N6-methyladeosine (m6A) plays an important role in virus infection and replication. Bombyx mori nuclear polyhedrosis is caused by Bombyx mori nucleopolyhedrovirus (BmNPV) infection. Expression levels of m6A-modification-related genes after the infection of BmNPV were detected at first. Then, expression levels of BmNPV nucleocapsid protein gene VP39 and envelope fusion protein gene GP64 after knockdown of METTL3in vitro were quantified to identify the effect of m6A modification on BmNPV. BmNPV firstly infects the larval midgut in case of oral infection. Subsequently, to clarify the relationship between m6A modification and resistance of the silkworm to BmNPV, we detected the expression levels of m6A-modification-related genes invivo before and after infection of BmNPV. The results indicated that low METTL3 level hindered the proliferation of BmNPV to some extent, and silkworm strain with low METTL3 level showed stronger resistance against BmNPV. This study will accumulate new experimental data for elucidating the resistance mechanism of silkworm against BmNPV. 相似文献

Microglial M1 depolarization mediated prolonged inflammation contributing to brain injury in ischemic stroke. Our previous study revealed that Genistein-3′-sodium sulfonate (GSS) exerted neuroprotective effects in ischemic stroke. This study aimed to explore whether GSS protected against brain injury in ischemic stroke by regulating microglial M1 depolarization and its underlying mechanisms. We established transient middle cerebral artery occlusion and reperfusion (tMCAO) model in rats and used lipopolysaccharide (LPS)-stimulated BV2 microglial cells as in vitro model. Our results showed that GSS treatment significantly reduced the brain infarcted volume and improved the neurological function in tMCAO rats. Meanwhile, GSS treatment also dramatically reduced microglia M1 depolarization and IL-1β level, reversed α7nAChR expression, and inhibited the activation of NF-κB signaling in the ischemic penumbra brain regions. These effects of GSS were further verified in LPS-induced M1 depolarization of BV2 cells. Furthermore, pretreatment of α7nAChR inhibitor (α-BTX) significantly restrained the neuroprotective effect of GSS treatment in tMCAO rats. α-BTX also blunted the regulating effects of GSS on neuroinflammation, M1 depolarization and NF-κB signaling activation. This study demonstrates that GSS protects against brain injury in ischemic stroke by reducing microglia M1 depolarization to suppress neuroinflammation in peri-infarcted brain regions through upregulating α7nAChR and thereby inhibition of NF-κB signaling. IntroductionProstate and breast cancer are the most prevalent primary malignant human tumors globally. Prostatectomy and breast conservative surgery remain the most common definitive treatment option for the >500,000 men and women newly diagnosed with localized prostate and breast cancer each year only in the US. Morphological examination is the mainstay of diagnosis but margin under-sampling of the excised cancer tissue may lead to local recurrence. In despite of the progress of non-invasive optical imaging, there is still a clinical need for targeted optical imaging probes that could rapidly and globally visualize cancerous tissues.MethodsElevated expression of junctional adhesion molecule-A (JAM-A) on tumor cells and its multiple pro-tumorigenic activity make the JAM-A a candidate for molecular imaging. Near-infrared imaging probe, which employed anti-JAM-A monoclonal antibody (mAb) phthalocyanine dye IR700 conjugates (JAM-A mAb/IR700), was synthesized and used to identify and visualize heterotopic human prostate and breast tumor mouse xenografts in vivo.ResultsThe intravenously injected JAM-A mAb/IR700 conjugates enabled the non-invasive detection of prostate and breast cancerous tissue by fluorescence imaging. A single dose of JAM-A mAb/IR700 reduced number of mitotic cancer cells in vivo, indicating theranostic ability of this imaging agent. The JAM-A mAb/IR700 conjugates allowed us to image a specific receptor expression in prostate and breast tumors without post-image processing.ConclusionThis agent demonstrates promise as a method to image the extent of prostate and breast cancer in vivo and could assist with real-time visualization of extracapsular extension of cancerous tissue. 相似文献

Loss of hand function and finger dexterity are main disabilities in the upper limb after stroke. An electromyography (EMG)-driven hand robot had been developed for post-stroke rehabilitation training. The effectiveness of the hand robot assisted whole upper limb training was investigated on persons with chronic stroke (n = 10) in this work. All subjects attended a 20-session training (3–5 times/week) by using the hand robot to practice object grasp/release and arm transportation tasks. Significant motor improvements were observed in the Fugl-Meyer hand/wrist and shoulder/elbow scores (p < 0.05), and also in the Action Research Arm Test and Wolf Motor Function Test (p < 0.05). Significant reduction in spasticity of the fingers as was measured by the Modified Ashworth Score (p < 0.05). The training improved the muscle co-ordination between the antagonist muscle pair (flexor digitorum (FD) and extensor digitorum (ED)), associated with a significant reduction in the ED EMG level (p < 0.05) and a significant decrease of ED and FD co-contraction during the training (p < 0.05); the excessive muscle activities in the biceps brachii were also reduced significantly after the training (p < 0.05). 相似文献

目的:探讨体外培养脐带血单个核细胞定向诱导分化为不同阶段红系祖细胞的动力学变化情况。方法:用0.5%甲基纤维素沉降脐带血红细胞及人淋巴细胞分离液密度梯度离心法得到单个核细胞,在含EPO、SCF、IGF-1等细胞因子的无血清培养体系中诱导其定向分化为红系祖细胞,观察细胞增殖、存活率、细胞集落形成情况,并检测不同阶段细胞红系特异性表面标志CD71和CD235a的表达。结果:随着培养时间的延长,细胞数逐渐增多,14 d细胞可扩增140倍左右,收集诱导后的细胞进行瑞氏吉姆萨染色,可见大量红系祖细胞,诱导后的细胞集落形成能力强,形成的克隆大部分为红系集落。诱导过程中,14 d前CD71、CD235a的表达逐渐增高。按细胞表面标志表达的不同可将诱导的细胞分为4群,分别对应红系祖细胞的不同阶段;随着诱导天数的增加,各时间点细胞对应的早期红系祖细胞群(P2、P3)比例逐渐下降,中晚期红系祖细胞群(P4、P5)的比例逐渐上升。结论:无血清培养基添加细胞因子组合的红系诱导培养体系可较好地诱导扩增红系祖细胞,流式分选可获得相对均一而处于不同分化阶段的红系祖细胞群体。获得了红系祖细胞体外分化的动力学数据,为今后进一步优化红系诱导分化体系获得均一的红系祖细胞奠定了基础,并对未来利用干细胞制备均一的红系祖细胞应用于临床治疗有一定的指导作用。 相似文献

Hybridization between incipient species is more likely to produce sterile or inviable F1 offspring in the heterogametic (XY or ZW) sex than in the homogametic (XX or ZZ) sex, a phenomenon known as Haldane's rule. Population dynamics associated with Haldane's rule may play an important role in early speciation of sexually reproducing organisms. The dynamics of the hybrid zone maintained by incomplete hybrid inferiority (sterility/inviability) in the heterogametic sex (a ‘weak’ Haldane's rule) caused by a Bateson–Dobzhansky–Muller incompatibility was modelled. The influences and interplays of the strengths of incompatibility, dispersal, density‐dependent regulation (DDR) and local adaptation of incompatible alleles in a scenario of short‐range dispersal (the stepping‐stone model) were examined. It was found that a partial heterogametic hybrid incompatibility could efficiently impede gene flow and maintain characteristic clinal noncoincidence and discordance of alleles. Density‐dependent regulation appears to be an important factor affecting hybrid zone dynamics: it can effectively skew the effects of the partial incompatibility and dispersal as measured by effective dispersal, clinal structures and density depression. Unexpectedly, local adaptation of incompatible alleles in the parental populations, which would be critical for the establishment of the incompatibility, exerts little effect on hybrid zone dynamics. These results strongly support the plausibility of the adaptive origin of hybrid incompatibility and ecological speciation: an adaptive mutation, if it confers a marginal fitness advantage in the local population and happens to cause epistatic inferiority in hybrids, could efficiently drive further genetic divergence that may result in the gene becoming an evolutionary hotspot. 相似文献

It has been pointed out that tea (Camellia sinensis (L.) O. Kuntze) prefers ammonium (NH 4 + ) over nitrate (NO 3 ? ) as an inorganic nitrogen (N) source. 15N studies were conducted using hydroponically grown tea plants to clarify the characteristics of uptake and assimilation of NH 4 + and NO 3 ? by tea roots. The total 15N was detected, and kinetic parameters were calculated after feeding 15NH 4 + or 15NO 3 ? to tea plants. The process of N assimilation was studied by monitoring the dynamic 15N abundance in the free amino acids of tea plant roots by GC-MS. Tea plants supplied with 15NH 4 + absorbed significantly more 15N than those supplied with 15NO 3 ? . The kinetics of 15NH 4 + and 15NO 3 ? influx into tea plants followed a classic biphasic pattern, demonstrating the action of a high affinity transport system (HATS) and a low affinity transport system (LATS). The V max value for NH 4 + uptake was 54.5 nmol/(g dry wt min), which was higher than that observed for NO 3 ? (39.3 nmol/(g dry wt min)). KM estimates were approximately 0.06 mM for NH 4 + and 0.16 mM for NO 3 ? , indicating a higher rate of NH 4 + absorption by tea plant roots. Tea plants fed with 15NH 4 + accumulated larger amounts of assimilated N, especially glutamine (Gln), compared with those fed with 15NO 3 ? . Gln, Glu, theanine (Thea), Ser, and Asp were the main free amino acids that were labeled with 15N under both conditions. The rate of N assimilation into Thea in the roots of NO 3 ? -supplied tea plants was quicker than in NH 4 + -supplied tea plants. NO 3 ? uptake by roots, rather than reduction or transport within the plant, seems to be the main factor limiting the growth of tea plants supplied with NO 3 ? as the sole N source. The NH 4 + absorbed by tea plants directly, as well as that produced by NO 3 ? reduction, was assimilated through the glutamine synthetase-glutamine oxoglutarate aminotransferase pathway in tea plant roots. Cloning and expression analysis of a CMS-related gene BcCoi1 from Brassica campestris ssp. chinensis
BcCoi1, a cytoplasmic male sterility related gene, which was isolated from flower buds of Brassica campestris ssp. chinensis Makino using the RACE technology, was characterized and submitted to the NCBI GenBank (accession no. GU263836). The gene encodes a 67.78-kD protein containing 16 leucine-rich repeats and an N-terminal F-box motif and is extremely similar to Arabidopsis thaliana Coi1 gene. The Southern blot showed that BcCoi1 belongs to a multigene family. In A. thaliana, the Coi1 gene is involved in jasmonate signaling, and Coi1 mutant displayed male sterility. In this study, qPCR results demonstrated that BcCoi1 was accumulated in stamens and was significantly higher expressed in flower organs of the maintainer line than in the CMS one. At the microsporocyte development stage, the gene was expressed at a significantly lower extent in the CMS line than in the maintainer line. This expression profile presumes that BcCoi1 plays a role in early microspore development in non-heading Chinese cabbage. 相似文献

The cDNA clone of ascorbate oxidase gene was isolated from non-heading Chinese cabbage (Brassica campestris ssp. chinensis Makino, cv. Suzhouqing) and characterized. Sequence analysis showed that there was a high similarity between this sequence (named BcAO) and its homologues in other plant species. Southern blotting indicated that more than one nuclear gene encoded this enzyme in non-heading Chinese cabbage. The mRNA level of the BcAO gene in leaves was monitored by real-time PCR at different developmental stages and under different stress conditions. Results showed that the expression of BcAO was upregulated by light, and the BcAO gene responded to copper stress as well. After inoculation with Alternaria brassicae, the expression of BcAO in the leaves was increased in general and peaked at 12 and 72 h post inoculation, with much higher expression at the later date. Cloning the BcAO gene will enable us to further understand its function and would provide useful information for resistance breeding program for non-heading Chinese cabbage. 相似文献
