钼、硼对大豆叶片膜脂过氧化及体内保护系统的影响

The study deals with the changes of membrane peroxidation and endogenous protective system with different supplementation of molybdenum (Mo) and/or boron (B) concentration in soybean (Glycine max L.) leaves at three developmental stages (5-trifoliate stage, initiation of flowering, and peak podsetting stage) in three pot-grown soybean varieties (“Zhechun No.3”, “Zhechun No.2”, “3811”). The control plants under low Mo and low B exhibited an increasing of membrane permeability (MP), malondialdehyde (MDA) and proline (Pro) contents, polyphenol oxidase (PPO) and ascorbate oxidase (AO) activities and a decrease of ascorbate (AsA) contents, superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (AP) and catalase (CAT) activities. Application of Mo or B alone raised the ability of anti-oxidant of soybean leaves. The results indicated that the anti-oxidant enzymes (including SOD, POD, CAT and AP) related closely to anti-oxidant compounds (including AsA and Pro). There was some difference between the effects of Mo and B on the anti-oxidant, and a synergetic effect was observed between Mo and B. Some genetic variation in the responses to Mo and B was found among the three soybean varieties which was related to the activities of the total anti-oxidant systems.     

miR-10a Regulates Proliferation of Human Cardiomyocyte Progenitor Cells by Targeting GATA6

microRNAs (miRNAs) play essential roles in cardiogenesis. The altered expression of miRNAs can result in cardiac malformations by inducing abnormalities in the behavior of cardiac cells. However, the role of miR-10a in the regulation of cardiomyocyte progenitor cells (CMPCs) remains undetermined. In the present study, we found that up- or down-regulation of miR-10a inhibited or promoted the proliferation of human CMPCs, respectively, without affecting their differentiation toward cardiomyocytes. miR-10a bound to GATA6 directly and reduced GATA6 expression. Over-expression of GATA6 greatly attenuated the miR-10a-mediated inhibitory effect on the proliferation of human CMPCs. Thus, our results indicate that miR-10a could effectively modulate the proliferation of human CMPCs by targeting GATA6. The finding provides novel insights into the potency of miR-10a during heart development.     

Glutamate receptors and signal transduction in learning and memory

The plasticity of the central nervous system helps form the basis for the neurobiology of learning and memory. Long-term potentiation (LTP) is the main form of synaptic plasticity, reflecting the activity level of the synaptic information storage process, and provides a good model to study the underlying mechanisms of learning and memory. The glutamate receptor-mediated signal pathway plays a key role in the induction and maintenance of LTP, and hence the regulation of learning and memory. The progress in the understanding of the glutamate receptors and related signal transduction systems in learning and memory research are reviewed in this article.     

spindlin1基因的原核表达及多克隆抗体制备

目的:明确人新基因spindlin1的组织表达谱、相互作用蛋白。方法与结果:应用PCR技术获得spindlin1的开放读框,并将其插入原核表达载体pGEX-2T,转化大肠杆菌BL21(DE3),经IPTG诱导后,融合蛋白GST-Spindlin1获得了可溶性表达。用SDS-PAGE胶上的目的条带做抗原制备了多克隆抗体,进行了抗体纯化。结论:spindlin1在OVCar3细胞中可能以二聚体形式存在,这一现象还有待于进一步研究。

Top-down modulations from dorsal stream in lexical recognition: an effective connectivity FMRI study

Both the ventral and dorsal visual streams in the human brain are known to be involved in reading. However, the interaction of these two pathways and their responses to different cognitive demands remains unclear. In this study, activation of neural pathways during Chinese character reading was acquired by using a functional magnetic resonance imaging (fMRI) technique. Visual-spatial analysis (mediated by the dorsal pathway) was disassociated from lexical recognition (mediated by the ventral pathway) via a spatial-based lexical decision task and effective connectivity analysis. Connectivity results revealed that, during spatial processing, the left superior parietal lobule (SPL) positively modulated the left fusiform gyrus (FG), while during lexical processing, the left SPL received positive modulatory input from the left inferior frontal gyrus (IFG) and sent negative modulatory output to the left FG. These findings suggest that the dorsal stream is highly involved in lexical recognition and acts as a top-down modulator for lexical processing.     

Rapid determination of gatifloxacin in biological samples and pharmaceutical products using europium‐sensitized fluorescence spectrophotometry

A europium‐sensitized fluorescence spectrophotometry method using an anionic surfactant, sodium dodecyl benzene sulphonate (SDBS), was developed for the determination of gatifloxacin (GFLX). The GFLX–Eu³⁺–SDBS system was studied and it was found that SDBS significantly enhanced the fluorescence intensity of the GFLX–Eu³⁺ complex (about 25‐fold). The optimal experimental conditions were determined as follows: excitation and emission wavelengths of 338 and 617 nm, pH 7.5, 3.0 × 10^–6 mol/L europium(III), and 5.0 × 10^–5 mol/L SDBS. The enhanced fluorescence intensity of the system (ΔI_f) showed a good linear relationship with the concentration of GFLX over the range 1.0 × 10^–8–8.0 × 10^–7 mol/L with a correlation coefficient of 0.9990. The detection limit (S:N = 3) was determined as 1.0 × 10^–9 mol/L. This method has been successfully applied for the determination of GFLX in pharmaceuticals and human urine/serum samples. Compared with most other methods reported, the rapid and simple procedure proposed here offered higher sensitivity, wider linear range and good stability. A novel in vitro retinal differentiation model by co-culturing adult human bone marrow stem cells with retinal pigmented epithelium cells

Human retinal pigment epithelium (HRPE) cells are important in maintaining the normal physiology within the neurosensory retina and photoreceptors. Recently, transplantation of HRPE has become a possible therapeutic approach for retinal degeneration. By negative immunoselection (CD45 and glycophorin A), in this study, we have isolated and cultivated adult human bone marrow stem cells (BMSCs) with multilineage differentiation potential. After a 2- to 4-week culture under chondrogenic, osteogenic, adipogenic, and hepatogenic induction medium, these BMSCs were found to differentiate into cartilage, bone, adipocyte, and hepatocyte-like cells, respectively. We also showed that these BMSCs could differentiate into neural precursor cells (nestin-positive) and mature neurons (MAP-2 and Tuj1-positive) following treatment of neural selection and induction medium for 1 month. Furthermore, the plasticity of BMSCs was confirmed by initiating their differentiation into retinal cells and photoreceptor lineages by co-culturing with HRPE cells. The latter system provides an ex vivo expansion model of culturing photoreceptors for the treatment of retinal degeneration diseases.     

2011-2017年大连市输入性卵形疟原虫感染病例的实验室诊断

目的分析大连市2011-2017年7例输入性卵形疟原虫感染病例的病原、抗原及核酸检测结果,以提高卵形疟原虫诊断的准确率和效率。方法收集7例疑似疟疾患者的血样,对血样进行血涂片镜检、快速疟疾诊断试纸条检测(RDT)及荧光定量PCR检测。结果血涂片镜检结果显示7例患者均感染卵形疟原虫。RDT检测结果显示2例阴性,另外5例为泛疟原虫抗原阳性。荧光定量PCR结果显示7例患者均检出疟原虫属和卵形疟原虫核酸。结论综合镜检、RDT及荧光定量PCR检测结果确定该7例患者均为卵形疟原虫感染。
