We follow up on a suggestion by Rolls and co-workers, that the effects of competitive learning should be assessed on the shape
and number of spatial fields that dentate gyrus (DG) granule cells may form when receiving input from medial entorhinal cortex
(mEC) grid units. We consider a simple non-dynamical model where DG units are described by a threshold-linear transfer function,
and receive feedforward inputs from 1,000 mEC model grid units of various spacing, orientation and spatial phase. Feedforward
weights are updated according to a Hebbian rule as the virtual rodent follows a long simulated trajectory through a single
environment. Dentate activity is constrained to be very sparse. We find that indeed competitive Hebbian learning tends to
result in a few active DG units with a single place field each, rounded in shape and made larger by iterative weight changes.
These effects are more pronounced when produced with thousands of DG units and inputs per DG unit, which the realistic system
has available, than with fewer units and inputs, in which case several DG units persists with multiple fields. The emergence
of single-field units with learning is in contrast, however, to recent data indicating that most active DG units do have multiple
fields. We show how multiple irregularly arranged fields can be produced by the addition of non-space selective lateral entorhinal
cortex (lEC) units, which are modelled as simply providing an additional effective input specific to each DG unit. The mean
number of such multiple DG fields is enhanced, in particular, when lEC and mEC inputs have overall similar variance across
DG units. Finally, we show that in a restricted environment the mean size of the fields is unaltered, while their mean number
is scaled down with the area of the environment.

Cadmium is a nephrotoxic metal widely used in industry and the main source of Cd in general population is smoking. Considering that the source of Cd in cigarettes is the tobacco leaf, the exposure to Cd was evaluated in workers employed at a tobacco leaf processing factory. Blood and urinary Cd levels were measured by flameless atomic absorption spectrometry in 87 workers and 35 controls. Urinary enzymes, total protein, albumin and uric acid were also determined to investigate the possible nephrotoxic effects of Cd. Blood Cd levels were significantly higher in workers (1.63 +/- 1.95 microg/L) than in controls (0.91 +/- 1.15 microg/L) (p = 0.044). The increase observed in urinary Cd levels of workers was non significant (0.56 +/- 0.5 microg/g creatinine in workers and 0.46 +/- 0.5 microg/g creatinine in controls). Both in workers and in controls, subjects smoking >10 cigarettes/day showed significantly increased blood Cd levels compared to non-smokers (p = 0.000 and p = 0.011, respectively). In workers, urinary alkaline phosphatase (ALP), gamma glutamyl transferase (GGT), total protein, and uric acid were observed to be significantly increased (p = 0.013, p = 0.000, p = 0.000, p = 0.025, respectively), ALP, GGT and total protein being positively correlated with Cd in urine. In conclusion, the workers in the tobacco leaf processing factory were found to be exposed to Cd compared to the general population. The increase in the urinary enzymes and proteins suggests that an exposure to Cd affects kidney functions even below the toxic limits generally accepted. 相似文献

The antioxidant and anticancer properties of a medicinal plant, Betula platyphylla var. japonica were investigated. The total methanol extract of B. platyphylla var. japonica had protective effects against hydrogen peroxide (H2O2) in the Chinese hamster lung fibroblast (V79-4) cell line and induced apoptotic cell death in human promyelocytic leukemia (HL-60) cells, a cancer cell line. B. platyphylla var. japonica extract significantly increased cell viability against H2O2. The extract also showed high 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (IC50 2.4 microg/ml) and lipid peroxidation inhibitory activity (IC50 below 4.0 microg/ml). Furthermore, B. platyphylla var. japonica extract reduced the number of V79-4 cells arrested in G2/M in response to H2O2 treatment and increased the activities of several cellular antioxidant enzymes, including superoxide dismutase, catalase and glutathione peroxidase. Treatment with B. platyphylla var. japonica extract induced cytotoxicity and apoptosis in HL-60 cells, as shown by nucleosomal DNA fragmentation, increases in the subdiploid cell population, and fluorescence microscopy. B. platyphylla var. japonica extract gradually increased the expression of pro-apoptotic Bax and led to the activation of caspase-3 and cleavage of PARP. These findings suggest that B. platyphylla var. japonica exhibits potential antioxidant and anticancer properties. 相似文献

The protein-tyrosine phosphatase Shp2 plays an essential role in growth factor and integrin signaling, and Shp2 mutations cause developmental defects and/or malignancy. Previous work has placed Shp2 upstream of Ras. However, the mechanism of Shp2 action and its substrate(s) are poorly defined. Additional Shp2 functions downstream of, or parallel to, Ras/Erk activation also are proposed. Here, we show that Shp2 promotes Src family kinase (SFK) activation by regulating the phosphorylation of the Csk regulator PAG/Cbp, thereby controlling Csk access to SFKs. In Shp2-deficient cells, SFK inhibitory C-terminal tyrosines are hyperphosphorylated, and the tyrosyl phosphorylation of multiple SFK substrates, including Plcgamma1, is decreased. Decreased Plcgamma1 phosphorylation leads to defective Ras activation on endomembranes, and may help account for impaired Erk activation in Shp2-deficient cells. Decreased phosphorylation/activation of other SFK substrates may explain additional consequences of Shp2 deficiency, including altered cell spreading, stress fibers, focal adhesions, and motility. 相似文献

细胞自噬是植物逆境应答过程中最常见的保护机制之一。动物中,自噬相关基因抵御镉(Cd)毒害的功能研究较清楚,但植物却知之甚少。文中以芹菜品种‘皇后’为试材,采用外源Cd(终浓度为0、2、4、8mg/L)添加营养液水培处理,利用转录组测序(RNA-seq)技术筛选细胞自噬相关差异基因并进行q RT-PCR验证。结果表明Cd胁迫对芹菜植株产生了明显的毒害作用,并与浓度间产生了量效关系。在筛选的8个差异表达的自噬相关基因中,ATG8a、ATG8f、ATG13、AMPK-1、AMPK-2基因随Cd浓度升高表达上调,ATG12、VPS30和VPS34则先上调后下降,说明自噬相关基因可能通过表达上调增加了自噬小体结构以抵御Cd毒性作用;而高浓度Cd(8mg/L)可能超出芹菜的耐受范围,导致多个自噬基因又出现表达下调趋势。以上结果有助于后期自噬相关基因的功能研究,为进一步探讨芹菜对Cd胁迫的耐性机制提供参考依据。 相似文献
