The effect of alpha-tocopherol on the structure and thermotropic phase behaviour of 1-palmitoyl-2-oleoyl-phosphatidylethanolamine dispersed in excess water was examined by synchrotron X-ray diffraction and differential scanning calorimetry. Small- and wide-angle X-ray scattering intensity profiles were recorded from mixed dispersions containing up to 20 mol% alpha-tocopherol during temperature scans over the range 10-75 degrees C. These showed that a domain enriched in alpha-tocopherol phase separated from pure phospholipid in the mixture. This domain tends to have inverted hexagonal structure which coexists with phospholipid bilayers depleted of alpha-tocopherol. The scattering intensity and dimensions of the phase are dependent on the temperature and proportion of alpha-tocopherol in the mixture. Phase separations were also manifest in calorimetric scans of the mixed dispersions evidenced from the appearance of multiple peaks at temperatures corresponding to transitions observed in the X-ray scattering experiments. The effect of alpha-tocopherol in the range 0-20 mol% on the phase behaviour and structure of the phospholipid as observed from the X-ray scattering and calorimetric results have been used to construct a partial phase diagram of the mixture in the temperature range 10-75 degrees C. This shows that alpha-tocopherol has a marked tendency to partition from bilayers of the phospholipid to form an enriched domain in which the phospholipid assumes a hexagonal-II structure.  相似文献

Background

The associations between common variants in the fat mass- and obesity-associated (FTO) gene and obesity-related traits may be age-dependent and may differ by sex. The present study aimed to assess the association of FTO rs9939609 with body mass index (BMI) and the risk of obesity from childhood to adolescence, and to determine the age at which the association becomes evident.

Methods

Totally 757 obese and 2,746 non-obese Chinese children aged 6–18 years were genotyped for FTO rs9939609. Of these, a young sub-cohort (n = 777) aged 6–11 years was reexamined 6 years later. Obesity was defined using the sex- and age-specific BMI cut-offs recommended by the International Obesity Task Force.

Results

The associations of FTO rs9939609 with BMI and obesity did not appear until children reached 12–14 years. The variant was associated with an increased BMI in boys (β = 1.50, P = 0.004) and girls (β = 0.97, P = 0.018), respectively. Thereafter, the magnitude of association increased in girls at ages 15–18 years (β = 2.02, P<0.001), but not boys (β = 0.10, P>0.05). Age was found to interact with the variant on BMI (P<0.001) and obesity (P = 0.042) only in girls. In the sub-cohort, the associations of FTO rs9939609 with BMI (β = 1.07, P = 0.008) and obesity (OR = 2.09, 95% CI: 1.12, 3.91) were only observed 6 years later (ages 12–18 years) in girls, even after adjusting for baseline BMI.

Conclusions

The association between FTO rs9939609 and obesity-related traits may change from childhood to adolescence in Chinese individuals, and the association may start as early as age 12 years, especially in girls.  相似文献

A newly isolated anti-Streptococcus suis bacteriocin-producing strain LPL1-5 was obtained from healthy unweaned piglets' fecal matter, and was designated as Lactobacillus pentosus LPL1-5 based on morphology, biochemical properties, and 16S rDNA sequencing analysis. The medium composition for enhanced bacteriocin production by L. pentosus LPL1-5 was optimized by statistical methodology. Yeast extract, K(2)HPO(4)?·?3H(2)O, and MnSO(4)?·?H(2)O were identified as significant components influencing pentocin LPL1-5 production using the Plackett-Burman method. Response surface methodology was applied for further optimization. The concentrations of medium components for enhanced pentocin LPL1-5 production were as follows (g/L): lactose 20.00, tryptone 10.00, beef extract 10.00, yeast extract 14.00, MnSO(4)?·?H(2)O 0.84, K(2)HPO(4)?·?3H(2)O 4.92, triammonium citrate 2.00, Na-acetate 5.00, MgSO(4)?·?7H(2)O 0.58, Tween 80 1.00. Under the optimized condition, a value of 3154.65?±?27.93 IU/mL bacteriocin activity was achieved, which was 4.2-fold that of the original medium.  相似文献

基于宁夏全区121个土壤剖面样点,研究不同深度(0—30、30—80、80—120cm)土壤有机碳的空间分布特征,通过相关性、方差分解和构建结构方程模型,分析气候、植被和土壤因素对宁夏不同深度土壤有机碳的影响及其作用途径。结果表明:(1)宁夏不同深度土壤有机碳含量均呈现中间低南北高的空间分布趋势,0—120cm剖面土壤有机碳含量随着土壤深度的增加而降低,土壤有机碳含量均值为5.49g/kg,变异系数达90.71%,含量偏低且空间异质性强。(2)各土层土壤有机碳与年均气温、干燥度、碳酸钙、pH均呈现极显著的负相关性(P<0.01),与年均降水量、相对湿度、植被净初级生产力、全氮、全磷、全钾、钙离子、阳离子交换量、粘粒含量均呈现极显著的正相关性(P<0.01)。(3)各因素对不同土层土壤有机碳的作用方式和影响程度有差异。土壤全氮、全磷、碳酸钙、阳离子交换量均能直接影响土壤有机碳,年均气温和植被净初级生产力对土壤有机碳的直接影响效应不显著,主要通过土壤属性间接影响土壤有机碳含量。随着土层深度的增加,气候和植被因素作用明显减弱,土壤因素作用增强并成为主要影响因素。该结果有助于宁夏土...  相似文献

Up-regulation of integrin alpha(v)beta(3) has been shown to play a key role in tumor angiogenesis and metastasis. In this study, we evaluated the role of integrin alpha(v)beta(3) in breast cancer cell resistance to ionizing irradiation (IR) and tested the anti-tumor efficacy of combining integrin alpha(v) siRNA and IR. Colonogenic survival assay, cell proliferation, apoptosis, and cell cycle analysis were carried out to determine the treatment effect of siRNA, IR, or combination of both on MDA-MB-435 cells (integrin alpha(v)beta(3)-positive). Integrin alpha(v)beta(3)-negative MCF-7 cells exert more radiosensitivity than MDA-MB-435 cells. IR up-regulates integrin alpha(v)beta(3) expression in MDA-MB-435 cells and integrin alpha(v) siRNA can effectively reduce both alpha(v) and alpha(v)beta(3) integrin expression, leading to increased radiosensitivity. Integrin alpha(v) siRNA also promotes IR-induced apoptosis and enhances IR-induced G2/M arrest in cell cycle progression. This study, with further optimization, may provide a simple and highly efficient treatment strategy for breast cancer as well as other integrin alpha(v)beta(3)-positive cancer types.  相似文献
