluripotent teratocarcinoma cells.  相似文献

Triplet conjugants of Paramecium multimicronucleatum, in each of which the third cell unites by its anterior and to a posterior part of one of the conjugating pair, are induced by conjugation inducing chemicals, KCl and acriflavine in a Ca11-poor condition. If the triplets are prematurely transferred to medium free of such chemicals, the third cells usually separate and no macronuclear fragmentation occurs in them. But in third cells of long lasting triplets, macromuclear fragmentation regularly begings at ~12 In after chemical treatment Staluhzaton of cell union and determmation of macronuclear fragmentation occured between ~100 and ~130 min after initiation of chemical treatment. Electron-microscopic observation of the stabilized third cell revealed that the two cell membranes at the uniting region were partially fused to form cytoplasmic bridges. A new experimental system for investigating platyfish-swordtail hybrid melanomas was developed using ornamental platyfish ( Xiphophorus maculatus ) and swordtails ( Xiphophorus helleri ). The incidence of melanomas in the hybrids was studied in relation to individual age factors. Based on the age of the onset of the melanomas, two types of the melanomas were discerned and were designated as the fry and the adult melanomas, respectively. The fry melanomas were characterized by early onset, wide-ranged lesions, an invasive nature owing to densely pigmented cells, and a tendency toward regression. The adult melanomas were characterized by occurrence in the later stage of life, a small number of primary sites, and rapid, expanding, and invasive growth due to sparsely pigmented cells.  相似文献

The color of hair and wool in mammals and feathers in birds is mostly determined by the quantity and quality of melanins that are synthesized in follicular melanocytes and transferred to keratinocytes. There are two chemically distinct types of melanin pigments: the black to brown eumelanins and the yellow to reddish pheomelanins. Melanins in sheep wool and human hair of various colors were characterized by HPLC methods to estimate 5,6-dihydroxyindole-2-carboxylic acid (DHICA)-derived units in eumelanins and benzothiazine units in pheomelanins. Melanins were also characterized by spectrophotometric methods after differential solubilization in alkalies. It was demonstrated that 1) black wool in Asiatic sheep contains eumelanin with the DHICA content similar to black mouse melanin, while black to brown melanins from human hair contain much lower ratios of DHICA-derived units, comparable to the slaty mutation in mice, 2) dark brown to brown hair in human contains eumelanin whose chemical properties are indistinguishable from those of black hair, 3) dark red wool and red human hair contain pheomelanic pigments whose chemical properties are rather different from those of yellow pheomelanins in mice, and 4) light brown, blonde, and red hairs in human can be differentiated from each other with this methodology.  相似文献

In mallards the bright nuptial plumage of the drake represents the neutral, sex hormone-independent coloration of the species that both sexes eventually exhibit after castration. We compared the pheo- and eumelanin contents of feathers from the head, breast, flank, and under-tail coverts in five groups of mallards after the post-nuptial molt in summer: intact hens, intact drakes, castrated drakes, castrated drakes injected with testosterone during the spring, and castrated drakes injected with 5α-dihydrotestosterone during the spring. In the head feathers and under-tail coverts, the gonadal hormones of the intact birds and the testosterone injections into castrates significantly reduced the eumelanin content, tended to increase the pheomelanin content, and, thereby, changed the melanin type from eumelanic in the untreated castrates to mixed melanic in the other three groups. The eumelanin contents of the flank feathers did not differ among the groups, but the pheomelanin contents at this site was significantly elevated in the two intact groups and the testosterone-treated compared to the uninjected castrates. Again, the melanin type changed from eumelanic in the castrates to mixed melanic in the other three groups. The high pheomelanin content of the breast feathers in the castrated birds was significantly reduced in the hens, intact drakes, and testosterone-injected castrates with a concomitant tendency for elevated eumelanin contents. At this site, a change occurred from pheomelanic to mixed melanic. 5α-dihydrotestosterone was clearly less effective than testosterone in affecting the melanin contents in castrates and resulted in an intermediate coloration. The differing effects of the two androgens might be a result of differences in their conversion to estrogens.  相似文献

Microanalysis of eumelanin is based on the formation of pyrrole-2,3,5-tricarboxylic acid (PTCA) on permanganate oxidation followed by its HPLC determination. A problem in this method was that the oxidation gave concave, exponential curves when the amounts of PTCA formed were plotted against the amounts of sample oxidized. The problem has been mostly overcome by adding a homogenate of 5 mg of a mouse liver to the oxidation medium. Sepia melanin, C57BL black mouse hair, B16 mouse melanoma, and MM418 human melanoma cells were oxidized in the absence or presence of the liver homogenate. The yields of PTCA increased about 1.5-fold by adding the liver homogenate and the calibration curves became linear or almost linear. With the improved method the PTCA values from various types of samples can be reliably compared.  相似文献
