Twelve polymorphic microsatellite markers were isolated from an AC-enriched genomic library of Buff-throated partridge (Tetraophasis szechenyii). The allele number of these loci ranged from three to 13 (average 7.75 per locus) in tested individuals. Polymorphism information content ranged from 0.532 to 0.882 with an average of 0.721. Average observed and expected heterozygosities were 0.559 (range from 0.240 to 0.792) and 0.772 (range from 0.621 to 0.910), respectively. These microsatellite markers will be useful for the assessment of genetic diversity, relatedness identification of family and analysis of population structure in wild buff-throated partridge. 相似文献

Currently, mortality compost is managed by temperature as extent of tissue degradation is difficult to assess. In the present study, field-scale mortality compost was constructed with composted brain tissue (Brain) and compost adjacent to brain tissue (CAB) sampled over 230 d. Following genomic DNA extraction, bovine-specific mitochondrial DNA (Mt-DNA) and bacterial 16S rDNA fragments were quantified using real-time PCR. Genomic DNA yield of Brain and CAB decreased rapidly (89-98%) and stabilized after 7 d. Compared to d 0, Brain Mt-DNA rapidly decreased (84-91% reduction on d 7). In CAB, Mt-DNA dramatically increased until d 28 (up to 34,500 times) thereafter decreasing by 77-93% on d 112. Quantification of bovine Mt-DNA indicates tissue degradation was initially characterized by rapid decomposition and release of cell contents into surrounding compost matrix followed by further degradation of Mt-DNA by flourishing microorganisms. It is suggested that gene duplication plays an important role in adaptation and evolution of plants. In this study, we examined whether the genus Rheum, with extensive diversification in the Qinghai-Tibeten Plateau (QTP) and adjacent regions, possessed the duplication of the chalcone synthase (CHS) genes and whether it underwent positive evolution. Here we cloned CHS-like genes from 16 Rheum species. Phylogenetic analyses suggested that CHS-like genes from Rheum comprised two monophyletic lineages (subfamilies), both of which were sisters to another related genus. The results showed that a genus-specific duplication occurred after this genus originated. The comparison of non-synonymous/synonymous substitution ratios between the two lineages in Rheum further indicated that a few sites along the duplicate branch underwent positive selection. The findings indicate that the duplication of Rheum species could contribute to their adaptive ability to rapidly changing environments which was resulted from the large-scale uplifts stages of QTP. 相似文献

Diabetic retinopathy (DR) is a major complication of diabetes and a leading cause of blindness in working-age Americans. DR is traditionally regarded as a disorder of blood–retina barriers, and the leakage of blood content is a major pathological characteristic of the disease. While the breakdown of the endothelial barrier in DR has been investigated extensively, the vascular leakage through the retinal pigment epithelium (RPE) barrier in the disease has not been widely acknowledged. As the blood content leaked through the RPE barrier causes excessive water influx to the retina, the breakdown of the RPE barrier is likely to play a causative role in the development of some forms of diabetic macular edema, a major cause of vision loss in DR. In this article, we will discuss the clinical evidences of the diabetes-induced RPE barrier breakdown, the alteration of the RPE in diabetes, the molecular and cellular mechanism of RPE barrier breakdown, and the research tools for the analysis of RPE barrier leakage. Finally, we will discuss the methodology and potential applications of our recently developed fluorescent microscopic imaging for the diabetes- or ischemia-induced RPE barrier breakdown in rodents. 相似文献
