以文心兰浅绿条纹突变体为材料,分析叶片光合色素含量和组成、叶绿素合成前体物质含量以及叶绿素荧光参数的变化,观察突变体叶绿体超微结构的改变,以探寻其叶色变异的生理基础。结果表明:(1)突变体叶绿素a(Chl a)、叶绿素b(Chl b)、类胡萝卜素(Car)和总叶绿素(Chl)含量分别比叶色正常植株显著降低了37.1%、34.0%、30.8%和36.3%。(2)突变体叶绿素生物合成受阻于胆色素原(PBG)到尿卟啉原Ⅲ(UrogenⅢ)的反应步骤。(3)突变体叶绿体发育存在明显的缺陷,基粒数目及基粒片层的垛叠层数明显减少,嗜锇颗粒及囊泡较多。(4)突变体初始荧光(Fo)比正常植株高39%,最大荧光(Fm)、最大光化学效率(Fv/Fm)、PSⅡ有效光化学效率(Fv′/Fm′)和PSⅡ实际光化学效率(ΦPSⅡ)均显著低于正常植株,但光化学淬灭系数(qP)和非光化学淬灭系数(NPQ)与正常植株无显著差异。研究结果说明,文心兰叶绿素生物合成受阻和叶绿体结构发育不良,导致叶绿素的含量下降,致使突变体叶片呈现浅绿条纹,光能利用率降低。  相似文献

摘要:【目的】分析蓝细菌病毒A-4L在鱼腥藻(Anabaena sp.) PCC 7120藻苔中形成同心圆噬斑的原因,阐明A-4L的一个重要生物学特征,为分离、鉴定和筛选新的水华蓝藻病毒提供借鉴。【方法】用初始滴度为2.8×1010PFU/mL的A-4L悬液感染鱼腥藻,在不同时间点收集裂解液绘制一步生长曲线,获得A-4L对鱼腥藻的潜伏期和释放量。将适量A-4L悬液感染不同培养时间的藻苔,逐日观察和记录藻苔病变情况。培养藻苔并接种适量A-4L悬液,分别置于完全持续光照(Light:Dark=24 h:0 h,L:D=24 h:0 h)条件;完全周期光照(L:D=14 h:10h)条件;或前3 d周期光照转后3 d持续光照的条件下,比较不同光照条件对同心圆噬斑形成的影响。然后挑取单个噬斑进行扩大培养,纯化后,负染电镜观察A-4L的超微形态。【结果】A-4L的潜伏期为0.5-2h,释放量约为247 IU/cell (Infectious Units)。在周期光照条件下,藻苔接种A-4L 3-4 d后,出现同心圆噬斑,且同心圆数量与攻毒后的天数(n)有相关性,为“n-1”;同心圆间距约为3 mm。与周期光照条件相比,在持续光照条件下未形成同心圆噬斑。而在周期光照条件转持续光照条件下,由先周期光照时所形成的同心圆在转持续光照后逐渐消失,证实同心圆噬斑的形成依赖于周期光照。负染电镜观察显示A-4L具有一个近似球形的头部,直径约为50 nm以及长度约为10 nm的尾部,形态与短尾蓝细菌病毒相似。【结论】A-4L是一株能形成同心圆噬斑的蓝细菌病毒,并揭示其同心圆噬斑形成的关键条件是周期光照。  相似文献

Background

Non-proliferative diabetic retinopathy is the early stage of diabetic retinopathy. Automatic detection of non-proliferative diabetic retinopathy is significant for clinical diagnosis, early screening and course progression of patients.

Methods

This paper introduces the design and implementation of an automatic system for screening non-proliferative diabetic retinopathy based on color fundus images. Firstly, the fundus structures, including blood vessels, optic disc and macula, are extracted and located, respectively. In particular, a new optic disc localization method using parabolic fitting is proposed based on the physiological structure characteristics of optic disc and blood vessels. Then, early lesions, such as microaneurysms, hemorrhages and hard exudates, are detected based on their respective characteristics. An equivalent optical model simulating human eyes is designed based on the anatomical structure of retina. Main structures and early lesions are reconstructed in the 3D space for better visualization. Finally, the severity of each image is evaluated based on the international criteria of diabetic retinopathy.

Results

The system has been tested on public databases and images from hospitals. Experimental results demonstrate that the proposed system achieves high accuracy for main structures and early lesions detection. The results of severity classification for non-proliferative diabetic retinopathy are also accurate and suitable.

Conclusions

Our system can assist ophthalmologists for clinical diagnosis, automatic screening and course progression of patients.

This study was to investigate the properties of mechanomyography (MMG), or muscle sound, of the paretic muscle in the affected side of hemiplegic subjects after stroke during isometric voluntary contractions, in comparison with those from the muscle in the unaffected side of the hemiplegic subjects and from the healthy muscle of unimpaired subjects. MMG and electromyography (EMG) signals were recorded simultaneously from the biceps brachii muscles of the dominant arm of unimpaired subjects (n=5) and the unaffected and affected arms of subjects after stroke (n=8), when performing a fatiguing maximal voluntary contraction (MVC) associated with the decrease in elbow flexion torque, and then submaximal elbow flexions at 20%, 40%, 60% and 80% MVCs. The root mean squared (RMS) values, the mean power frequencies (MPF, in the power density spectrum, PDS) of the EMG and MMG, and the high frequency rate (HF-rate, the ratio of the power above 15Hz in the MMG PDS) were used for the analysis. The MMG RMS decreased more slowly during the MVC in the affected muscle compared to the healthy and unaffected muscles. A transient increase could be observed in the MMG MPFs from the unaffected and healthy muscles during the MVC, associated with the decrease in their simultaneous EMG MPFs due to the muscular fatigue. No significant variation could be seen in the EMG and MMG MPFs in the affected muscles during the MVC. The values in the MPF and HF-rate of MMG from the affected muscles were significantly lower than those from the healthy and unaffected muscles (P<0.05) at the high contraction level (80% MVC). Both the MMG and EMG RMS values in the healthy and unaffected groups were found to be significantly higher than the affected group (P<0.05) at 60% and 80% MVCs. These observations were related to an atrophy of the fast-twitch fibers and a reduction of the neural input in the affected muscles of the hemiplegic subjects. The results in this study suggested MMG could be used as a complementary to EMG for the analysis on muscular characteristics in subjects after stroke.  相似文献

In the past decades the rapid growth of molecular diagnostics (based on either traditional PCR or isothermal amplification technologies) meet the demand for fast and accurate testing. Although isothermal amplification technologies have the advantages of low cost requirements for instruments, the further improvement on sensitivity, speed and robustness is a prerequisite for the applications in rapid pathogen detection, especially at point-of-care diagnostics. Carbon‐based supercapacitors store charge through the adsorption of electrolyte ions onto the carbon surface. Therefore, it would be more attractive for the enhanced charge storage if the locations for storing charge can be extended from carbon surface to space. Here, a novel spatial charge storage mechanism based on counterion effect from Fe(CN)63? ions bridged by oxygen groups and confined into honeycomb‐carbon frameworks is presented, which can provide additionally spatial charge storage for electrical double‐layer capacitances in a negative potential region and pseudocapacitances from Fe(CN)63?/Fe(CN)64? in a positive potential region. More importantly, an ultrafast supercapacitor based on this novelty carbon can be charged/discharged within 0.7 s to deliver both high specific energy of 15 W h kg?1 and ultrahigh specific power of 79.1 kW kg?1 in 1 m Na2SO4 electrolyte, much higher than those of previously reported asymmetric supercapacitors in aqueous electrolytes, as well as excellent cycling stability. These features suggest a new generation of ultrafast asymmetric supercapacitors as novel high‐performance energy storage devices.  相似文献

The non‐structural protein 4B (NS4B) of the hepatitis C virus (HCV) is an endoplasmic reticulum (ER) membrane protein comprising two consecutive amphipathic α‐helical domains (AH1 and AH2). Its self‐oligomerization via the AH2 domain is required for the formation of the membranous web that is necessary for viral replication. Previously, we reported that the host‐encoded ER‐associated reticulon 3 (RTN3) protein is involved in the formation of the replication‐associated membranes of (+)RNA enteroviruses during viral replication. In this study, we demonstrated that the second transmembrane region of RTN3 competed for, and bound to, the AH2 domain of NS4B, thus abolishing NS4B self‐interaction and leading to the downregulation of viral replication. This interaction was mediated by two crucial residues, lysine 52 and tyrosine 63, of AH2, and was regulated by the AH1 domain. The silencing of RTN3 in Huh7 and AVA5 cells harbouring an HCV replicon enhanced the replication of HCV, which was counteracted by the overexpression of recombinant RTN3. The synthesis of viral RNA was also increased in siRNA‐transfected human primary hepatocytes infected with HCV derived from cell culture. Our results demonstrated that RTN3 acted as a restriction factor to limit the replication of HCV.  相似文献

The peripheral fibers that extend from auditory neurons to hair cells are sensitive to damage, and replacement of the fibers and their afferent synapse with hair cells would be of therapeutic interest. Here, we show that RGMa, a repulsive guidance molecule previously shown to play a role in the development of the chick visual system, is expressed in the developing, newborn, and mature mouse inner ear. The effect of RGMa on synaptogenesis between afferent neurons and hair cells, from which afferent connections had been removed, was assessed. Contact of neural processes with hair cells and elaboration of postsynaptic densities at sites of the ribbon synapse were increased by treatment with a blocking antibody to RGMa, and pruning of auditory fibers to achieve the mature branching pattern of afferent neurons was accelerated. Inhibition by RGMa could thus explain why auditory neurons have a low capacity to regenerate peripheral processes: postnatal spiral ganglion neurons retain the capacity to send out processes that respond to signals for synapse formation, but expression of RGMa postnatally appears to be detrimental to regeneration of afferent hair cell innervation and antagonizes synaptogenesis. Increased synaptogenesis after inhibition of RGMa suggests that manipulation of guidance or inhibitory factors may provide a route to increase formation of new synapses at deafferented hair cells. © 2013 Wiley Periodicals, Inc.
