Deletion or mutation of the SMN1 (survival of motor neurons) gene causes the common, fatal neuromuscular disease spinal muscular atrophy. The SMN protein is important in small nuclear ribonucleoprotein (snRNP) assembly and interacts with snRNP proteins via arginine/glycine-rich domains. Recently, SMN was also found to interact with core protein components of the two major families of small nucleolar RNPs, fibrillarin and GAR1, suggesting that SMN may also function in the assembly of small nucleolar RNPs. Here we present results that indicate that the interaction of SMN with GAR1 is mediated by the Tudor domain of SMN. Single point mutations within the Tudor domain, including a spinal muscular atrophy patient mutation, impair the interaction of SMN with GAR1. Furthermore, we find that either of the two arginine/glycine-rich domains of GAR1 can provide for interaction with SMN, but removal of both results in loss of the interaction. Finally, we have found that unlike the interaction of SMN with the Sm snRNP proteins, interaction with GAR1 and fibrillarin is not enhanced by arginine dimethylation. Our results argue against post-translational arginine dimethylation as a general requirement for SMN recognition of proteins bearing arginine/glycine-rich domains.  相似文献

利用RAPD标记技术,对8个沙梨(Pyrus pyrifolia)主栽品种进行RAPD分析。结果显示,从33个10碱基随机引物中筛选出了2个多态性高的引物S2075和S1296,扩增位点分别为11个和10个,以此为基础建立了8个品种的DNA指纹图谱;根据这2个引物中任何一个的扩增图谱均可以将这8个品种区分。研究结果为沙梨品种的区别与鉴定提供了一种有效方法。  相似文献

通过咸水灌溉沙土土质生长的幼龄胡杨,分析了咸水灌溉沙土土壤盐分分布累积特点、盐分胁迫对胡杨的耗水生长关系、叶绿素、Pro、MDA的影响,结果表明:(1)在1.2—3 g/L范围内,微咸水灌溉沙土处于脱盐状态,6—12 g/L咸水灌溉使沙土积盐大增。在整个生长周期内,微咸水和咸水灌溉,0—200 cm内土体的总盐都呈累积趋势。(2)咸水灌溉胡杨,不同盐分处理的生长耗水关系可以用对数模型描述。(3)盐分胁迫下,胡杨叶片内叶绿素含量呈抛物线递减,Pro和MDA含量则呈现抛物线递增趋势。说明短期内咸水灌溉对土壤安全和胡杨的生长影响有限,可用咸水解决生态缺水现状,3种生理指标可用来衡量胡杨的盐胁迫程度,以此为指导提高人工造林的成活率。  相似文献

Four diverse disturbance types, namely, farmland, rotationally grazed grassland, overgrazed grassland and forbidden grazing grassland, were identified in the Alashan desert region of Inner Mongolia. Rodents were sampled in April, July and October in 2002 and 2003 using the trap-day method. Their spatial and temporal niches among the four disturbance types were assessed using the Shannon-Wiener niche breadth index and the Pianka niche overlap index. The Shannon-Wiener niche breadth index was the highest for Allactaga sibirica and Meriones meridianus (0.925 and 0.908, respectively), intermediate for Cricetulus eversmanni, Phodopus roborovskii and Citellus dauricus (0.789, 0.782 and 0.711, respectively), and lowest for Cardiocranius paradoxus (0.003). The Pianka niche overlap indices of six-paired species were the highest and varied from 0.900 to 1.000, suggesting their spatial niche overlapped almost completely. Conversely, the spatial niche overlap index for Meriones unguiculatus, Cardiocranius paradoxus, and Salpingotus kozlovi was zero, thus indicating a complete absence of competition among them. In addition, the temporal niche breadth and overlap indices varied greatly in relation to disturbance types and season. The level of temporal niche overlap in spring and autumn was low while in summer it was high. 通过原生质体融合和培养获得苜蓿红豆草属间体细胞杂种植株。采用一种简便方法从杂种组织再生植株叶片、红豆草羟脯氨酸抗性系再生植株叶片和苜蓿根癌农杆菌702转化系愈伤组织提取DNA用于RAPD和Southern杂交分析。随机引物扩增结果显示两种亲本的RAPD多态具有明显差异。在所用20种随机引物中,6种产生较多的DNA片段。杂种组织具有两种亲本特有的DNA片段,但倾向于排除红豆草亲本的染色体,表明该杂种为非对称杂种,两种亲本染色体之间可能发生了重组。由于红豆草DNA的介入,杂种组织表现出较强的分化能力。分别利用RAPD扩增得到的OPA141000bp红豆草羟脯氨酸抗性系特异产物和OPA141600bp苜蓿根癌农杆菌702转化系特异产物为探针进行Southern分子杂交,证明杂种组织同时具有这两种DNA片段的同源序列。  相似文献

干旱与条锈病复合胁迫对小麦的生理影响   总被引:1,自引:0,他引:1  
以抗旱性和抗病性不同的小麦为材料,以正常生长为对照,观察了病原菌和水分复合胁迫对小麦叶片相对含水量、活性氧代谢以及对抗氰呼吸的发生、运行的影响。讨论了在干旱与病原菌侵染复合胁迫下,抗氰呼吸在植物抗逆机制中所扮演的角色。复合胁迫下,抗病小麦显然具备更强的水分调控能力,而感病品种不能有效控制病叶水分散失。水分胁迫能引起抗氰呼吸的下降,但不能抵消因病原菌侵染引起的抗氰呼吸的增强,条锈菌侵染对小麦抗氰呼吸的影响远远大于水分胁迫。病原菌侵染和水分复合胁迫下,活性氧产生的速率表现出累加效应,而抗氰呼吸表现出和基质抗氧化酶的活性互补。植物交替氧化酶在干旱与病原菌侵染复合胁迫中具有重要的抗氧化功能,并可能调节着逆境下物质与能量需求间的矛盾。  相似文献

中间锦鸡儿(Caragana intermedia)耐旱、耐寒、耐盐碱,是西北干旱地区的重要固沙灌木,筛选其优良抗逆境基因,可以作为林草基因工程的基因源。该研究在中间锦鸡儿干旱胁迫转录组文库中找到1条CiPUB22 (plant U box 22)基因的cDNA全长序列,CiPUB22基因包括1 260 bp开放阅读框,编码419个氨基酸。实时荧光定量PCR结果表明,在脱水、盐和ABA处理1 h后CiPUB22基因表达量上升并达到最高水平,分别为对照表达量的12倍、35倍和7倍,干旱处理后12 d达到最高值,为对照的2.5倍,表明CiPUB22的转录水平受非生物胁迫诱导。构建CiPUB22基因的过表达载体并转化野生型拟南芥,对转基因纯合体株系抗逆性分析发现,在150 mmol/L NaCl、1 μmol/L ABA和400 mmol/L甘露醇处理下,过表达株系的萌发率均低于野生型,说明过表达CiPUB22基因降低了拟南芥在种子萌发过程中对盐和渗透胁迫的耐受性。  相似文献

在对毛锦藓科的分类学修订中,我们报道3个种和1变种在中国的新分布记录,它们是:外弯小锦藓Brotherella recurvans、纤枝疣胞藓Clastobryum cuculligerum、海岛麻锦藓Taxithelium liukiuense、扭尖金枝藓阔叶变种Trismegistia calderensis var. rigida。这些新记录物种的标本分别采自海南、香港和云南。  相似文献

Plastin-3 plays a key role in cancer cell proliferation and invasion, but its prognostic value in pancreatic cancer (PACA) remains poorly defined. In this study, we show that PLS3 messenger RNA is overexpressed in PACA tissue compared with normal tissue. We accumulated 207 cases of PACA specimens to perform immunohistochemical analysis and demonstrated that PLS3 levels correlate with T-classification (p < .001) and pathology (p < .001). Furthermore, overall survival rates (p < .001) in tumors with high PLS3 expression were poor, as assessed through Kaplan–Meier survival analysis. PLS3 was found to be an independent prognostic factor for PACA through multivariate Cox regression analysis. Moreover, we found that PLS3 enhances the proliferation and invasion of tumor cells as assessed through Cell Counting Kit-8, wounding healing assays, and Transwell assays. The upregulation of PLS3 also led to enhanced phosphatidylinositol-3 kinase/protein kinase B signaling in PACA cells. These data suggest that PLS3 is a biomarker to estimate PACA progression and represents a molecular target for PACA therapy.  相似文献
