Activity patterns and time budgets are 2 important aspects of animal behavior that researchers use to investigate ecological influences on individual behavior. We collected data on activity patterns and time budgets in 1 group of François’ langurs (Trachypithecus francoisi) from August 2003 to July 2004 in the Nonggang Nature Reserve, Guangxi Province, China, via instantaneous scan sampling method with 15-min intervals. The diurnal activity pattern of François’ langurs showed morning and afternoon feeding peaks, with a midday resting peak. Seasonal change was apparent in the activity pattern: 2 significant feeding peaks occurred in the dry season and only 1 significant feeding peak in the rainy season. The group spent an average of 51.5% of the daytime resting. Feeding and moving accounted on average for 23.1% and 17.3% of the activity budget, respectively. Subjects spent little time on social activities, averaging 2% for grooming and 5.5% for playing. Their time budgets showed significant seasonal variation: they spent a greater proportion of time on feeding and less time on resting and grooming in the dry season than in the rainy season. They also differed among different sex-age classes: immatures spent more time playing, whereas adults devoted more time to resting, feeding, and grooming. Correlations between time budgets and food items or food availability clearly indicated that François’ langurs might adopt an energy-maximizing strategy when preferred foods were scarce in the dry season. 相似文献

Sponges are a source of compounds with potential pharmaceutical applications. In this article, methods of sponge cell culture for production of these bioactive compounds are reviewed, and new approaches for overcoming the problem of metabolite supply are examined. The use of embryos is proposed as a new source of sponge material for cell culture. Stem cells are present in high amounts in embryos and are more versatile and resistant to infections than adult cells. Additionally, genetic engineering and cellular research on apoptotic mechanisms are promising new fields that might help to improve cell survival in sponge-cell lines. We propose that one topic for future research should be how to reduce apoptosis, which appears to be very high in sponge cell cultures. 相似文献

Characterization of novel linuron-mineralizing bacterial consortia enriched from long-term linuron-treated agricultural soils 总被引:2,自引:0,他引:2
Linuron-mineralizing cultures were enriched from two linuron-treated agricultural soils in the presence and absence of a solid support. The cultures contained linuron-degrading bacteria, which coexisted with bacteria degrading either 3,4-dichloroaniline (3,4-DCA) or N,O-dimethylhydroxylamine (N,O-DMHA), two common metabolites in the linuron degradation pathway. For one soil, the presence of a solid support enriched for linuron-degrading strains phylogenetically related to but different from those enriched without support. Most linuron-degrading consortium members were identified as Variovorax, but a Hydrogenophaga and an Achromobacter strain capable of linuron degradation were also obtained. Several of the linuron-degrading isolates also degraded 3,4-DCA. Isolates that degraded 3,4-DCA but not linuron belonged to the genera Variovorax, Cupriavidus and Afipia. Hyphomicrobium spp. were involved in the metabolism of N,O-DMHA. Whereas several isolates degraded linuron independently, more efficient degradation was achieved by combining linuron and 3,4-DCA-degraders or by adding casamino acids. These data suggest that (1) linuron degradation is performed by a group of metabolically interacting bacteria rather than by individual strains, (2) there are other genera in addition to Variovorax that degrade linuron beyond 3,4-DCA, (3) linuron-degrading consortia of different origins have a similar composition, and (4) interactions between consortium members can be complex and can involve exchange of both metabolites and other nutrients. 相似文献

We investigated the geometric representations underlying the perception of 2-D contour curvature. 88 arcs representing lower and upper halves of concentric circles, or halves of ellipses derived mathematically through planar projection by affinity with the circles, a special case of Newton's transform, were generated to produce curved line segments with negative and positive curvature and varying sagitta (sag) and/or aspect ratio. Aspect ratio is defined here as the ratio between the sagitta and the chord-length of a given arc. The geometric properties of the arcs suggest a regrouping into four structural models. The 88 stimuli were presented in random order to 16 observers eight of whom were experienced in the mathematical and visual analysis of 2-D curvature ('expert observers'), and eight of whom were not ('non-expert observers'). Observers had to give a number, on a psychophysical scale from 0 to 10, that was to reflect the magnitude of curvature they perceived in a given arc. The results show that the subjective magnitude of curvature increases exponentially with the aspect ratio and linearly with the sagitta of the arcs for both experts and nonexperts. Statistical analysis of the correlation coefficients of linear fits to individual data represented on a logarithmic scale reveals significantly higher correlation coefficients for aspect ratio than for sagitta. The difference is not significant when curves with the longest chords only (7 degrees -10 degrees ) are considered. The geometric model that produces the best psychometric functions is described by a combination of arcs of vertically and horizontally oriented ellipses, indicating that perceptual sensations of 2-D contour curvature are based on geometric representations that suggest properties of 3-D structures. A 'buckled bar model' is shown to optimally account for the perceptual data of all observers with the exception of one expert. His perceptual data can be linked to a more analytical, less 'naturalistic' representation originating from a specific perceptual experience, which is discussed. It is concluded that the structural properties of 'real' objects are likely to determine even the most basic geometric representations underlying the perception of curvature in 2-D images. We conducted a systematic assessment and comparative study on the biochemical and cellular characteristics of cultured cotton
cells during the entire process of somatic embryogenesis (SE). All staged cultures were widely investigated in this assay.
Cell and tissue ectogenesis manipulation combined with flow cytometry (FCM) was employed to cellular study during the whole
totipotency process of dedifferentiation and redifferentiation. We identified two phases of chromatin decondensation during
the dedifferentiation and redifferentiation. At the same time, sharp increase in the ratio of indoleacetic acid (IAA), isopentenyladenosine
group (iPAs) at the same stage of cell dedifferentiation and redifferentiation process serve as distinct biochemical maker
of dedifferentiation and SE initiation with the unique feature. Our results suggest the two phases of chromatin reorganization
associated with endogenous auxin/cytokinin dynamic activity may underlie dedifferentiation and redifferentiation during the

ADP-glucose pyrophosphorylase (AGPase) plays a key role in regulating starch biosynthesis in cereal seeds and is likely the
most important determinant of seed strength. The Escherichia coli mutant glgC gene (glgC16), which encodes a highly active and allosterically insensitive AGPase, was introduced into maize (Zea mays L.) under the control of an endosperm-specific promoter. Developing seeds from transgenic maize plants showed up to 2–4-fold
higher levels of AGPase activity in the presence of 5 mM inorganic phosphate (Pi). Transgenic plants with higher cytoplasmic
AGPase activity under Pi-inhibitory conditions showed increases (13–25%) in seed weight over the untransformed control. In
addition, in all transgenic maize plants, the seeds were fully filled, and the seed number of transgenic plants had no significant
difference compared with that of untransformed control. These results indicate that increasing cytoplasmic AGPase activity
has a marked effect on sink activity and, in turn, seed weight in transgenic maize plants. 相似文献

A recent publication by Kim et al. on penetrating both human embryonic kidney cells and mouse embryonic stem cells with Si nanowires highlights the increasing interest in using proven semiconductor materials not only to detect specific biomolecules in solutions but also to deliver genetic material or potentially screen for the presence of particular molecules at the cell level. Soil organic matter (SOM) is often separated by physical means to simplify a complex matrix into discrete fractions. A frequent
approach to isolating two or more fractions is based on differing particle densities and uses a high density liquid such as
sodium polytungstate (SPT). Soil density fractions are often interpreted as organic matter pools with different carbon (C)
turnover times, ranging from years to decades or centuries, and with different functional roles for C and nutrient dynamics.
In this paper, we discuss the development and mechanistic basis of common density-based methods for dividing soil into distinct
organic matter fractions. Further, we directly address the potential effects of dispersing soil in a high density salt solution
on the recovered fractions and implications for data interpretation. Soil collected from forested sites at H. J. Andrews Experimental
Forest, Oregon and Bousson Experimental Forest, Pennsylvania was separated into light and heavy fractions by floatation in
a 1.6 g cm−3 solution of SPT. Mass balance calculations revealed that between 17% and 26% of the original bulk soil C and N content was
mobilized and subsequently discarded during density fractionation for both soils. In some cases, the light isotope was preferentially
mobilized during density fractionation. During a year-long incubation, mathematically recombined density fractions respired
∼40% less than the bulk soil at both sites and light fraction (LF) did not always decompose more than the heavy fraction (HF).
Residual amounts of tungsten (W) present even in well-rinsed fractions were enough to reduce microbial respiration by 27%
compared to the control in a 90-day incubation of Oa material. However, residual W was nearly eliminated by repeated leaching over the year-long incubation, and is not likely
the primary cause of the difference in respiration between summed fractions and bulk soil. Light fraction at Bousson, a deciduous
site developed on Alfisols, had a radiocarbon-based mean residence time (MRT) of 2.7 or 89 years, depending on the interpretation
of the radiocarbon model, while HF was 317 years. In contrast, both density fractions from H. J. Andrews, a coniferous site
developed on andic soils, had approximately the same MRT (117 years and 93 years for LF and HF). At H. J. Andrews the organic
matter lost during density separation had a short MRT (19 years) and can account for the difference in respired CO2 between the summed fractions and the bulk soil. Recognition and consideration of the effects of the density separation procedure
on the recovered fractions will help prevent misinterpretation and deepen our understanding of the specific role of the recovered
organic matter fractions in the ecological context of the soil studied. 相似文献

Extraction of intracellular protein from Escherichia coli is traditionally achieved by mechanical, chemical or enzymatic disruption technology. In this study, a novel thermolysis method was used to disrupt E. coli cells to release a recombinant thermostable esterase. We found that heat treatment of E. coli was highly effective to destroy the integrity of bacterial cell walls and release the recombinant hyperthermophilic esterase at temperatures above 60 degrees C. The effects of temperature, pH and cell concentration on the efficiency of cell disruption were examined. The most effective temperature for cell disruption was at 80 degrees C. The pH and cell concentration had only minor effect on the release of the hyperthermophilic esterase. In addition, we found that the hyperthermophilic esterase could be purified at the early stage of the thermolysis, which is a major advantage of the thermolysis method. Finally, a comparison between thermolysis and traditional methods for the disruption of cells and the release of the thermostable enzyme was made. 相似文献
