Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7‐hydroxycoumarin‐derived carbonothioate‐based probe ( CC‐Hg ) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7‐hydroxyl, which reinforced the intramolecular charge transfer process. The CC‐Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0–2 μM and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.  相似文献

Background

Until now, radiographic manifestations of multidrug-resistant pulmonary tuberculosis (MDR- TB) in patients with diabetes mellitus (DM) have not been reported. We conducted a study to investigate the imaging features of pulmonary computed tomography (CT) for type 2 diabetic (T2DM) patients with MDR-TB.

Methods

The clinical data and pulmonary CT findings of 39 type 2 diabetic patients with MDR-TB, 46 type 2 diabetic patients with drug-susceptible tuberculosis (DS-TB), and 72 pure drug-susceptible TB cases (without T2DM and MDR) treated at Dalian Tuberculosis Hospital from 2012 to 2015 were collected, and the clinical features and imaging differences of the three groups were compared.

Results

The clinical characteristics of the three groups of patients were not significantly different except with respect to age and previous treatment history. However, on imaging, the patients with MDR-TB showed consolidation in and above the pulmonary segments was significantly more extensive than that seen in the DS-TB group with or without T2DM.

Conclusion

Consolidation in or above multiple pulmonary segments with multiple mouth-eaten cavities and bronchial damage on pulmonary CT images in type 2 diabetic patients with tuberculosis suggests the possibility of multi-drug resistance.  相似文献

In order to identify genes involved in oogenesis in shrimp, an ovarian cDNA library of Marsupenaeus japonicus was screened using a suppression-subtraction hybridization (SSH)-enriched probe. More than 20 genes were identified as differentially expressed genes between the ovary and the testis. Unexpectedly, one of these genes is a ribosomal protein that is normally considered a housekeeping gene. Northern blot shows that the shrimp ribosomal protein L24 gene (srpl24) is 0.6 kb in length. The expression level of srpl24 in the ovary is much higher than in the testis. Bioinformatics analyses show that srpl24 encodes a protein of 164 aa with a predicted molecular mass of 18.2 kDa, which is a cytoplasmic ribosomal protein. Real time PCR analyses demonstrated that the relative abundance of srpl24 mRNA in the different organs is: ovary > testis, hepatopancreas, muscle and eye. The highest expression level of srpl24 in the ovary suggests that srpl24 has an important role in oogenesis. It is the first reported rpl24 in crustaceans and is the first reported rpl24 that is differentially expressed between the ovary and the testis in animals.  相似文献

用RDP试剂提取三丁基锡暴露诱导的杂色鲍肝胰腺总RNA,经Oligotex纯化得到 mRNA;应用SMART技术合成双链cDNA,双链特异核酸酶(DSN)进行双链cDNA的均一化,构建了杂色鲍三丁基锡暴露诱导下的均一化cDNA文库.原始文库的库容为4.3× 106 CFU/ml,重组率为 97.9%.从文库中随机挑选了3 288个克隆进行测序,得到3 048个高质量EST序列,其中有 370条 Contigs,2 103条 Singlets,Unigenes共 2 473条,冗余率为18.86%.以上结果说明该文库质量较好,为进一步筛选相关功能基因打下基础;较低的冗余率说明该文库值得继续使用大规模ESTs测序的方法寻找相关功能基因,并为进一步使用基因芯片技术研究相关功能基因的表达谱提供便利.  相似文献

Metabolic reprogramming of non‐cancer cells residing in a tumor microenvironment, as a result of the adaptations to cancer‐derived metabolic and non‐metabolic factors, is an emerging aspect of cancer–host interaction. We show that in normal and cancer‐associated fibroblasts, breast cancer‐secreted extracellular vesicles suppress mTOR signaling upon amino acid stimulation to globally reduce mRNA translation. This is through delivery of cancer‐derived miR‐105 and miR‐204, which target RAGC, a component of Rag GTPases that regulate mTORC1 signaling. Following amino acid starvation and subsequent re‐feeding, 13C‐arginine labeling of de novo synthesized proteins shows selective translation of proteins that cluster to specific cellular functional pathways. The repertoire of these newly synthesized proteins is altered in fibroblasts treated with cancer‐derived extracellular vesicles, in addition to the overall suppressed protein synthesis. In human breast tumors, RAGC protein levels are inversely correlated with miR‐105 in the stroma. Our results suggest that through educating fibroblasts to reduce and re‐prioritize mRNA translation, cancer cells rewire the metabolic fluxes of amino acid pool and dynamically regulate stroma‐produced proteins during periodic nutrient fluctuations.  相似文献

Knowledge of leaf responses to elevated atmospheric [CO2] (CO2 concentration) is integral to understanding interactions between vegetation and global change. This work deals with responses of leaf mass‐based nitrogen concentration (Nm) and specific leaf area (SLA). It assesses the statistical significance of factors perceived as influential on the responses, and quantifies how the responses vary with the significant factors identified, based on 170 data cases of 62 species compiled from the literature. Resultant equations capture about 41% of the variance in the data for percent responses of Nm and SLA, or about 95% of the variance for Nm and SLA at 57–320% normal [CO2]; these performance statistics also hold for leaf area‐based N concentration and specific leaf weight. The equations generalize that: (i) both Nm and SLA decline as [CO2] increases; (ii) proportional decline of Nm is greater with deciduous woody species and with plants of normally low Nm, increases with pot size in growth chamber and greenhouse settings and with temperature and photosynthetic photon flux density (PPFD), and is mitigated by N fertilization; and (iii) proportional decline of SLA depends on pot size and PPFD similarly to Nm, increases with leaf life span and water vapour pressure deficit in enclosed experiments, and decreases with prolonged exposure to elevated [CO2] among broadleaf woody species in field conditions. The results highlight great uncertainty in the percent‐response data and reveal the potential feasibility to estimate Nm and SLA at various magnitudes of elevated [CO2] from a few key plant and environmental factors of broad data bases.  相似文献
