Myocardial infarction (MI) leads to cardiac remodelling and heart failure. Cardiomyocyte apoptosis is considered a critical pathological phenomenon accompanying MI, but the pathogenesis mechanism remains to be explored. MicroRNAs (miRs), with the identity of negative regulator of gene expression, exist as an important contributor to apoptosis. During the experiment of this study, MI mice models were successfully established and sequencing data showed that the expression of miR-23a-5p was significantly enhanced during MI progression. Further steps were taken and it showed that apoptosis of cardiac cells weakened as miR-23a-5p was downregulated and on the contrary that apoptosis strengthened with the overexpression of miR-23a-5p. To explore its working mechanisms, bioinformatics analysis was conducted by referring to multi-databases to predict the targets of miR-23a-5p. Further analysis suggested that those downstream genes enriched in several pathways, especially in the PI3K/Akt singling pathway. Furthermore, it demonstrated that miR-23a-5p was negatively related to the phosphorylation of PI3K/Akt, which plays a critical role in triggering cell apoptosis during MI. Recilisib-activated PI3K/Akt singling pathway could restrain apoptosis from inducing miR-23a-5p overexpression, and Miltefosine-blocked PI3K/Akt singling pathway could restrict apoptosis from inhibiting miR-23a-5p reduction. In conclusion, these findings revealed the pivotal role of miR-23a-5p-PI3K/Akt axis in regulating apoptosis during MI, introducing this novel axis as a potential indicator to detect ischemic heart disease and it could be used for therapeutic intervention.  相似文献

Purpose

China is the world’s largest producer and consumer of refined and reclaimed copper because of the rapid economic and industrial development of this country. However, only a few studies have analyzed the environmental impact of China’s copper industry. The current study analyzes the life cycle environmental impact of copper production in China.

Methods

A life cycle impact assessment using the ReCiPe method was conducted to estimate the environmental impact of refined and reclaimed copper production in China. Uncertainty analysis was also performed based on the Monte-Carlo simulation.

Results and discussion

The environmental impact of refined copper was higher than that of reclaimed copper in almost all categories except for human toxicity because of the direct atmospheric arsenic emission during the copper recycling stage. The overall environmental impact for the refined copper production was mainly attributed to metal depletion, freshwater ecotoxicity, marine ecotoxicity, and water depletion potential impact. By contrast, that for the reclaimed copper production was mainly caused by human toxicity impact.

Conclusions

Results show that the reclaimed copper scenario had approximately 59 to 99% more environmental benefits than those of the refined copper scenario in most key categories except for human toxicity, in which a similar environmental burden was observed between both scenarios. The key factors that reduce the overall environmental impact for China’s copper industry include decreasing direct heavy metal emissions in air and water, increasing the national recycling rate of copper, improving electricity consumption efficiency, replacing coal with clean energy sources for electricity production, and optimizing the efficiency of copper ore mining and consumption.

Achieving light harvesting is crucial for the efficiency of the solar cell. Constructing optical structures often can benefit from micro‐nanophotonic imprinting. Here, a simple and facile strategy is developed to introduce a large area grating structure into the perovskite‐active layer of a solar cell by utilizing commercial optical discs (CD‐R and DVD‐R) and achieve high photovoltaic performance. The constructed diffraction grating on the perovskite active layer realizes nanophotonic light trapping by diffraction and effectively suppresses carrier recombination. Compared to the pristine perovskite solar cells (PSCs), the diffraction‐grating perovskite devices with DVD obtain higher power conversion efficiency and photocurrent density, which are improved from 16.71% and 21.67 mA cm?2 to 19.71% and 23.11 mA cm?2. Moreover, the stability of the PSCs with diffraction‐grating‐structured perovskite active layer is greatly enhanced. The method can boost photonics merge into the remarkable perovskite materials for various applications.  相似文献

Aggrecanases are now believed to be the principal proteinases responsible for aggrecan degradation in osteoarthritis. Given their potential as a drug target, we solved crystal structures of the two most active human aggrecanase isoforms, ADAMTS4 and ADAMTS5, each in complex with bound inhibitor and one wherein the enzyme is in apo form. These structures show that the unliganded and inhibitor-bound enzymes exhibit two essentially different catalytic-site configurations: an autoinhibited, nonbinding, closed form and an open, binding form. On this basis, we propose that mature aggrecanases exist as an ensemble of at least two isomers, only one of which is proteolytically active.  相似文献

The enantiomerization and enantioselective bioaccumulation of metalaxyl by a single dose of exposure to Tenebrio molitor larvae under laboratory condition were studied by high‐performance liquid chromatography tandem mass spectroscopy (HPLC‐MS/MS) based on a ChiralcelOD‐3R [cellulosetris‐tris‐(3, 5‐dichlorophenyl‐carbamate)] column. Exposure of enantiopure R‐metalaxyl and S‐metalaxyl in Tenebrio molitor larvae exhibited significant enantiomerization, with formation of the R enantiomers from the S enantiomers, and vice versa, which might be attributed to the chiral pesticide catalyzed by a certain enzyme in Tenebrio molitor larvae. Enantiomerization was not observed in wheat bran during the period of 21 d. In addition, bioaccumulation of rac‐metalaxyl in Tenebrio molitor larvae was enantioselective with a preferential accumulation of S‐metalaxyl. These results showed that enantioselectivity was caused not only by actual degradation and metabolism but also by enantiomerization, which was an important process in the environmental fate and behavior of metalaxyl enantiomers. 通过对不同地理株骨条藻形态和亚显微结构的观察和描述 ,鉴定为中肋骨条藻 ,但发现在刺长、细胞间隙、细胞链形态、链上细胞数 ,色素体个数方面存在差别 ;通过不同 N∶ P营养盐实验 ,结果表明 ,在 N∶ P为 16∶ 1时 ,东海株和胶州湾株中肋骨条藻均得到最大生长率 ,分别为 1.6 6 d1 和 1.5 3d- 1 ,东海株中肋骨条藻最大生长率要高于胶州湾株中肋骨条藻 ,这表明在不同的海域环境条件下 ,中肋骨条藻的生长均有各自地域性特征。应用 RAPD技术对两地理株中肋骨条藻进行鉴别 ,以其全 DNA为模板进行 RAPD扩增 ,两不同地理株的中肋骨条藻表现不同的 DNA多态性 ,同样以各自的小亚基片段为模板进行的 RAPD扩增条带也表现出差异性。实验结果表明 ,两株中肋骨条藻应为种下的不同变种或亚种  相似文献

微藻化感作用是一种极其复杂的生理、生态学现象。选取强壮前沟藻指数生长初期Ⅰ和平台生长初期Ⅱ两个阶段的滤液对中肋骨条藻、海洋原甲藻、锥状斯氏藻及球等鞭金藻生长的影响进行了研究,并萃取了阶段Ⅱ的粗提物,抑藻检测表明其具有"杀藻"效应,通过GC/MS分析该粗提物中具有潜在化感作用的物质种类。研究发现强壮前沟藻两个生长阶段的滤液对中肋骨条藻均产生强烈致死效应(phaseⅠ:F=15.18475,P=0.00298<0.05;phaseⅡ:F=6.24559,P=0.03149<0.05);锥状斯氏藻在强壮前沟藻滤液中生长,实验结束时两个阶段中的细胞密度分别是对照组的79.3%和68.9%;海洋原甲藻在强壮前沟藻生长阶段Ⅱ滤液实验的最后3d,其生长受到显著抑制(F=4.84438,P=0.04925<0.05);而等鞭金藻在强壮前沟藻两个生长阶段滤液中被抑制现象不明显(P>0.05)。强壮前沟藻滤液实验表明,强壮前沟藻能够向微环境中分泌代谢产物来抑制中肋骨条藻和海洋原甲藻的生长,并且这种抑制效应具有种类特殊对应性。上述实验结果还表明,强壮前沟藻生长阶段Ⅱ的滤液具有的生长抑制作用较为明显。采用乙酸乙酯萃取强壮前沟藻生长阶段Ⅱ滤液中的代谢产物,检测发现其代谢粗提物具有溶藻效应,GC/MS分析结果表明粗提物中存在4种可能产生化感抑制作用的物质,其中二丁基羟基甲苯(Butylated Hydroxytoluene BHT)被认为具有抗滤过性病原体和抗微生物活性。  相似文献

城市绿地是城市自然景观的重要组成部分,探讨不同影响因素对城市绿地降温效应的影响规律,有助于城市绿地的空间布局规划与管理,以便更好地缓解城市热环境。以南京紫金山森林公园为研究区,沿陵园路—中山陵方向选取了5个植被结构相似的林地作为观测点,并以水泥地面为参照点,利用HOBO小型气象观测站对温度、湿度和风速等因子进行定点同步观测,并分别采用LAI-2200植物冠层分析仪、鱼眼相机和GPS手持导航仪获取各观测点的叶面积指数、天空可见度和高程信息,然后采用统计分析和相关分析方法,分别对绿地降温效应的日变化规律及其影响因素进行了定量分析。结果表明:各站点的温度变化趋势基本一致;与参考点相比,5个观测点在白天均有明显降温效果,13:00—16:00降温效应最强,而晚上降温效果很弱,甚至出现轻微的保温效应;降温幅度随大气温度的升高而增大,随相对湿度和风速的增大而减小;绿地叶面积指数、天空可见度和高程等因子对绿地降温效应在不同时段会产生不同程度的影响:叶面积指数与天空可见度在9:00—19:00间对降温效应的影响作用较为明显,但由于高温的影响,午后(13:00—15:00)影响作用较弱;在夜间,高程是主要影响因子。研究结果可为城市公园绿地的合理布局与绿化建设提供依据和参考。  相似文献
