The embryogenic callus of "Red Marsh" grapefruit was stored in vitro by slow growth culture method for one year, and survived with a significant weight increment over that period. The survivers regenerated somatic embryos more easily than the controls. Eight callus lines were used for genetic analyses. Although chromosome number variations were verified by cytological examination both in the controls and the stored samples, the ploidy level remained relatively stable during the storage period. Randomly amplified polymorphic DNA (RAPD) analysis was performed to detect DNA sequence variation. No difference in RAPD pattern was found with the 102 primers used. However, a methylation sensitive amplified polymorphism (MSAP) assay showed DNA methylation changes in the stored samples compared with the controls. 相似文献

从水稻(Oryza sativa L.)、烟草(Nicotiana tabacum L.)和黑松(Pinus thunbergiiParl.)等植物的22对叶绿体SSR引物中筛选出 5对能用于柑橘叶绿体SSR分析的引物,应用这5对引物对9个组合的柑橘体细胞杂种的叶绿体遗传进行了分析.结果表明:这些组合再生的杂种中叶绿体都呈现随机分离,该现象与以前报道的RFLP分析结果一致,而且其可靠性已被CAPS分析所证实.表明柑橘叶绿体SSR同RFLP及CAPS一样可靠,并且更简单高效、易于操作,特别适合对柑橘等植物体细胞杂种进行早期胞质遗传组成分析. 相似文献

In the present paper, attempts were made to explore the possibility of employing ultraviolet (UV) irradiation in citrus asymmetric
fusion for transfer of limited amount of favorable traits from a desirable cultivar to a target one. Exposure of Satsuma mandarin
(Citrus unshiu Marc.) embryogenic protoplasts to UV at an intensity of 300 μW cm−2 led to reduced viability, especially under long irradiation duration. The protoplasts could not grow during culture when
they were irradiated for over 30 s. Terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling (TUNEL) assay revealed
extensive DNA fragmentation in the UV-irradiated protoplasts compared with those without UV treatment. Electrofusion between
UV-irradiated protoplasts of Satsuma mandarin (donor) with those of Jincheng (C. sinensis Osbeck, recipient), a local cultivar of superior quality, gave rise to regeneration of several lines of shoots, which failed
to root despite enormous endeavors. Ploidy analysis via flow cytometry and chromosome counting showed that four selected shoots
were either diploid, triploid or tetraploid. Random amplified polymorphism DNA (RAPD) and amplified fragment length polymorphism
(AFLP) confirmed the shoots, irrespective of their ploidy level, as putative somatic hybrids. Cleaved amplified polymorphism
sequences (CAPS) demonstrated that the shoots predominantly got their cytoplasmic components, in terms of chloroplast (cp)
and mitochondrion DNA, from Jincheng, along with possible recombination of cpDNA in some shoot lines. The current data indicated
that UV-based asymmetric fusion could also be employed in citrus somatic hybridization with the intention of creating novel
germplasms, which may provide an alternative approach for cultivar improvement. 相似文献

In the present paper attempts were made to induce chromosome doubling of ‘Meiwa’ kumquat (Fortunella crassifolia) protoplasts and ‘Frost’ navel orange (Citrus sinensis Osbeck) embryogenic callus via colchicine treatment. Colchicine decreased protoplast viability, delayed protoplast division and inhibited callus growth, indicating presence of toxicity to cells. Cell lines established from ‘Meiwa’ protoplasts treated with 0.01 and 0.1% colchicine for 8, 16 and 24 h at each concentration showed different responses when they were cultured on embryoid-induction medium. Flow cytometry (FCM) demonstrated that tetraploids were detected in cell lines and embryoids from all of the treatments, with the highest frequency being 19.23%. As for ‘Frost’, tetraploid cells were only detected when the callus was treated with 0.1% colchicine for either 4 or 8 days, from which plantlets were regenerated. FCM and chromosome counting confirmed them as true tetraploids. The diploid cells were more active in mitotic division during a 12-day culture and smaller in size than their tetraploid counterpart. Potential applications of the novel tetraploid germplasms obtained through in vitro chromosome doubling to citrus cultivar improvement are discussed. 相似文献

以纽荷尔脐橙为试材,分析了湖北省13个产地果实的常规品质和果皮色差值。结果显示,不同产地纽荷尔脐橙果实品质差异明显。系统聚类结果表明,秭归县、巴东县和兴山县的常规品质聚为第一类,长阳县、郧县、公安县、枝江市和宜都市聚为第二类,而丹江口市、阳新县、通城县、天门市和京山县聚为第三类;就果皮色差值来看,长阳县、郧县、兴山县和宜都市聚为第一类,巴东县、京山县、枝江市、丹江口市、阳新县、公安县、秭归县聚为第二类,天门市和通城县聚为第三类。综合分析常规品质和果皮色差值可以看出,三峡库区的兴山县、秭归县、巴东县纽荷尔脐橙综合品质最好,邻近三峡库区的长阳县、宜都市、枝江市,丹江库区的郧县和卷桥库区的公安县次之,丹江口市、阳新县、京山县,尤其是天门市和通城县的品质表现不如上述各个产地。因此湖北省纽荷尔脐橙应重点集中在三峡库区这一优势区域内优先发展,同时向其邻近的适宜地区辐射,而在其它产地宜选择小气候适宜地区适当发展。 相似文献
