Formation of Ascorbate Oxidase in Cultured Pumpkin Cells

Ascorbate oxidase activity rapidly increased during callus formationfrom pumpkin fruit tissue. The activity reached a maximum at5 days after transfer and then declined. In callus which hadbeen subcultured at about 4-week intervals for more than oneyear, the activity also increased after transfer to fresh mediumand reached a maximum in the early logarithmic phase of growth.Light had little effect on the appearance of ascorbate oxidaseactivity in pumpkin callus. In the callus grown in the presenceof 10µM CuSO₄, the activity was about 10 times that inthe presence of 0.1 µM CuSO₄, suggesting that the formatonof ascorbate oxidase in pumpkin callus is stimulated by copper,a prosthetic metal of the enzyme. From 45 to 75% of the totalascorbate oxidase activity in pumpkin cell suspension cultureswas found in the medium. Ascorbate oxidase activity in the medium,as well as that in the cells, increased soon after transferto fresh medium, and reached a maximum at about 5 days. pTiAch 5 TR区与在植物中积累agropine相关的三个基因片段已导入pINIIA或pACYC184,在大肠杆菌微细胞中表达出杂合蛋白。三个基因片段的pINIIA重组质粒的表达得到稳定蛋白、不稳定蛋白和在两个相表达出相似蛋白的三种结果。基因2'HindⅢ片段上的UGAA序列是在两个相表达相似蛋白的原因。从产生的杂合蛋白分子量和读码方式看,pTiAch 5中基因2'和基因1'的结构与pTi 15955近似,但由基因0'片段产生的杂合蛋白分子量比根据pTi15955 DNA序列数据推测的稍大。从表达强度和微细胞操作看,这个系统还不适应从大肠杆菌中方便地纯化杂合蛋白的需要。     

鳗鲡幼鱼耳石日轮的研究

本文报道采自辽东半岛沿岸鳗鲡(Anguilla japonica)的白仔鳗和经人工培育的当年幼鳗耳石日轮生长的观察结果。白仔鳗和幼鳗耳石平均直径均与体全长成直线相关。12尾白仔鳗耳石的平均日轮数146.3,据此推测其产卵期为11—12月。观察证实从咸淡水转人到淡水生活的幼鳗耳石的环纹有过渡带存在。     

Glycosphingolipid expression in spontaneously aborted fetuses and placenta from blood group p women. Evidence for placenta being the primary target for anti-Tja-antibodies

A 12-week-old fetus and one 17-week-old fetus + placenta were obtained after spontaneous abortions from two women of blood group p. The 17-week-old fetus was dissected into intestine, liver, brain and residual tissue. Nonacid glycosphingolipid fractions were prepared from the tissues. Glycolipid characterization was carried out using thin layer chromatography immunostained with monoclonal antibodies and bacteria and by¹H NMR spectroscopy and mass spectrometry. In the placental fraction substantial amounts of globotetraosylceramide (P-antigen) and globotriaosylceramide (P^k-antigen) were identified. In contrast, the fetuses contained only trace amounts of these structures, as revealed by immunostaining. These results indicate that the primary target for the antibodies of the anti-Tj^a serum is the placenta tissue, resulting in termination of the pregnancy.     

Mutation analysis and prenatal diagnosis in a Lesch-Nyhan family showing non-random X-inactivation interfering with carrier detection tests

Summary A nonsense mutation at the CpG-site in the codon for Arg(169) in the gene for hypoxanthine phosphoribosyltransferase (hprt) was identified by genomic polymerase chain reaction (PCR) and DNA sequencing in cultured fibroblasts from two brothers with Lesch Nyhan's syndrome. The recurrence of mutation at this CpG-site in several unrelated Lesch-Nyhan families suggests that deamination of 5-methylcytosine is a possible mechanism for mutagenesis. The level of hprt-mRNA in the fibroblasts of the patients was similar to that in healthy controls, whereas hprt-enzyme activity was not detectable. The mutation in this family was also identified in five female relatives and prenatally in a male fetus. Unexpectedly, results from hair follicle analyses and fibroblast selection studies in 8-azaguanine and 6-thioguanine medium showed a non-carrier phenotype in three of the female heterozygotes, whereas X-inactivation mosaicism was demonstrated in one heterozygote. A possible explanation for the apparent non-random X-inactivation in this family is the co-existence of the hprt mutation with an undefined X-linked lethal mutation. This observation is of practical relevance for carrier detection in other Lesch-Nyhan families.     

去甲肾上腺素在大鼠缰核引起的心血管效应及其机制

在乌拉坦麻醉的大鼠,研究了去甲肾上腺素(NE)在缰核(Hb)引起的心血管效应及其机制。Hb内微量注射NE使平均动脉压和心率呈剂量依赖性增加。用α受体阻断剂酚妥拉明预处理Hb,可明显减弱NE在Hb引起的心血管效应,但β受体阻断剂心得安或生理盐水不引起任何影响。Hb内微量注射海人酸使平均动脉压和心率明显增加,但Hb内微量注射利多卡因则不能引起明显的心血管效应。上述结果表明,Hb内NE在调节心血管活动中起重要作用,而这种效应可能是通过激活α受体使Hb兴奋的结果。     

NO—样松驰因子在大鼠止血带休克中的作用

为探讨NO-样松驰因子(NO-LRF)在休克中的效应及其病理生理意义,本工作在大鼠止血带休克(ToS)模型上发现离体灌流的主动脉环对去甲肾上腺素的反应性降低,组织cGMP含量增加。用NO前体L-精氨酸(L-Arg)或NO合成阻断剂L-NNA,可溶性乌苷酸环化酶抑制剂亚甲蓝灌流,分别增强或减轻ToS动物主动脉的上述变化,而且这些药物的作用不受血管内皮是否存在的影响。实验结果提示非内皮细胞源的NO-LRF是引起ToS动物血管低反应性的因素之一。体内实验表明 L-Arg治疗缓解,而L-NNA恶化ToS病程,提示NO-LRF可能参与机体的适应保护机制。
