Methamphetamine and other drugs activate a small proportion of all neurons in the brain. We previously developed a fluorescence‐activated cell sorting (FACS)‐based method to characterize molecular alterations induced selectively in activated neurons that express the neural activity marker Fos. However, this method requires pooling samples from many rats. We now describe a modified FACS‐based method to characterize molecular alterations in Fos‐expressing dorsal striatal neurons from a single rat using a multiplex pre‐amplification strategy. Fos and NeuN (a neuronal marker) immunohistochemistry indicate that 5–6% of dorsal striatum neurons were activated 90 min after acute methamphetamine injections (5 mg/kg, i.p.) while less than 0.5% of neurons were activated by saline injections. We used FACS to separate NeuN‐labeled neurons into Fos‐positive and Fos‐negative neurons and assessed mRNA expression using RT‐qPCR from as little as five Fos‐positive neurons. Methamphetamine induced 3–20‐fold increases of immediate early genes arc, homer‐2, c‐fos, fosB, and its isoforms (ΔfosB and a novel isoform ΔfosB‐2) in Fos‐positive but not Fos‐negative neurons. Immediate early gene mRNA induction was 10‐fold lower or absent when assessed in unsorted samples from single dorsal striatum homogenates. Our modified method makes it feasible to study unique molecular alterations in neurons activated by drugs or drug‐associated cues in complex addiction models.

The genetic code specifies 20 common amino acids and is largely preserved in both single and multicellular organisms. Unnatural amino acids (Uaas) have been genetically incorporated into proteins by using engineered orthogonal tRNA/aminoacyl-tRNA synthetase (RS) pairs, enabling new research capabilities and precision inaccessible with common amino acids. We show here that Escherichia coli tyrosyl and leucyl amber suppressor tRNA/RS pairs can be evolved to incorporate different Uaas in response to the amber stop codon UAG into various proteins in Caenorhabditis elegans. To accurately report Uaa incorporation in worms, we found that it is crucial to integrate the UAG-containing reporter gene into the genome rather than to express it on an extrachromosomal array from which variable expression can lead to reporter activation independent of the amber-suppressing tRNA/RS. Synthesizing a Uaa in a dipeptide drives Uaa uptake and bioavailability. Uaa incorporation has dosage, temporal, tRNA copy, and temperature dependencies similar to those of endogenous amber suppression. Uaa incorporation efficiency was improved by impairing the nonsense-mediated mRNA decay pathway through knockdown of smg-1. We have generated stable transgenic worms capable of genetically encoding Uaas, enabling Uaa exploitation to address complex biological problems within a metazoan. We anticipate our strategies will be generally extendable to other multicellular organisms.  相似文献

Background

One of the most common types of brain-computer interfaces (BCIs) is called a P300 BCI, since it relies on the P300 and other event-related potentials (ERPs). In the canonical P300 BCI approach, items on a monitor flash briefly to elicit the necessary ERPs. Very recent work has shown that this approach may yield lower performance than alternate paradigms in which the items do not flash but instead change in other ways, such as moving, changing colour or changing to characters overlaid with faces.

Methodology/Principal Findings

The present study sought to extend this research direction by parametrically comparing different ways to change items in a P300 BCI. Healthy subjects used a P300 BCI across six different conditions. Three conditions were similar to our prior work, providing the first direct comparison of characters flashing, moving, and changing to faces. Three new conditions also explored facial motion and emotional expression. The six conditions were compared across objective measures such as classification accuracy and bit rate as well as subjective measures such as perceived difficulty. In line with recent studies, our results indicated that the character flash condition resulted in the lowest accuracy and bit rate. All four face conditions (mean accuracy >91%) yielded significantly better performance than the flash condition (mean accuracy = 75%).

Conclusions/Significance

Objective results reaffirmed that the face paradigm is superior to the canonical flash approach that has dominated P300 BCIs for over 20 years. The subjective reports indicated that the conditions that yielded better performance were not considered especially burdensome. Therefore, although further work is needed to identify which face paradigm is best, it is clear that the canonical flash approach should be replaced with a face paradigm when aiming at increasing bit rate. However, the face paradigm has to be further explored with practical applications particularly with locked-in patients.  相似文献

Analyses of mitochondrial DNA (mtDNA) and nonrecombining Y chromosome (NRY) variation in the same populations are sometimes concordant but sometimes discordant. Perhaps the most dramatic example known of the latter concerns Polynesians, in which about 94% of Polynesian mtDNAs are of East Asian origin, while about 66% of Polynesian Y chromosomes are of Melanesian origin. Here we analyze on a genome-wide scale, to our knowledge for the first time, the origins of the autosomal gene pool of Polynesians by screening 377 autosomal short tandem repeat (STR) loci in 47 Pacific Islanders and compare the results with those obtained from 44 Chinese and 24 individuals from Papua New Guinea. Our data indicate that on average about 79% of the Polynesian autosomal gene pool is of East Asian origin and 21% is of Melanesian origin. The genetic data thus suggest a dual origin of Polynesians with a high East Asian but also considerable Melanesian component, reflecting sex-biased admixture in Polynesian history in agreement with the Slow Boat model. More generally, these results also demonstrate that conclusions based solely on uniparental markers, which are frequently used in population history studies, may not accurately reflect the history of the autosomal gene pool of a population.  相似文献

The functional analysis of the sodium exchanger SOS1 from wheat, TaSOS1, was undertaken using Saccharomyces cerevisiae as a heterologous expression system. The TaSOS1 protein, with significant sequence homology to SOS1 sodium exchangers from Arabidopsis and rice, is abundant in roots and leaves, and is induced by salt treatment. TaSOS1 suppressed the salt sensitivity of a yeast strain lacking the major Na+ efflux systems by decreasing the cellular Na+ content while increasing K+ content. Na+/H+ exchange activity of purified plasma membrane from yeast cells expressing TaSOS1 was higher than controls transformed with empty vector. These results demonstrate that TaSOS1 contributes to plasma membrane Na+/H+ exchange.  相似文献

The influence of zinc (Zn) on physiological and biochemical parameters was studied to elucidate the mechanism of Zn resistance in Phragmites australis. Zn concentrations in roots, stems and leaves increased with exogenous Zn concentration, while Zn content in roots was much higher than in shoots. X-ray microanalysis was used to reveal compartments in which Zn accumulated in root cortex. Zinc concentrations followed a gradient with the sequence: intercellular space>cell wall >vacuole >cytoplasm, indicating that most Zn was immobilized in the apoplast or sequestered into the vacuolar lumen. Sequential extraction of various Zn chelates revealed that the ratio of Zn extracted with different extraction media was markedly different. Ethanol, HAc (acetic acid) and NaCl-extractable Zn were dominant in both roots and leaves of P. australis. Zn-binding protein fractions were found in the roots and leaves after gel filtration chromatography, among which a polypeptide with an apparent molecular mass of 14kDa bound Zn most effectively. Two newly synthesized polypeptides of 58 and 45kDa appeared under Zn pollution, whereas a prominent fraction of 72kDa disappeared. The involvement of Zn distribution in plant tissues, subcellular compartments and chelates and Zn-inducing proteins in the acclimation mechanism of P. australis to Zn pollution is discussed.  相似文献

The patterns of resource translocation among interconnected ramets within a clone may show diversity. In order to understand the pattern of inter-ramet water translocation in natural clones of the rhizomatous shrub Hedysarum laeve (Leguminosae), which frequently dominates inland dunes in semi-arid areas of China, acid fuchsin dye application combined with spectrometric analysis was employed. Dye solutions were fed to the base of plants (near the parent ramet) and to the apical portions (near one of the youngest shoots) and the amount of dye in shoots and rhizomes was measured spectrometrically after 24 h. The total amount of dye translocated from the base in the acropetal direction was approximately four times as high as that of basipetal translocation from cut sections near the apex. Most of the dye was detected in the shoots with very little found in the rhizomes. The average percentage allocation of dye from donor ramet transferred to recipient ramets in the ramet hierarchy closer, due to acropetal and/or basipetal translocation, was significantly greater than those in the ramet hierarchy less close to it. The ramet-hierarchy order had a great influence on dye translocation in either the acropetal or basipetal direction. It is inferred that the donor ramet would transport water towards its recipient ramets in different ramet hierarchies in unequal proportions in this species.  相似文献

采用生物信息学方法对人CDC73 (cell division cycle 73)基因编码蛋白的理化性质、亲疏水性、跨膜区域、信号肽区域、二级结构、三级结构、蛋白质之间的相互作用、亚细胞定位进行预测分析。使用多种分析软件对人CDC73基因编码蛋白进行预测分析。研究可知,人CDC73基因属于抑癌基因,该基因编码一个由531个氨基酸组成的肿瘤抑制因子Parafibromin,其等电点为9.63,半衰期为30 h且在哺乳动物中高度保守;二级结构预测发现13个α螺旋和10个β折叠片层,三级结构预测结果的可靠性达69.01%,亚细胞定位主要分布于细胞质及细胞核。由本研究可知,人CDC73基因编码蛋白是一个存在核定位序列的不稳定亲水蛋白,在细胞内广泛分布并参与多种生命活动过程,且能够抑制肿瘤的产生。对人CDC73基因编码蛋白结构和功能的预测分析,可为其进一步的研究提供一定的理论依据,也为相关疾病的诊治提供新的思路。  相似文献
