The impact of uncoupling protein (UCP) 1, UCP3 and UCP3s expressed in yeast on oxidative phosphorylation, membrane potential and H+ transport is determined. Intracellular ATP synthesis is inhibited by UCP3, much more than by UCP1, while similar levels of UCP3 and UCP1 exist in the mitochondrial fractions. Measurements of membrane potential and H+ efflux in isolated mitochondria show that, different from UCP1, with UCP3 and UCP3s there is a priori a preponderant uncoupling not inhibited by GDP. The results are interpreted to show that UCP3 and UCP3s in yeast mitochondria are in a deranged state causing uncontrolled uncoupling, which does not represent their physiological function.  相似文献

帕金森病(Parkinson‘s disease,PD)是中枢神经系统多巴胺(Dopamine,DA) 能神经元退行性疾病,其发病率具有明显的性别差异。性腺类固醇激素,尤其是雌激素,可在垂体、下丘脑、中脑边缘系统和黑质-纹状体(nigrostriatal,NS)系统等水平影响DA能神经递质系统的功能。而且,雌激素的使用剂量和时间的不同,可影响DA能神经元的活性。本文就雌激素与DA的关系及PD发病具有性别差异方面的新近进展作简要概述。  相似文献

In this communication, the effects of trehalose and culture system on protocorm-like body (PLB) growth were investigated. PLB derived from Phalaenopsis and Doritaenopsis cultivars, which were grown on solidified trehalose amended NP medium showed higher proliferation rate than on NP medium containing sucrose. For P. ‘Hwa Feng Red Jewel’ and Dtp. ‘Mount Beauty×Su’s Red Lip’, the proliferation rates on solidified trehalose media were almost two times higher than those on sucrose media after 8-week culture. However, Knudson C (KC) medium did not reveal the similar results between trehalose and sucrose. In liquid culture system, both trehalose amended NP and KC media brought about better results than sucrose containing media for PLB proliferation. For culture system test, solidified media showed higher proliferation rate than liquid media under the same medium composition. Roller bottles were more suitable than flask-shaking cultures in liquid systems for PLB proliferation.  相似文献

胚胎干细胞起源于植入前胚胎的内细胞团(inner cell mass,ICM),能够在体外进行增殖并能分化成三个胚层的所有细胞类型.如果能有效地将胚胎干细胞(embryonic stem cell,ES细胞)诱导分化为特定的细胞类型,ES细胞将成为细胞替代治疗中供体细胞的一个理想来源,从而可以应用于帕金森病(Parkinson's disease,PD)、糖尿病、心脓死等的退行性病变疾病的治疗.近年来,将ES细胞来源的多巴胺能神经元用于PD治疗的研究越来越广泛.现对这些研究中有关基因表达调控、实验研究进展、临床应用以及所遇到的问题作一综述.虽然这些研究还没能最终大面积应用于临床.但为PD的治疗提供了一个广阔的应用前景.  相似文献

Aminolevulinic acid (ALA) was produced by recombinant Escherichia coli BL21(DE3) (pET28-A.R-hemA) harboring the ALA synthase gene (hemA) from Agrobacterium radiobacter zju-0121. The effects of inducers on the ALA synthase activity and ALA productivity were evaluated. The results indicated that a low isopropyl-beta-D-thiogalactoside (IPTG) concentration (0.05 mmol/L) was favorable for high expression of ALA synthase, which resulted in higher ALA productivity. For metabolic engineering applications, lactose was a better substitute of IPTG for active enzyme expression. When lactose concentration was 5 mmol/L, the specific ALA synthase activity and ALA productivity reached 16.7 nmol/(min . mg of protein) and 1.15 g/L, respectively, which were about 15% and 43% higher than those induced by IPTG.  相似文献

The process of mechanotransduction of bone, the conversion of a mechanical stimulus into a biochemical response, is known to occur in osteoblasts in response to fluid shear stress. In order to understand the reaction of osteoblasts to various times of flow perfusion, osteoblasts were seeded on three-dimensional scaffolds, and cultured in the following conditions: continuous flow perfusion, intermittent flow perfusion, and static condition. We collected samples on day 4, 8 and 12 for analysis. Osteoblast proliferation was demonstrated by cell proliferation and scanning electron microscopy assay. Additionally, the expression of known markers of differentiation, including alkaline phosphatase and osteocalcin, were tested by qRT-PCR and alkaline phosphatase activity assay, and the deposition of calcium was used as an indicator of mineralization demonstrated by calcium content assay. The results supported that low fluid shear stress plays an important role in the activation of osteoblasts: enhance cell proliferation, increase calcium deposition, and promote the expression of osteoblastic markers. Furthermore, the continuous flow perfusion is a more favorable environment for the initiation of osteoblast activity compared with intermittent flow perfusion. Therefore, the force and time of fluid shear stress are important parameters for osteoblast activation.  相似文献

There is an apparent allometric relationship between peak frequency of echolocation and body size in rhinophilids. However, some rhinolophids deviate from this rule. To date this variation has been explained as a result of partitioning of communication channels. An alternative hypothesis that food resource partitioning results in this divergence in expected frequencies was tested by comparing prey selection between Rhinolophus macrotis Blyth, 1844 and Rhinolophus lepidus Blyth, 1844 in Yunnan Province, China. These two sympatric species are morphologically similar but acoustically divergent: R. macrotis has an echolocation frequency significantly lower than that predicted by the allometric relationship, whereas that of R. lepidus agreed with expectations. Prey selection experiments, conducted in a flight tent, indicated that the dominant prey taxa of R. macrotis were Lasiocampidae, Arctiidae and Noctuidae, whilst that of R. lepidus were Arctiidae, Noctuidae and Ichneumonidae. R. macrotis ate more earless moths and fewer eared moths than R. lepidus, and R. macrotis fed on larger prey in general and captured a wider size range than that captured by R. lepidus. These results confirmed the existence of finely tuned trophic niche differentiation and suggested that food resource partitioning is one of the factors leading to lower peak frequency of calls in R. macrotis.  相似文献

目的探讨MMP-9和PCNA在宫颈癌组织中的联合表达、相关性及其临床意义。方法采用组织芯片和免疫组化技术检测143例宫颈浸润癌(ICC)、20例癌旁正常宫颈上皮(NCE)中MMP-9和PCNA的表达,统计分析MMP-9、PCNA表达与病理分级、临床分期和淋巴结转移等临床病理因素的关系。结果 MMP-9、PCNA在宫颈癌组织中的阳性率分别为89.5%(128/143)和93.7%(134/143),较正常宫颈组织中的阳性率40.0%(8/20)和15.0%(3/20)显著增高(P=0.000)。MMP-9的表达与病理分级(r=0.342,P=0.000)、淋巴结转移(r=0.197,P=0.018)和间质浸润深度(r=0.203,P=0.015)呈正相关。PCNA表达与临床分期(r=0.228,P=0.006)、病理分级(r=0.330,P=0.000)呈正相关。MMP-9和PCNA在宫颈癌组织中的表达强度呈正相关(r=0.228,P=0.006),二者表达一致的比例高达87.41%(125/143)。结论MMP-9和PCNA在宫颈癌中的异常表达与肿瘤的分化、侵袭和发展密切相关,联合检测二者的表达对于进一步理解宫颈癌的生物学行为和判断预后有一定价值。  相似文献
