An exploration of the role of Sertoli cells on fetal testis development using cell ablation strategy

Sertoli cells (SCs) are presumed to be the center of testis differentiation because they provide both structural support and biological regulation for spermatogenesis. Previous studies suggest that SCs control germ cell (GC) count and Leydig cell (LC) development in mouse testes. However, the regulatory role of SCs on peritubular myoid (PTM) cell fate in fetal testis has not been clearly reported. Here, we employed Amh‐Cre; diphtheria toxin fragment A (DTA) mouse model to selectively ablate SCs from embryonic day (E) 14.5. Results found that SC ablation in the fetal stage caused the disruption of testis cords and the massive loss of GCs. Furthermore, the number of α‐smooth muscle actin‐labeled PTM cells was gradually decreased from E14.5 and almost lost at E18.5 in SC ablation testis. Interestingly, some Ki67 and 3β‐HSD double‐positive fetal LCs could be observed in Amh‐Cre; DTA testes at E16.5 and E18.5. Consistent with this phenomenon, the messenger RNA levels of Hsd3b1, Cyp11a1, Lhr, Star and the protein levels of 3β‐HSD and P450Scc were significantly elevated by SC ablation. SC ablation appears to induce ectopic proliferation of fetal LCs although the total LC number appeared reduced. Together, these findings bring us a better understanding of SCs’ central role in fetal testis development. 相似文献

Abstract Mining activities have introduced various heavy metals and metalloids to surrounding soil environments, causing adverse impacts to the ecological environment system. The extremely high concentration of various heavy metals and metalloids make the Xikuangshan (Hunan, China) an excellent model to assess their ecological risk. In this study, the soil samples from 26 locations of different land use methods in four areas (smelting area, road nearby ore, mining area, and ore tailing area) in Xikuangshan with different levels of various heavy metals and metalloids (Sb, As, Pb, and Cd) were analyzed; in addition, the index of geo-accumulation and the potential ecological risk index were used to evaluate ecological risk. The results showed that the average contents of Sb, As, Pb, and Cd in all soil samples were 4368.222?mg·kg?1, 40.722?mg·kg?1, 248.013?mg·kg?1, and 40.941?mg·kg?1, respectively, implying serious contamination of compound pollution of heavy metals in soil. The concentration of heavy metals in soil among smelting area, road nearby ore, mining area, and ore tailing area showed significant distribution characteristics of region because different mining activities such as smelting, mining, transportation, and stacking caused different pollution intensity. Moreover, the contents of Sb in soil samples decreased successively in residue field, wasteland, forestland, sediment, grassland, and vegetable field, and the contents of Sb in vegetable-field and ecological restoration grassland were relatively low, which indicate that the method of grassland ecological restoration is an effective method to control antimony pollution in soil. The results of ecological risk assessment showed that the antimony mining area was seriously polluted by Sb, As, Pb, and Cd, and had strong ecological risk, and Sb and Cd were the most important pollution factors, which indicated that the pollution of Sb and Cd should be a major concern of relevant departments of environment and health. 相似文献

Abstract Identifying the source effect on heavy metals to human health risk is essential for devising and implementing restoration policies for polluted soils. For this purpose, eight heavy metals (As, Cd, Hg, Cr, Cu, Ni, Pb, and Zn) in soil profile samples (0–10, 10–20, 20–30, and 30–40?cm) collected in the area around aluminum-plastic manufacturing facilities (APMF) were determined. An absolute principal component score multiple linear regression (APCS-MLR) model supported by a health risk assessment (HRA) model was developed to determine the source apportionment of soil heavy metals and contribution rate of pollution sources to human health risk. Results showed significant accumulations of eight metals in the topsoil (0–20?cm), parent material, transportation, APMF, and agricultural practices were the four major contributing sources for heavy metals in soils, with average contribution percentages of 21.69%, 24.99%, 29.60%, and 14.25%, respectively. Carcinogenic risk factors for adults (1.23E-04) and children (1.32E-04) were found to be above the acceptable level (1E-06 to 1E-04). The health risk quantification results indicated that parent material, APMF, transportation, agricultural practices, and unidentified factors accounted for 55.76%, 14.48%, 12.09%, 10.13%, and 7.54% of the carcinogenic risk for children and adults. The adverse impacts of Cd, Zn, and Pb accumulations in soil coming from APMF activities were significant and need to receive more attention. 相似文献

AbstractGroundwater quality is defined by various water quality parameters. The aims of the research are to understand the relationships among different groundwater quality parameters and to trace the sources and affecting factors of groundwater pollution via statistical and multivariate statistical techniques. The 36 shallow groundwater samples collected from shallow pumping wells in Yan’an City were analyzed for various water quality parameters. Correlation analysis, principal component analysis (PCA), hierarchical cluster analysis (HCA), and multivariable linear regressions (MLR) were jointly used in this study to explore the sources and affecting factors of groundwater pollution. The study reveals that the mineral dissolution/precipitation and anthropogenic input are the main sources of the physicochemical indices and trace elements in the groundwater. Groundwater chemistry is predominantly regulated by natural processes such as dissolution of carbonates, silicates, and evaporates and soil leaching, followed by human activities as the second factor. Climatic factors and land use types are also important in affecting groundwater chemistry. Cl– is the greatest contributor to the overall groundwater quality revealed by the two regression models. The first model which has eight dependent variables is high in model reliability and stability, and is recommended for the overall groundwater quality prediction. The study is helpful for understanding groundwater quality variation in urban areas. 相似文献

Climate change is altering phenology; however, the magnitude of this change varies among taxa. Compared with phenological mismatch between plants and herbivores, synchronization due to climate has been less explored, despite its potential implications for trophic interactions. The earlier budburst induced by defoliation is a phenological strategy for plants against herbivores. Here, we tested whether warming can counteract defoliation‐induced mismatch by increasing herbivore‐plant phenological synchrony. We compared the larval phenology of spruce budworm and budburst in balsam fir, black spruce, and white spruce saplings subjected to defoliation in a controlled environment at temperatures of 12, 17, and 22°C. Budburst in defoliated saplings occurred 6–24 days earlier than in the controls, thus mismatching needle development from larval feeding. This mismatch decreased to only 3–7 days, however, when temperatures warmed by 5 and 10°C, leading to a resynchronization of the host with spruce budworm larvae. The increasing synchrony under warming counteracts the defoliation‐induced mismatch, disrupting trophic interactions and energy flow between forest ecosystem and insect populations. Our results suggest that the predicted warming may improve food quality and provide better growth conditions for larval development, thus promoting longer or more intense insect outbreaks in the future. 相似文献

Flowers have developed different strategies to attract pollinators through visual or olfactory signals. Most flowers offer pollinators a reward (e.g. nectar and pollen) for the pollination service. However, one‐third of Orchidaceae have been shown not to provide a reward. Calanthe are terrestrial orchids distributed throughout China, Nepal, Japan and tropical Asia. Despite its high diversity, the pollination biology of Calanthe remains largely unknown, even though it is an important aspect of plant conservation. In the study, through field surveying, there were three Hesperiidae butterflies pollinating two species of Calanthe and the pollination behavior differed between the two species of Calanthe, which might lead to different fruit setting rates. There was no nectar in the flowers of the two species, indicating deceptive pollination. Using a glass cylinder experiment, it was deduced that the two species of Calanthe were most likely to attract pollinators by generalized food deception. Interestingly, Hesperiidae butterflies were traditionally thought to be nectar thieves and generally do not transmit pollinia. However, our findings showed that, in this case, the thieves were deceived by the plants and pollinated them for free. 相似文献

The incidence of melanoma is rising globally including China. Comparing to Caucasians, the incidence of non‐cutaneous melanomas is significantly higher in Chinese. Herein, we performed genomic profiling of 89 Chinese surgically resected primary melanomas, including acral (n = 54), cutaneous (n = 22), and mucosal (n = 13), by hybrid capture‐based next‐generation sequencing. We show that mucosal melanomas tended to harbor more pathogenic mutations than other types of melanoma, though the biological significance of this finding remains uncertain. Chromosomal arm‐level alterations including 6q, 9p, and 10p/q loss were highly recurrent in all subtypes, but mucosal melanoma was significantly associated with increased genomic instability. Importantly, 7p gain significantly correlated with unfavorable clinical outcomes in non‐cutaneous melanomas, representing an intriguing prognostic biomarker of those subtypes. Furthermore, focal amplification of 4q12 (KIT, KDR, and PDGFRα) and RAD51 deletion were more abundant in mucosal melanoma, while NOTCH2 amplification was enriched in acral melanoma. Additionally, cutaneous melanomas had higher mutation load than acral melanomas, while mucosal melanomas did not differ from other subtypes in mutation burden. Together, our data revealed important features of acral and mucosal melanomas in Chinese including distinctive driver mutation pattern and increased genomic instability. Dendrolimus spp. are important destructive pests of conifer forests, and Dendrolimus punctatus Walker (Lepidoptera; Lasiocampidae) is the most widely distributed Dendrolimus species. During periodic outbreaks, this species is said to make “fire without smoke” because large areas of pine forest can be quickly and heavily damaged. Yet, little is known about the molecular mechanisms that underlie the unique ecological characteristics of this forest insect. Here, we combined Pacific Biosciences (PacBio) RSII single‐molecule long reads and high‐throughput chromosome conformation capture (Hi‐C) genomics‐linked reads to produce a high‐quality, chromosome‐level reference genome for D. punctatus. The final assembly was 614 Mb with contig and scaffold N50 values of 1.39 and 22.15 Mb, respectively, and 96.96% of the contigs anchored onto 30 chromosomes. Based on the prediction, this genome contained 17,593 protein‐coding genes and 56.16% repetitive sequences. Phylogenetic analyses indicated that D. punctatus diverged from the common ancestor of Hyphantria cunea, Spodoptera litura and Thaumetopoea pityocampa ~ 108.91 million years ago. Many gene families that were expanded in the D. punctatus genome were significantly enriched for the xenobiotic biodegradation system, especially the cytochrome P450 gene family. This high‐quality, chromosome‐level reference genome will be a valuable resource for understanding mechanisms of D. punctatus outbreak and host resistance adaption. Because this is the first Lasiocampidae insect genome to be sequenced, it also will serve as a reference for further comparative genomics. 相似文献

ABSTRACT 12-O-tetradecanoylphorbol-13-acetate (TPA), is a major active constituent of the seed oil of Croton tiglium L., has pharmacological activity for the treatment of acute myeloid leukemia patients. Diethyldithiocarbamate (DTC) is a potent inhibitor of NF-κB show activity of anticancer. In this study, we determined the effect of DTC and TPA in combination on HL-60 cells cultured in vitro and in vivo. In this study, we have shown that DTC and TPA synergistically inhibited the growth of HL-60 cells and strongly induced apoptosis in the cells. Mechanistic studies showed that the combined effects of DTC and TPA were associated with a decrease in Bcl-2. The animal experiment showed that the combination of DTC and TPA more potently inhibited the growth of HL-60 tumors than either agent alone. Our results indicate that the administration of TPA and DTC in combination may be an effective strategy for inhibiting the growth of acute myeloid leukemia cells. 相似文献
