Representatives of the genus Proterorhinus (tubenose gobies) from the water bodies of the Ponto-Caspian Basin were examined for sequence polymorphism of the mitochondrial DNA fragment containing the cytochrome b (Cyt-b) gene. A total of ten haplotypes were discovered, which formed two groups. Thus, the data obtained indicated the existence of only two taxonomically valid phylogenetic lineages, represented by (1) marine and brackish-water populations of the Black Sea and (2) freshwater populations of the whole Ponto-Caspian Basin, along with the brackish-water population of the Caspian Sea. Based on an analysis of the tubenose goby haplotype distribution patterns, the colonization patterns of this group of fishes (phylogeography) in the freshwater drainages of Ponto-Caspian Basin are examined. It was established that the basin of the ancient Caspian Sea was the major donor area for the formation of the freshwater and brackish-water populations. In the Ponto-Caspian Basin, two centers of the tubenose gobies speciation and distribution are distinguished. One of these centers is associated with the modern northwestern part of the Black Sea, and another one is confined to the Caspian Sea. It is suggested that the modern colonization of the Volga River by tubenose goby occurred from the Caspian Sea.  相似文献

The first data are presented on mtDNA diversity in Besermyans, the Finno-Ugric ethnic group related to Udmurts. An analysis of mtDNA polymorphism showed that Besermyans stood out from the other populations of Volga-Ural region due to the presence of a large proportion of the Mongoloid component. The sample of Besermyans contained East Eurasian haplotypes not detected in ethnic populations of the Volga region and Cisurals, while they were detected in South Siberia, mostly among Turkic-speaking populations. An analysis of the genetic distances between Besermyans and the neighboring ethnic groups showed that Besermyans were distant from other populations of Volga-Ural region and close to Turkic-speaking populations of South Siberia. Thus, the data obtained favor the suggestion on the mixed Udmurto-Turkic origin of Besermyans.  相似文献

Ramie (Boehmeria nivea L. Gaud) fibers extracted from the stem bast is one of the most important natural fibers. Ramie growth is severely hindered by drought stress but is promoted by gibberellins (GAs). In order to investigate ramie morphological and physiological responses to drought stress and GA3 treatment, four groups of potted ramie (severe drought stress (DS), severe drought stress and spraying with GA3 (DS + GA3), normal watering and spraying with GA3 (control + GA3), and normal watering as a control) were tested. The result showed that, comparing with the ramie growing under well watering condition, a decrease in chlorophyll a (Chl a) and carotenoid and an increase in proline and soluble sugar contents were commonly observed in drought-stressed and GA3-treated ramie. Different responses of the stem morphological traits, fiber yield, and seven physiological characteristics (relative water content, the activities of POD, SOD, and CAT enzymes, the contents of Chl b, endogenous GAs and MDA) were observed between drought-stressed and GA3-treated plants. When the ramie suffering drought stress was sprayed with GA3 (in (DS + GA3) group), the responses of some physiological traits (POD, SOD, CAT, MDA, and endogenous GAs) and morphological traits (stem shape and fiber yield) to drought stress disappeared completely or partially, and the plant presented similar characteristics of well-watered ramie in these traits. These results suggested that the application of exogenous GA3 can improve the drought tolerance of ramie.  相似文献

The effect of the ubiquitous downregulation of insulin receptor (InR) gene expression on the metabolism of juvenile hormone (JH) and dopamine (DA) in young females of D. melanogaster under normal conditions and heat stress is studied. The level of JH degradation and alkaline phosphatase activity (ALP, an enzyme regulating DA synthesis) were used as indicators of JH and DA levels, respectively. We demonstrated that, under normal conditions, the ubiquitous inhibition of the InR gene expression in D. melanogaster females induced an increase in the JH degradation and ALP activity. As we have already shown, this is indicative of the decrease in the concentration of the above hormones. It was also found that the total inactivation of InR does not affect the initiation of JH and DA metabolic system response to heat stress; however, it does affect its intensity. Thus, the involvement of the insulin signaling pathway in the regulation of the JH and DA metabolism in Drosophila females was demonstrated in vivo under normal and stress conditions.  相似文献

Life span depends on many factors, including the level of reactive oxygen species, like superoxide radical. Superoxide radical is produced from oxygen in the course of the oxidation of NADPH to NADP+. The process is catalyzed by NADPH oxidase. In this study, genotype and allele distributions of the C242T (rs4673) polymorphism in the CYBA gene, which encodes the α subunit of NADPH oxidase (p22phox), were examined in the sample of long livers and in the population sample of the city of Tomsk. Statistically significantly higher frequency of T allele among female long livers (34.625%), compared to the females from Russian population (26.32%) was demonstrated (χ2 = 5.226; p = 0.022; OR = 1.48). Thus, the T allele is associated with a high life expectancy in females from the Russian population. No such association was observed for males from the same population.  相似文献

Transgenic plants of camelina (Camelina sativa (L.) Crantz) with the synthetic gene of antimicrobial peptide cecropin P1 (cecP1) were obtained. Agrobacterium-mediated transformation is performed using the binary vector pGA482::cecP1 by vacuum infiltration of flower buds. The presence of the cecP1 gene in the genome of plants was confirmed by PCR. CecP1 gene expression in transgenic plants was shown by Western blot analysis and by antimicrobial activity of plant extracts against the bacterial phytopathogene Erwinia carotovora. The plants of F0 and F1 generations had the normal phenotype and retained the ability to form viable seeds in self-pollination. cecP1 plants exhibit enhanced resistance to bacterial and fungal phytopathogens: Erwinia carotovora and Fusarium sporotrichioides. The increased sustainability of cecropin P1-expressing plants against salt stress is shown. The possibility of the integration of the cecP1 gene into the overall protective system of plants against biotic and abiotic stresses is discussed.  相似文献

New data on mitochondrial gene pool polymorphism of Maeotis (1st–3rd centuries CE) in the light of their relation with Sarmatian nomads are presented. Maeotis are more genetically various, compared to Sarmatians; both the age of Maeotian gene pool and their close interactions with neighboring tribes can be reasons for this. The study of relationships of Maeotis and Sarmatians suggests an intensive gene interchange between them, which influences significantly on the formation of the Maeotian gene pool.

Light regulates ascorbic acid (AsA) synthesis, which increases in the light, presumably reflecting a need for antioxidants to detoxify reactive molecules produced during photosynthesis. Here, we examine this regulation in Arabidopsis thaliana and find that alterations in the protein levels of the AsA biosynthetic enzyme GDP-Man pyrophosphorylase (VTC1) are associated with changes in AsA contents in light and darkness. To find regulatory factors involved in AsA synthesis, we identified VTC1-interacting proteins by yeast two-hybrid screening of a cDNA library from etiolated seedlings. This screen identified the photomorphogenic factor COP9 signalosome subunit 5B (CSN5B), which interacted with the N terminus of VTC1 in yeast and plants. Gel filtration profiling showed that VTC1-CSN5B also associated with the COP9 signalosome complex, and this interaction promotes ubiquitination-dependent VTC1 degradation through the 26S proteasome pathway. Consistent with this, csn5b mutants showed very high AsA levels in both light and darkness. Also, a double mutant of csn5b with the partial loss-of-function mutant vtc1-1 contained AsA levels between those of vtc1-1 and csn5b, showing that CSN5B modulates AsA synthesis by affecting VTC1. In addition, the csn5b mutant showed higher tolerance to salt, indicating that CSN5B regulation of AsA synthesis affects the response to salt stress. Together, our data reveal a regulatory role of CSN5B in light-dark regulation of AsA synthesis.  相似文献
