Generalized additive models (GAMs) have been widely used for flexible modeling of various types of outcomes. When the outcome in a GAM is subject to missing, practical analyses often assume that missingness is missing at random (MAR). This assumption can be of suspicion when the missingness is not by design. Evaluating the potential effects of alternative nonignorable missing data mechanism on the MAR inference from a GAM can be important but often challenging due to the complicatedness of alternative nonignorable models. We apply the index approach to local sensitivity (Troxel, Ma, and Heitjan 2004 (2004). Statistica Sinica 14 , 1221–1237) to evaluate the potential changes of the GAM estimates in the neighborhood of the MAR model. The approach avoids fitting any complicated nonignorable GAM. Only MAR estimates are required to calculate the resulting sensitivity index and adjust the GAM estimates to account for nonignorable missingness. Thus the proposed approach is considerably simpler to conduct, as compared with the alternative methods. The simulation study shows that the index provides valid assessment of the local sensitivity of the GAM estimates to nonignorable missingness. Background
Clostridium botulinum is a taxonomic designation for at least four diverse species that are defined by the expression of one (monovalent) or two (bivalent) of seven different C. botulinum neurotoxins (BoNTs, A-G). The four species have been classified as C. botulinum Groups I-IV. During brain development, each neuron must find and synapse with the correct pre- and postsynaptic partners. The complexity
of these connections and the relatively large distances some neurons must send their axons to find the correct partners makes
studying brain development one of the most challenging, and yet fascinating disciplines in biology. Furthermore, once the
initial connections have been made, the neurons constantly remodel their dendritic and axonal arbours in response to changing
demands. Neurexin and neuroligin are two cell adhesion molecules identified as important regulators of this process. The importance
of these genes in the development and modulation of synaptic connectivity is emphasised by the observation that mutations
in these genes in humans have been associated with cognitive disorders such as Autism spectrum disorders, Tourette syndrome
and Schizophrenia. The present review will discuss recent advances in our understanding of the role of these genes in synaptic

The rhizomatous sedge Carex brevicuspis can produce clumping ramets from shortened rhizomes (phalanx) and spreading ramets from elongated rhizomes (guerrilla) to form a combined clonal growth form. In this paper, changes in clonal growth and biomass allocation pattern of C. brevicuspis in response to sedimentation were studied. Four sedimentation depths (0, 3, 6, and 9 cm) were applied to 48 ramets in a randomized block design. Plants were harvested after 20 weeks. With increasing sedimentation depth, the proportion of spreading ramets to total ramets increased from 19.6% in 0 cm to 92.9% in 9 cm sedimentation treatments, whereas that of clumping ramets decreased from 80.4% to 7.1%, indicating a change of clonal growth form from phalanx to guerrilla as a response to sedimentation. With increasing sedimentation depth, biomass allocation to shoots and roots did not change, but rhizome mass ratio increased from 2.7% in 0 cm to 7.2% in 9 cm sedimentation treatments, suggesting that production of long rhizomes changes biomass allocation pattern. The results show that plasticity of clonal growth forms, by which more spreading ramets are produced, is an effective strategy to avoid sedimentation stress under our experimental conditions. 相似文献

【目的】为进一步研究镰刀菌Q7-31T产生的植物细胞壁降解酶的酶系信息。【方法】以1%(W/V)蛋白胨为氮源,0.5%(W/V)燕麦秸秆为碳源,20°C、120 r/min振荡培养3 d,诱导发酵培养菌株,获得的粗酶液经过Sephacry S-100凝胶柱层析和DEAE琼脂糖弱阴离子交换柱层析,最终得到纯化的内切葡聚糖酶,并对其进行酶学性质分析及串联质谱鉴定。【结果】研究表明:Egn21的分子质量为44.25 kDa,等电点为4.91;酶学特性研究显示:Egn21降解羧甲基纤维素的最适反应温度为40°C,在45°C以下比较稳定。该酶最适pH为6.0,在pH为5.0–8.0条件之间比较稳定。Co~(2+)、Zn~(2+)和Mg~(2+)对其没有明显作用,而Fe~(2+)、Ca~(2+)、K~(+)、Na~(+)和Mn~(2+)对酶活性有抑制作用,Hg~(2+)会使酶失去活性。【结论】从Q7-31T中分离纯化得到的内切葡聚糖酶Egn21,经过酶学特性与串联质谱鉴定结果显示其属于GH5家族。 相似文献

[目的]环二鸟苷酸c-di-GMP是细菌中广泛存在的第二信使,能够调控多种细胞功能。c-di-GMP的合成与水解分别由含有GGDEF结构域和EAL结构域的蛋白催化。本研究针对茎瘤固氮根瘤菌ORS571的GGDEF和EAL结构域相关蛋白进行基因组学分析,并对三个同时含有GGDEF和EAL结构域的蛋白(AZC_3085、AZC_3226和AZC_4658)进行功能研究。[方法]利用SMART数据库对含有GGDEF和EAL结构域的蛋白进行结构域预测。利用CLUSTALW程序对蛋白序列进行比较分析。通过同源重组的方法构建突变株,并对突变株的细胞运动能力、胞外多糖合成、生物膜形成及与豆科宿主的结瘤等表型进行测定。[结果]茎瘤固氮根瘤菌ORS571中一共存在37个GGDEF和EAL结构域蛋白。突变株△4658的运动能力较野生型有下降,但是其胞外多糖合成能力、生物膜形成能力和竞争性结瘤能力较野生型有提高。此外,实验结果表明突变株△4658的胞内c-di-GMP水平高于野生型。突变株△3085和△3226的各种表型与野生型相比没有明显差异。[结论]茎瘤固氮根瘤菌ORS571编码如此大数量的GGDEF和EAL结构域蛋白,表明c-di-GMP可能在其信号转导过程中起到非常重要的作用。同时具有GGDEF和EAL结构域的蛋白AZC_4658对茎瘤固氮根瘤菌ORS571的运动能力、胞外多糖合成、生物膜形成及与宿主的结瘤起到一定的调节作用。 相似文献
