Adaptive immune systems are present only in vertebrates. How do all the remaining animals withstand continuous attacks of permanently evolving pathogens? Even in the absence of adaptive immunity, every organism must be able to unambiguously distinguish "self" cells from any imaginable "nonself." Here, we analyzed the function of highly polymorphic gene vCRL1, which is expressed in follicle and blood cells of Ciona intestinalis, pointing to possible recognition roles either during fertilization or in immune reactions. By using segregation analysis, we demonstrate that vCRL1 locus is not involved in the control of self-sterility. Interestingly, genetic knockdown of vCRL1 in all tissues or specifically in hemocytes results in a drastic developmental arrest during metamorphosis exactly when blood system formation in Ciona normally occurs. Our data demonstrate that vCRL1 gene might be essential for the establishment of a functional blood system in Ciona. Presumably, presence of the vCRL1 receptor on the surface of blood cells renders them as self, whereas any cell lacking it is referred to as nonself and will be consequently destroyed. Calcium (Ca2+) plays crucial roles in regulation of pollen tube growth. The influx of Ca2+ into the pollen tube is mediated by ion channels, and the density and activity of Ca2+ channels in pollen plasma membranes critically determines their electrical properties. In this report, using whole-cell and single-channel patch-clamping techniques, we investigated developmental changes of hyperpolarization-activated Ca2+ channel activity in pear (Pyrus pyrifolia) pollen and its relationship with pollen viability. For both pollen and pollen tubes, hyperpolarization-activated Ca2+ channels had the same conductance and cAMP sensitivity, indicating that they were the same channels. However, the Ca2+ current density in pollen tube protoplasts was greater than that in pollen protoplasts. Compared with day-3 flowers’ pollen, hyperpolarization-activated Ca2+ current density was significantly lower in day 0 and day 3 flowers’ pollen, which was consistent with the pollen germination and pollen tube growth, indicating that pollen protoplasts’ increased Ca2+ current density may have enhanced the pollen viability. During pollen tube elongation, pollen tube plasma membrane Ca2+ current density increased with increased length pollen tubes up to 300 μm. All of these results indicated that hyperpolarization-activated Ca2+ channel activity was associated with in pear pollen development and may have a causal link between Ca2+ channel activity and pollen viability. 相似文献

expression pattern. Meanwhile, we compared the CAPN3 mRNA expression pattern in MB chicken at 10 weeks with a commercial meat type chicken line (S01) of the same age to identify
the unique expression pattern under different genetic background. A real time quantitative PCR (qRT-PCR) assay was developed
for an accurate measurement of its expression in various tissues from chickens at different ages (0, 2, 4, 6, 8, 10, and 12 weeks).
Expression of the CAPN3 mRNA was detected in the selected tissues, regardless of age. The breast muscle and leg muscle tissues had a significantly
higher expression than the other tissues from the same individual (P < 0.01). Overall, the CAPN3 mRNA level exhibited a “rise-decline” developmental change in detected tissues except for brain. The S01 chicken had a higher
expression of the CAPN3 mRNA in detected tissues than the MB chicken at 10 weeks. The present expression data of chicken CAPN3 gene may provide some information to shed light on the tissue and ontogenic expression pattern during chicken development. 相似文献
The decline in DNA repair capacity contributes to the age‐associated decrease in genome integrity in somatic cells of different species. However, due to the lack of clinical samples and appropriate tools for studying DNA repair, whether and how age‐associated changes in DNA repair result in a loss of genome integrity of human adult stem cells remains incompletely characterized. Here, we isolated 20 eyelid adipose‐derived stem cell (ADSC) lines from healthy individuals (young: 10 donors with ages ranging 17–25 years; old: 10 donors with ages ranging 50–59 years). Using these cell lines, we systematically compared the efficiency of base excision repair (BER) and two DNA double‐strand break (DSB) repair pathways—nonhomologous end joining (NHEJ) and homologous recombination (HR)—between the young and old groups. Surprisingly, we found that the efficiency of BER but not NHEJ or HR is impaired in aged human ADSCs, which is in contrast to previous findings that DSB repair declines with age in human fibroblasts. We also demonstrated that BER efficiency is negatively associated with tail moment, which reflects a loss of genome integrity in human ADSCs. Mechanistic studies indicated that at the protein level XRCC1, but not other BER factors, exhibited age‐associated decline. Overexpression of XRCC1 reversed the decline of BER efficiency and genome integrity, indicating that XRCC1 is a potential therapeutic target for stabilizing genomes in aged ADSCs. 相似文献
Gastric cancer (GC) is a prevalent malignant cancer of digestive system, identification of novel diagnostic and prognostic biomarkers for GC is urgently demanded. The aim of this study was to determine potential long noncoding RNAs (lncRNAs) associated with the pathogenesis and prognosis of GC. Raw noncoding RNA microarray data (GSE53137, GSE70880, and GSE99417) was downloaded from Gene Expression Omnibus (GEO) database. Differentially expressed genes between GC and adjacent normal gastric tissue samples were screened by an integrated analysis of multiple gene expression profile after gene reannotation and batch normalization. Differentially expressed genes were further confirmed by The Cancer Genome Atlas (TCGA) database. Competing endogenous RNA (ceRNA) network, Gene Ontology term and Kyoto Encyclopedia of Genes and Genomes pathway, survival analysis were extensively applied to identify hub lncRNAs and discover potential biomarkers related to diagnosis and prognosis of GC. In total of 246 integrated differential genes including 15 lncRNAs and 241 messenger RNAs (mRNAs) were obtained after intersections of differential genes between GEO and TCGA database. ceRNA network comprised of three lncRNAs (UCA1, HOTTIP, and HMGA1P4), 26 microRNAs (miRNAs) and 72 mRNAs. Functional analysis revealed that three lncRNAs were mainly dominated in cell cycle and cellular senescence. Survival analysis showed that HMGA1P4 was statistically related to the overall survival rate. For the first time, we identified that HMGA1P4, a target of miR-301b/miR-508, is involved in cell cycle and senescence process by regulating CCNA2 in GC. Finally, the expression levels of three lncRNAs were validated to be upregulated in GC tissues. Thus, three lncRNAs including UCA1, HOTTIP, and HMGA1P4 may contribute to GC development and their potential functions might be associated with the prognosis of GC. 相似文献
Sclerotinia stem rot, caused by Sclerotinia sclerotiorum, is a devastating disease in Henan Province, of the main rapeseed production areas in China. Fluazinam belongs to the broad‐spectrum phenylpyridinamine fungicides, which have high activity in inhibiting the mycelial growth of S. sclerotiorum. In this study, 191 field isolates were obtained from different oilseed rape fields in Henan Province, before being exposed to fluazinam in 2015. The baseline sensitivity of S. sclerotiorum to fluazinam was established. The effective concentration for 50% inhibition of mycelial growth (EC50) ranged from 0.0019 to 0.0337 μg/ml, and the mean EC50 value was 0.0084 ± 0.0055 μg/ml. The range of the frequency distribution was narrow. The results of a cross‐resistance assay revealed no cross‐resistance between fluazinam and carbendazim, dimethachlone, boscalid or fludioxonil. Field efficacy tests showed that the control efficacies of fluazinam (50% WG) applied at 150, 225 and 300 g ai ha?1 were 67%, 73% and 88%, respectively. In contrast, the control efficacies of boscalid (50% WG) and carbendazim (50% WP) applied at 225 and 1,500 g ai ha?1 were 71% and 52%, respectively. 相似文献
This study evaluated the effects of foliar spraying melatonin (MT) on the growth of salt-stressed rice. Seedlings were treated with 50 and 100 mM of NaCl and different concentrations of MT (25, 50, 100, 200, 300, and 400 μM) for 14 days. Different concentrations of MT could promote plant growth significantly under salt stress, particularly at concentrations of 200, 300, and 400 μM. A concentration of 200 μM MT was considered as optimal and used in a subsequent experiment on biomass, water content, antioxidation, mineral nutrition, salt absorption, and distribution of salt-stressed rice seedlings. Results showed that MT’s promoting effect on plant growth under salt stress was evident with time, particularly under high salt stress. MT improved the activities of antioxidant enzymes, reduced membrane lipid peroxidation, alleviated cell injury in plant leaves, and increased N content and Si accumulation in the leaves and roots under salt stress, particularly under high salinity. This compound also inhibited Na uptake and upward transport, but it promoted or maintained the uptake and upward transport of K and Ca in salt-stressed rice. Thus, MT improved the ion homeostasis of K/Na and Ca/Na in plants, particularly in the leaves. Foliar spraying of MT alleviated salt stress on rice by promoting nutrient accumulation or translocation, improving ion homeostasis, which is evident in the leaves, and consequently enhancing its salt resistance. The antioxidative improvement caused by MT might also be related to the improved ion homeostasis.
