白菜根肿病是由芸薹根肿菌(Plasmodiophora brassicae Woron)引起的一种常见土传病害,主要危害白菜的根部。根际是土壤-植物-微生物相互作用最活跃的关键微域,根际微生态系统中的微生物失衡是导致土传病害的重要因素,深入探究根际微生态与土传病害互作机制,有利于从根际微生物、抑病物质和功能代谢等方面挖掘防控土传病害安全高效的方法。本文综述了根际微生态与白菜根肿病的发生机制关系,从该病害的危害、发生的根际微生态机制及生防菌防治研究等方面综合分析了根际微生物调控白菜根肿病发生的机制,以期为白菜根肿病防控、促进土壤健康和维持根际微生态系统稳定提供理论依据。  相似文献

Ultra High Molecular Weight Polyethylene(UHMWPE)has been widely used as a bearing material for artificial joint replacementover forty years.It is usually crosslinked by gamma rays irradiation before its implantation into human body.In thisstudy,UHMWPE and UHMWPE/nano-hydroxyapatite(n-HA)composite were prepared by vacuum hot-pressing method.Theprepared materials were irradiated by gamma rays in vacuum and molten heat treated in vacuum just after irradiation.The effectof filling n-HA with gamma irradiation on tribological properties of UHMWPE was investigated by using friction and wearexperimental machine(model MM-200)under deionized water lubrication.Micro-morphology of worn surface was observedby metallographic microscope.Contact angle and hardness of the materials were also measured.The results show that contactangle and hardness are changed by filling n-HA and gamma irradiation.Friction coefficient and wear rate under deionized waterlubrication are reduced by filling n-HA.While friction coefficient is increased and wear rate is reduced significantly by gammairradiation.The worn surface of unfilled material is mainly characterized as adhesive wear and abrasive wear,and that of n-HAfilled material is mainly characterized as abrasive wear.After gamma irradiation,the degrees of adhesive and abrasive wear forunfilled material and abrasive wear of n-HA filled material are significantly reduced.Unfilled and filled materials after irradiationare mainly shown as slight fatigue wear.The results indicate that UHMWPE and UHMWPE/n-HA irradiated at the doseof 150 kGy can be used as bearing materials in artificial joints for its excellent wear resistance compared to original UHMWPE.  相似文献

应用形态学观察、流式细胞仪检测、Western印迹和DNA凝胶电泳等方法研究白藜芦醇对Jurkat细胞的作用。发现白藜芦醇处理组中细胞有皱缩、出泡、染色质边集等现象,但染色质浓缩呈散在团块状且不致密。细胞质结构疏松,线粒体肿胀,脊消失。少见凋亡小体。在白藜芦醇处理组,Western印迹可检测到弱的17kDacaspase-3条带,DNA凝胶电泳可以检测到梯状DNA和弥散条带;流式细胞仪在白藜芦醇处理组检测到大量PI单阳性细胞和少量膜联蛋白V单阳性细胞。Z-VAD-FMK干预后可以发现细胞死亡率降低,同时该组梯状DNA消失,但是大分子量弥散DNA条带依然可以检测到。结果表明白藜芦醇可以通过caspase依赖和非依赖途径导致Jurkat细胞死亡。此分子机制的明确将为白藜芦醇应用于临床白血病的治疗打下理论基础。  相似文献

In response to blood vessel injury, hemostasis is initiated by platelet activation, advanced by thrombin generation, and tempered by fibrinolysis. The primary fibrinolytic protease, plasmin, can be activated either on a fibrin-containing thrombus or on cells. Annexin A2 (A2) heterotetramer (A2·p11)(2) is a key profibrinolytic complex that assembles plasminogen and tissue plasminogen activator and promotes plasmin generation. We now report that, in endothelial cells, plasmin specifically induces activation of conventional PKC, which phosphorylates serine 11 and serine 25 of A2, triggering dissociation of the (A2·p11)(2) tetramer. The resulting free p11 undergoes ubiquitin-mediated proteasomal degradation, thus preventing further translocation of A2 to the cell surface. In vivo, pretreatment of A2(+/+) but not A2(-/-) mice with a conventional PKC inhibitor significantly reduced thrombosis in a carotid artery injury model. These results indicate that augmentation of fibrinolytic vascular surveillance by blockade of serine phosphorylation is A2-dependent. We also demonstrate that plasmin-induced phosphorylation of A2 requires both cleavage of A2 and activation of Toll-like receptor 4 on the cell surface. We propose that plasmin can limit its own generation by triggering a finely tuned "feedback" mechanism whereby A2 becomes serine-phosphorylated, dissociates from p11, and fails to translocate to the cell surface.  相似文献

Human coactosin-like protein (CLP) is a small (MW approximately 17 kDa) evolutionarily conserved actin-binding protein. It can bind to actin filaments but not globular actin and belongs to the fourth class of ADF-H-domain-containing proteins. Human CLP can also bind to 5LO, which plays an important role in cellular leukotriene synthesis. Although the structure of hCLP has been determined by both NMR and X-ray experiments, how hCLP binds to the actin filament is still a controversial question. To obtain insights into the structure of the complex, we studied the three-dimensional structure and backbone dynamics of hCLP using multidimensional NMR spectroscopy. Guided by the solution structure of the protein, a series of site-directed mutants were generated and their F-actin-binding activities were measured by high-speed cosedimentation assays. Furthermore, the structure model of the hCLP-F-actin complex was proposed using computational docking with the docking results filtered by the mutation data. Several previously untested residues (including T66, L89, R91, K102, D116 and E119) in hCLP were found important for the F-actin-binding activity. The extended region of beta4-beta5 of hCLP (residue 66-75) was found very flexible and very important for F-actin binding. The C-terminal residues of hCLP were not involved in F-actin binding, which was different from UNC-60B. Based on our hCLP-F-actin-binding model, different affinities of the four classes of ADF-H domain containing proteins for F-actin were explained.  相似文献

Six newly derived hybrid mouse embryonic stem (ES) cell lines and two inbred ES cell lines were tested for their ability to produce completely ES cell-derived mice by aggregation of ES cells with tetraploid embryos. Forty-five ES cell-tetraploid pups were generated from six hybrid ES cell lines and no pups from two inbred ES cell lines. These pups were found to have increased embryonic and placental weights than control mice. Twenty-two pups survived to adulthood and produced normal offsprings, and the other 23 pups died of several reasons including respiratory distress, abdomen ulcer-like symptoms, and foster failure. The 22 adult ES cell-tetraploid mice were completely ES cell-derived as judged by coat color and germline transmission, only two of them was found to have tetraploid component in liver, blood, and lung as analyzed by microsatellite loci. Our data suggested that genetic heterozygosity is a crucial factor for postnatal survival of ES cell-tetraploid mice, and tetraploid embryo aggregation using hybrid ES cells is a simple and efficient procedure for immediate generation of targeted mouse mutants from genetically modified ES cell clones, in contrast to the standard protocol, which involves the production of chimeras and several breeding steps.  相似文献

How do presynaptic inputs regulate synapse formation? In this issue of Neuron, Lin et al. show that the neurotransmitter acetylcholine decreases the stability of AChR clusters. This dispersing activity, which requires the serine/threonine kinase Cdk5, cooperates with positive signals from motoneurons to ensure high concentration of AChRs at the neuromuscular junction.  相似文献
