A two-dimensional flow model has been developed to simulate mass transport in a microchannel bioreactor with a porous wall. A two-domain approach, based on the finite volume method, was implemented. For the fluid part, the governing equation used was the Navier-Stokes equation; for the porous medium region, the generalized Darcy-Brinkman-Forchheimer extended model was used. For the porous-fluid interface, a stress jump condition was enforced with a continuity of normal stress, and the mass interfacial conditions were continuities of mass and mass flux. Two parameters were defined to characterize the mass transports in the fluid and porous regions. The porous Damkohler number is the ratio of consumption to diffusion of the substrates in the porous medium. The fluid Damkohler number is the ratio of the substrate consumption in the porous medium to the substrate convection in the fluid region. The concentration results were found to be well correlated by the use of a reaction-convection distance parameter, which incorporated the effects of axial distance, substrate consumption, and convection. The reactor efficiency reduced with reaction-convection distance parameter because of reduced reaction (or flux), and smaller local effectiveness factor due to the lower concentration in Michaelis-Menten type reactions. The reactor was more effective, and hence, more efficient with the smaller porous Damkohler number. The generalized results could find applications for the design of bioreactors with a porous wall.  相似文献

Baicalein and genistein were studied for the affinities for human serum albumin (HSA) in the presence and absence of three CdTe quantum dots (QDs) with different sizes. Three typical CdTe QDs with maximum emissions of 535 nm (green-emitting, G-QDs), 598 nm (yellow-emitting, Y-QDs), and 654 nm (red-emitting, R-QDs) were tested. The fluorescence intensities of HSA decreased remarkably with increasing concentration of QDs. Baicalein resulted in an obvious blue-shift of the λem of HSA from 340 to 334 nm. However, the extents of blue-shifts induced by baicalein and genistein in the presence of QDs were much bigger than that in the absence of QDs. The quenching process of baicalein for HSA was easily affected by the QDs size than that of genistein. QDs increased the quenching constant from 136.97% to 162.24% for baicalein. However, QDs only increased the quenching constants from 20.56% to 32.23% for genistein. G-QDs, Y-QDs, and R-QDs increased the affinities of baicalein for HSA about 3.02%, 6.38% and 9.40%. G-QDs, Y-QDs, and R-QDs increased the affinities of genistein for HSA about 2.56%, 13.46% and 19.44%. The binding affinities of baicalein and genistein for HSA increased with increasing QDs size.  相似文献

Protein tyrosine kinases play key roles in many molecular and cellular processes in diverse living organisms. Their proper functioning is crucial for the normal growth, development, and health in humans, whereas their dysfunction can cause serious diseases, including various cancers. As such, intense studies have been performed to understand the molecular mechanisms by which the activities of protein tyrosine kinases are regulated in mammalian cells. Particularly, small molecules that can modulate the activity of tyrosine kinases are of great importance for discovering therapeutic drug candidates for numerous diseases. Notably, heme cannot only serve as a prosthetic group for hemoglobins and enzymes, but it also is a small signaling molecule that can control the activity of diverse signaling and regulatory proteins. Using a computational search, we found that a group of non-membrane spanning tyrosine kinases contains one or more CP motifs that can potentially bind to heme and mediate heme regulation. We then used experimental approaches to determine whether heme can affect the activity of any of these tyrosine kinases. We found that heme indeed affects the phosphorylation of key tyrosine residues in Jak2 and Src, and is therefore able to modulate Jak2 and Src activity. Further experiments showed that Jak2 and Src bind to heme and that the presence of heme alters the sensitivity of Jak2 and Src to trypsin digestion. These results suggest that heme actively interacts with Jak2 and Src and alters their conformation.  相似文献

目的:研究TGF-α、TGF-β1与胰腺癌临床病理的关系.方法:免疫组织化学SABC法检测41例胰腺癌组织和12例正常胰腺组织中TGF-α、TGF-β1的表达情况,并分析其与病人的年龄、性别、肿瘤部位、病理分级和分期(UICC)等指标的相关性.结果:在41例胰腺癌组织中TGF-αTGF-β1的阳性表达率分别为73.2%、63.4%,在12例正常胰腺组织中的阳性表达率分别为16.7%、25.0%,两组之间存在显著差异(P=0.001).在胰腺癌组织中,TGF-β1的表达在不同分期中存在显著差异(P=0.019);TGF-α的表达与胰腺癌患者的年龄、性别、肿瘤部位、大小、分期及组织学分级无相关性(P>0.05).结论:TGF-α、TGF-β1在胰腺癌中高表达.TGF-β1与胰腺癌病理分期有关.  相似文献

Based on the theory of constitution of Traditional Chinese Medicine (TCM), the human population is divided into nine constitutions including one balanced constitution (Normality) and eight unbalanced constitutions (Yang-deficiency, Yin-deficiency, Phlegm-wetness, Qi-deficiency, Wetness-heat, Blood stasis, Depressed constitution, and Inherited special constitution). Different constitutions have specific metabolic features and different susceptibility to certain diseases. However, whether a genetic basis accounts for such constitution classification is yet to be determined. Here we performed a genetic study to assess the association between genetic variations of metabolic genes including PPARD, PPARG and APM1 and the constitutions. A total of 233 individuals of the Han population in China were classified into four groups, Normality, Yang-deficiency, Yin-deficiency and Phlegm-wetness with whom 23 single nucleotide polymorphisms (SNPs) in the three genes were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Biased distribution of PPARD rs2267669 and rs2076167, APM1 rs7627128 and rs1063539 in Yang-deficiency, PPARG Pro12Ala in Yin-deficiency and PPARD rs2076167, APM1 rs266729 and rs7627128 in Phlegm-wetness were observed. The frequencies of Haplotype13 (Hap13) of PPARG in Yin-deficiency, Hap25 of APM1 in Yang-deficiency and Hap2 of PPARD and Hap14 of PPARG in Phlegm-wetness, were significantly different from those in Normality, suggesting those might be group-associated haplotypes. These results suggested that single SNP and haplotypes of PPARD, PPARG and APM1 may underlie the genetic basis of the constitutions classified in TCM.  相似文献

Omega-3(ω-3) fatty acid desaturase transgenic pigs may improve carcass fatty acid composition. The use of transgenic pigs is also an excellent large animal model for studying the role of ω-3 fatty acids in the prevention and treatment of coronary heart disease and cancer. Transgenic pigs carrying synthesized fatty acid desaturase-1 gene (sFat-1) from Caenorhabditis briggsae by somatic cell nuclear transfer (SCNT) were produced for the first time in China. Porcine fetal fibroblast cells were transfected with a sFat-1 expression cassette by the liposome-mediated method. Transgenic embryos were reconstructed by nuclear transfer of positive cells into enucleated in vitro matured oocytes. A total of 1889 reconstructed embryos were transferred into 10 naturally cycling gilts. Nine early pregnancies were established, 7 of which went to term. Twenty-one piglets were born. The cloning efficiency was 1.1% (born piglets/transferred embryos). The integration of the sFat-1 gene was confirmed in 15 live cloned piglets by PCR and Southern blot except for 2 piglets. Expression of the sFat-1 gene in 12 of 13 piglets was detected with RT-PCR. The data demonstrates that an efficient system for sFat-1 transgenic cloned pigs was developed, which led to the successful production of piglets expressing the sFat-1 gene.  相似文献

Enzymatic transesterification of waste cooking oil, comprising fats, oil and grease (FOG), to produce fatty acid methyl esters (FAME) i.e. biodiesel, was investigated using a novel strain of the fungus Aspergillus niger, immobilized as a whole‐cell biocatalyst. Response surface methodology (RSM), with a five‐level‐three‐factor central composite rotatable design, was used to optimize the reaction and analyze the relationship of reaction variables and their coinfluent on the response i.e. FAME yield. Independent variables that affect the transesterification reaction include temperature, feedstock water content and enzyme amount. Using RSM, a second‐order polynomial equation was derived for FAME yield using multiple regression analysis. The second‐order polynomial regression model was highly significant (P<0.001) in predicting the actual relationship between the response and the variables, where a linear relationship was apparent between observed and predicted values (R2=0.9651). In addition, the predicted determination coefficient q2 i.e. 0.7723, also proved that the model has a high predictive ability. The validation experiments, under optimized conditions, showed that the predicted value of maximum FAME yield (i.e. 91.3%) was in close agreement with the experimental value (i.e. 91.8%).  相似文献

The Plio-Pleistocene history of C4 plant biomass in northwestern China has been documented from the loess-soil sequences of the Loess Plateau region. However, how C4 plants evolved in the warmer and low-elevation eastern China monsoon zone is still poorly known mainly because of the unavailability of well-dated geological records. In this study, a 203.6-m core of floodplain deposits was recovered from the North China Plain near Tianjin and dated magnetostratigraphically. The results define a chronosequence for the last 3.3 Ma. The late Quaternary portion of the core consists of fluvio-marine sediments while the rest of the section (3.3-0.6 Ma) contains abundant paleosols formed on the floodplain, as confirmed by soil micromorphological evidence. The stable carbon and oxygen isotopic composition of pedogenic carbonates was measured to document vegetation and climate changes. The results reveal mixed C3 and C4 vegetation with an estimated C4 abundance of ~ 40-60% from ~ 3.1 to ~ 2.2 Ma, and a subsequent gradual decline to ~ 25% until ~ 0.6 Ma. This trend is consistent with the data from the loess-soil sequences further west on the Loess Plateau, suggesting they are regionally significant changes. The lowering of growing-season temperature and/or drier conditions induced by global cooling would explain this overall decline.  相似文献
