对甜菊(Steviarebaudiana)愈伤组织中尚未发生器官分化的分生细胞团进行了超微结构研究.结果表明,在器官分化条件下,愈伤组织中形成的分生区域的细胞体积小,细胞核大,核仁明显,且具核仁泡,部分细胞核中含有核内含物.大量小液泡分布在细胞的边周或散布于整个细胞中.液泡中通常含有陷入的细胞质成分和膜状物.部分液泡的形成与内质网膨大有密切关系.同时也观察到由内质网形成的多圈膜和双层膜包围细胞质成分的同心环结构.高尔基体及其小泡丰富,有时聚集分布在细胞某一区域.核糖体密集,有的聚集成多聚核糖体.因此,愈伤组织中分生区的细胞与分生组织中的液泡化和分裂的细胞类似.分生区细胞的另一明显特征是出现质膜内陷.推测这些超微结构特征可能反映了甜菊愈伤组织器官分化前的某些形态变化。  相似文献

We have set up a system to rapidly freeze muscle fibers during contraction to investigate by electron microscopy the ultrastructure of active muscles. Glycerinated fiber bundles of rabbit psoas muscles were frozen in conditions of rigor, relaxation, isometric contraction, and active shortening. Freezing was carried out by plunging the bundles into liquid ethane. The frozen bundles were then freeze-substituted, plastic-embedded, and sectioned for electron microscopic observation. X-ray diffraction patterns of the embedded bundles and optical diffraction patterns of the micrographs resemble the x-ray diffraction patterns of unfixed muscles, showing the ability of the method to preserve the muscle ultrastructure. In the optical diffraction patterns layer lines up to 1/5.9 nm-1 were observed. Using this method we have investigated the myofilament lengths and concluded that there are no major changes in length in either the actin or the myosin filaments under any of the conditions explored.  相似文献

本文在国内首次报道普通小麦(Triticumaestivum,2n=6x=42)和鸭茅状摩擦禾(Tripsacumdactyloides,2n=4x=72)杂交成功。用33个春小麦品种(系)为母本,与鸭茅状摩擦禾杂交,授粉6636朵花,获得3083个颖果。颖果分为有胚和无胚两类。有胚颖果806个,占总颖果数的26.14%。33个母本中30个都能和鸭茅状摩擦禾杂交,14个母本的杂交亲和性(胚数/授粉花数×100)超过10%。4个最高的分别达到59.00%,39.00%,29.50%和22.66%与小麦和玉米(Zeamays,2n=2x=20)的杂交相比,鸭茅状摩擦禾同样对小麦Kr基因的作用不敏感,且与小麦的杂交亲和性要比与玉米杂交时高。加之鸭茅状摩擦禾为野生型,具有多年生、多分枝、花期长等特点,可以克服小麦和玉米杂交时花期不遇的困难。讨论了用它作为花粉给体的优越性及在小麦改良和产生小麦单倍体上的前景。  相似文献

This study investigated the effects of prolonged exercise, with and without glucose supplementation, on metabolism and sarcoplasmic reticulum (SR) Ca(2+)-handling properties in working vastus lateralis muscle. Fifteen untrained volunteers [peak O(2) consumption (Vo(2peak)) = 3.45 +/- 0.17 l/min; mean +/- SE] cycled at approximately 60% Vo(2peak) on two occasions, during which they were provided with either an artificially sweetened placebo beverage (NG) or a 6% glucose (G) beverage (~1.00 g carbohydrate/kg body mass). Beverage supplementation started at 30 min of exercise and continued every 15 min thereafter. SR Ca(2+) handling, metabolic, and substrate responses were assessed in tissue extracted from the vastus lateralis at rest, after 30 min and 90 min of exercise, and at fatigue in both conditions. Plasma glucose during G was 15-23% higher (P < 0.05) than those observed during NG following 60 min of exercise until fatigue. Cycle time to fatigue was increased (P < 0.05) by approximately 19% during G (137 +/- 7 min) compared with NG (115 +/- 6 min). Prolonged exercise reduced (P < 0.05) maximal Ca(2+)-ATPase activity (-18.4%), SR Ca(2+) uptake (-27%), and both Phase 1 (-22.2%) and Phase 2 (-34.2%) Ca(2+)-release rates during NG. The exercise-induced reductions in SR Ca(2+)-cycling properties were not altered during G. The metabolic responses to exercise were all unaltered by glucose supplementation, since no differences in respiratory exchange ratios, carbohydrate and lipid oxidation rates, and muscle metabolite and glycogen contents were observed between NG and G. These results indicate that the maintenance of blood glucose homeostasis by glucose supplementation is without effect in modifying the muscle metabolic, endogenous glycogen, or SR Ca(2+)-handling responses.  相似文献

Background and aims

To test the hypothesis that dominant plant species could acquire different nitrogen (N) forms over a spatial scale and they also have the ability to compete for available N with microbes.

Methods

A short-term 15N labeling experiment was conducted in the temperate grassland ecosystem of North China in July of 2013. Three N forms (NO3 ? , NH4 + and glycine) labeled with 15N were injected into the two soil depths (0–5 and 5–15 cm) surrounding each plant to explore N acquisition by plants and microbes. Three dominant plant species (Artemisia frigida, Cleistogenes squarrosa and Artemisia capillaris) were investigated.

Results

Two hours after 15N labeling, all three dominant plant species absorbed both organic and inorganic N, but different patterns were observed at two soil depths. Uptake of NO3 ? was significantly higher at 0–5 cm than at 5–15 cm soil depth among all the dominant plant species. 15N recovery by microbes was significantly higher than plants. However, 15N recovery by plants showed different patterns over soil depths.

Conclusions

Dominant plant species in the temperate grassland have different patterns in acquisition of N added to soil in organic form and absorption of inorganic N, and microbes were more effectively than plants at competing for N in a short-term period.
