Excised wheat (Triticum aestivum L.) leaves, when subjected to drought stress, increased ethylene production as a result of an increased synthesis of 1-aminocyclopropane-1-carboxylic acid (ACC) and an increased activity of the ethyleneforming enzyme (EFE), which catalyzes the conversion of ACC to ethylene. The rise in EFE activity was maximal within 2 h after the stress period, while rehydration to relieve water stress reduced EFE activity within 3 h to levels similar to those in nonstressed tissue. Pretreatment of the leaves with benzyladenine or indole-3-acetic acid prior to water stress caused further increase in ethylene production and in endogenous ACC level. Conversely, pretreatment of wheat leaves with abscisic acid reduced ethylene production to levels produced by nonstressed leaves; this reduction in ethylene production was accompanied by a decrease in ACC content. However, none of these hormone pretreatments significantly affected the EFE level in stressed or nonstressed leaves. The germination of lettuce (Lactuca sativa L.) seeds was greatly reduced when the seeds were heated at 97°C for 30 h prior to imbibition. This dormancy was effectively released when ethylene (1–100 ppm) or benzyladenine (BA) (0.005–0.05 mM) was applied during the imbibition period. Ethylene was not required during the early part of imbibition, but was essential during the period immediately prior to radicle protrusion. Treatment with 1-aminocyclopropane-1-carboxylic acid (ACC) (0.1–10 mM) stimulated germination, but was not as effective as ethylene or cytokinin treatment. During the germination of nondormant lettuce seeds, ethylene production increased rapidly and reached a peak at 24 h, which coincided with the emergence of the radicle, and then declined; the level of ACC increased as ethylene production rate increased, but remained at a high level after radicle protrusion. In heat-pretreated dormant lettuce seeds, the increases in percent germination, ethylene production, and ACC levels were all delayed and lower than those of nondormant seeds, and these increases were accelerated by treatment with ethylene or cytokinin.  相似文献

Summary Single unit spikes and evoked field potentials were recorded in different parts and depths of the corpus cerebelli and auricle of immobilized rays before and after stimulating with light, electric fields, touch, tail bending and direct shock to mechanoreceptive nerves of the lateral line.Discrete areas of the cerebellum are responsive to these modalities and the areas show limited overlap; they are all distinct from the area reported by Plassmann to be responsive to angular acceleration. The visual and tactile-proprioceptive areas are large; the electric area is small. Most units are excited only by one modality.The tail is represented only in the posterior lobe; trigeminal innervation extends from the posterior onto the anterior lobe, suggesting some topographic projection.The dynamic characteristics of the responses were examined particulary in the visual units. To a flash, units discharge up to six bursts of spikes in 500 ms. This pattern is reduced at repetition rates > 1/s; above ca. 4/s units tend to fire irregularly. Various kinds of units are found in respect to the succession of responses to short trains of flashes. Cyclic guanosine monophosphate (cGMP) signalling plays a fundamental role in many cell types, including platelets. cGMP has been implicated in platelet formation, but mechanistic detail about its spatio-temporal regulation in megakaryocytes (MKs) is lacking. Optogenetics is a technique which allows spatio-temporal manipulation of molecular events in living cells or organisms. We took advantage of this method and expressed a photo-activated guanylyl cyclase, Blastocladiella emersonii Cyclase opsin (BeCyclop), after viral-mediated gene transfer in bone marrow (BM)-derived MKs to precisely light-modulate cGMP levels. BeCyclop-MKs showed a significantly increased cGMP concentration after illumination, which was strongly dependent on phosphodiesterase (PDE) 5 activity. This finding was corroborated by real-time imaging of cGMP signals which revealed that pharmacological PDE5 inhibition also potentiated nitric oxide-triggered cGMP generation in BM MKs. In summary, we established for the first-time optogenetics in primary MKs and show that PDE5 is the predominant PDE regulating cGMP levels in MKs. These findings also demonstrate that optogenetics allows for the precise manipulation of MK biology.  相似文献

Osteoarthritis (OA) is a common joint disease featured by the deterioration of articular cartilage and chondrocyte death. Emerging evidence has indicated that circular RNAs (circRNAs) play an essential role in OA progress. Here, we found that the expression of circHIPK3 was significantly decreased in human and mouse OA cartilage. Knocking down circHIPK3 increased apoptosis and intracellular ROS level in HC‐a chondrocytes. We performed proteomic studies and identified that circHIPK3 regulated chondrocyte apoptosis through the mitochondrial pathway. Results of JC‐1 staining and western blot further confirmed that mitochondrial outer membrane permeabilization was promoted in HC‐a chondrocytes transfected by circHIPK3 siRNA. In terms of mechanism, we showed that PON2 functioned as a potential target of circHIPK3 to regulate chondrocyte apoptosis. Moreover, we revealed that circHIPK3 interacted with miR‐30a‐3p to regulate PON2 expression in chondrocytes. Taken together, our findings suggested that circHIPK3 regulated chondrocyte apoptosis by mitochondrial pathway, and targeting the circHIPK3/miR‐30a‐3p/PON2 axis might be a potential strategy for OA treatment.

The current study revealed the important role of circHIPK3 in regulating chondrocyte apoptosis and maintaining extracellular matrix (ECM) homeostasis. Mechanistically, circHIIPK3 might serve as a sponge of miR‐30a‐3p to regulate PON2 expression. The downregulation of circHIIPK3 resulted in the increased expression of miR‐30a‐3p and decreased expression of PON2, thus leading to mitochondrial pathway apoptosis and ECM destruction.  相似文献

为了合理利用和保护天敌进行卵形短须螨、双斑长跗萤叶甲和假眼小绿叶蝉的综合防治,用灰色系统分析方法和生态位分析法对合肥地区白毫早茶园3种主要害虫与其捕食性天敌在数量、时间、空间等方面关系进行分析,利用害虫与天敌关系密切指数之和综合评判9种天敌与3种害虫关系密切的前四位天敌。2015年卵形短须螨的前四位天敌是鳞纹肖蛸(5.3079)、三突花蟹蛛(5.1716)、锥腹肖蛸(4.8367)和草间小黑蛛(4.7869);2016年前四位天敌依次是三突花蟹蛛(5.3975)、鳞纹肖蛸(4.9414)、茶色新圆蛛(4.8757)、锥腹肖蛸(4.6815)。对两年结果综合分析,卵形短须螨的前四位天敌依次是三突花蟹蛛(10.5691)、鳞纹肖蛸(10.2493)、茶色新圆蛛(9.6353)和锥腹肖蛸(9.5182)。2015年双斑长跗萤叶甲的前四位天敌依次是锥腹肖蛸(5.6926)、异色瓢虫(5.6976)、八斑球腹蛛(5.5101)和斜纹猫蛛(5.4552);2016年依次是茶色新圆蛛(5.2909)、锥腹肖蛸(5.2710)、鳞纹肖蛸(5.1063)和斜纹猫蛛(5.0703)。对两年结果综合评判,双...  相似文献

基于条件价值法(CVM)开展了三亚海域海洋保护区生态系统多样性和重要海洋物种多样性的维持服务价值评估。于2014年12月通过面对面调研的形式对三亚当地城镇居民进行问卷调查,建立了支付意愿(WTP)多元线性回归方程,再将此回归模型推广到海南全省城镇居民,从而评估了三亚海域海洋保护区生态系统多样性和重要海洋物种多样性的维持服务价值;同时也对CVM研究中的相关问题如问卷设计、偏差分析和CVM的可靠性与有效性等进行了验证和探讨。研究结果表明:(1)2014年三亚海域海洋保护区生态系统多样性及重要海洋物种多样性的维持服务总价值分别为3.734亿元和4.442亿元。(2)受教育程度和家庭人均年收入等社会经济信息变量对支付意愿具有显著的影响水平,符合经济学理论。(3) CVM作为陈述偏好性价值评估方法,评估结果受偏差的影响较大。为了尽可能减小偏差,需要从问卷的设计、调查员筛选培训、调查方式和被调查者等诸多方面严格把关。(4) CVM问卷支付数额的设计较好地界定了支付意愿的范围和分布形态,支付意愿具有较高的统计效率,CVM的可靠性得到较好的验证,显示了CVM评估法在估算生态系统服务非使用价值的有效性。鉴于居民的学历和收入在短时间内很难得到提升,建议相关海洋管理部门加强对三亚海域海洋保护区和海洋生物多样性的宣传教育,增强居民对三亚海洋保护区和海洋珍稀濒危生物的认知及保护意识。  相似文献

[目的]筛选稳定表达口蹄疫病毒衣壳蛋白的牛肾细胞(Madin-Darby bovinekidney,MDBK)株.[方法]采用聚合酶链式反应(Polymerase chain reaction,PCR)方法从重组质粒pMD-P1-2A和pMD-3C中分别扩增口蹄疫病毒衣壳前体蛋白P1-2A基因和蛋白酶3C基因,将两基因依次插入逆转录病毒载体pBABE-puro.重组逆转录病毒载体pBABE-puro/P1-2A-3C和pVSV-G质粒载体用脂质体介导共转染GP2-293包装细胞.产生的重组逆转录病毒感染MDBK细胞后使用嘌呤霉素筛选抗性细胞.利用克隆环套取法得到单克隆细胞.经间接免疫荧光和酶联免疫吸附测定(Enzyme-linkedimmunosorbent assay,ELISA)方法检测MDBK细胞中衣壳蛋白的表达,并在电镜下观察口蹄疫病毒空衣壳.[结果]成功筛选到稳定表达口蹄疫病毒衣壳蛋白的MDBK细胞株,衣壳前体蛋白P1-2A在蛋白酶3C裂解作用下正确组装成空衣壳.[结论]该研究为口蹄疫亚单位疫苗的研制提供了实验材料.  相似文献
