中山杉(Taxodium ‘Zhongshansha’)具有极强的耐淹性, 但其耐淹机理仍没有明确。该研究以‘中山杉118’ (Taxodium ‘Zhongshansha 118’)幼苗为对象, 在经过93天不同水淹处理(对照、水浸、浅淹、深淹)后测定中山杉叶片和根系的无氧呼吸酶活性、淀粉及可溶性糖含量、生物量以及根系活力, 从能量消耗的角度初步探索了中山杉的耐淹性。结果表明: 长期水淹使中山杉叶片与根系中3种无氧呼吸酶(乙醇脱氢酶、丙酮酸脱羧酶、乳酸脱氢酶)活性显著增加, 且叶片与根系的乙醇脱氢酶活性均高于乳酸脱氢酶活性, 中山杉的根系和叶片是通过加强以酒精发酵为主的无氧呼吸适应长期缺氧环境; 不同水淹处理的叶片中3种无氧呼吸酶活性均高于根系, 叶片对缺氧环境更加敏感; 中山杉叶片和根系淀粉、可溶性糖含量均随水淹深度的增加显著增加, 根系淀粉含量显著高于叶片, 可溶性糖含量低于叶片; 中山杉根系淀粉含量高是其能够长期忍受水淹的重要原因, 且中山杉适应长期水淹的策略为忍耐型; 经受长期水淹后中山杉根茎结合部长出气生根及茎基部膨大, 同时根系外壁的木质化能将根系与外部水淹环境隔离, 具有很强的耐淹性, 可作为湿地生态修复、消落带生物治理的优良植物材料。  相似文献

以辣椒品种“超辣九号”为试材,采用15%的PEG6000模拟干旱,研究了0.1μmol·L^-1外源24-表油菜素内酯(EBR)处理对干旱胁迫下辣椒叶片快速叶绿素荧光诱导动力学曲线(OJIP)的影响。结果表明:干旱胁迫降低了辣椒叶片的光化学效率和光合性能,导致干旱光抑制的发生。干旱胁迫既损伤了辣椒叶片PSⅡ供体侧放氧复合体(OEC),同时也对PSⅡ反应中心和受体侧造成伤害,阻碍了光合电子传递;干旱胁迫还导致单位叶面积有活性反应中心数目(RC/CS)的下降,并降低了单位叶面积吸收的光能(ABS/CS)、捕获的光能(TRo/CS)和进行电子传递的能量(ETo/CS),同时诱导了单位叶面积热耗散(DIo/CS)的增加。这说明辣椒遭受干旱胁迫后启动了相应的防御机制,一方面通过PSⅡ的可逆失活减少光能吸收与传递,另一方面通过促进热耗散减少过剩激发能的积累。EBR处理改善了干旱胁迫下辣椒叶片PSⅡ受体侧的电子传递,缓解了单位叶面积有活性反应中心数目的减少,优化了光合电子传递的进行,并维持相对较高的热耗散能力,从而减轻了干旱光抑制程度,对干旱胁迫下辣椒叶片光合机构和光合性能起到保护作用。  相似文献

Background

Although the ultrastructure of the schistosome esophageal gland was described >35 years ago, its role in the processing of ingested blood has never been established. The current study was prompted by our identification of MEG-4.1 expression in the gland and the observation of erythrocyte uncoating in the posterior esophagus.

Methodology/Principal Findings

The salient feature of the posterior esophagus, characterized by confocal and electron microscopy, is the enormous increase in membrane surface area provided by the plate-like extensions and basal invaginations of the lining syncytium, with unique crystalloid vesicles releasing their contents between the plates. The feeding process was shown by video microscopy to be divided into two phases, blood first accumulating in the anterior lumen before passing as a bolus to the posterior. There it streamed around a plug of material revealed by confocal microscopy as tethered leucocytes. These were present in far larger numbers than predicted from the volume of the lumen, and in varying states of damage and destruction. Intact erythrocytes were detected in the anterior esophagus but not observed thereafter, implying that their lysis occurred rapidly as they enter the posterior. Two further genes, MEGs 4.2 and 14, were shown to be expressed exclusively in the esophageal gland. Bioinformatics predicted that MEGs 4.1 and 4.2 possessed a common hydrophobic region with a shared motif, while antibodies to SjMEG-4.1 showed it was bound to leucocytes in the esophageal lumen. It was also predicted that MEGs 4.1 and 14 were heavily O-glycosylated and this was confirmed for the former by 2D-electrophoresis and Western blotting.

Conclusions/Significance

The esophageal gland and its products play a central role in the processing of ingested blood. The binding of host antibodies in the esophageal lumen shows that some constituents are antibody targets and could provide a new source of vaccine candidates.  相似文献

研究了锦熟黄杨(Buxus sempervirens L.)在低温胁迫下不同光照条件(12 h光照/12 h黑暗、24 h全光、24 h全黑)对其抗氧化酶活性的影响,结果表明:低温不同光照条件下细胞膜透性和丙二醛含量均高于对照(20℃ 12 h光照/12 h黑暗),细胞膜透性和丙二醛含量在5℃ 24 h光照条件下最大;低温胁迫下SOD活性高于对照,并在5℃ 24 h光照条件下达到最高值;CAT的活性仍维持较高水平,5℃ 12 h光照/12 h黑暗显著高于对照及其它处理;在低温有光照条件下,POD活性升高,黑暗条件下POD活性低于对照。低温胁迫下POD、SOD和CAT的活性均呈上升趋势,可能是植株具有较强抗性的原因。  相似文献

We isolated an MgATG1 gene encoding a serine/threonine protein kinase from the rice blast fungus Magnaporthe grisea. In the DeltaMgatg1 mutant, in which the MgATG1 gene had been deleted, autophagy was blocked; the mutant also showed fewer lipid droplets in its conidia, lower turgor pressure of the appressorium, and such defects in morphogenesis as delayed initiation and slower germination of conidia. As a result of lower turgor pressure of the appressorium, the DeltaMgatg1 mutant lost its ability to penetrate and infect the two host plants, namely, rice and barley. However, normal values of the parameters and infective abilities were restored on reintroducing an intact copy of the MgATG1 gene into the mutant. Autophagy is thus necessary for turnover of organic matter during the formation of conidia and appressoria and for normal development and pathogenicity in M. grisea.  相似文献

The NG2 chondroitin sulfate proteoglycan is a membrane-spanning molecule expressed by immature precursor cells in a variety of developing tissues. In tightly adherent cell lines with a flattened morphology, NG2 is organized on the cell surface in linear arrays that are highly co-localized with actin and myosin-containing stress fibers in the cytoskeleton. In contrast, microtubules and intermediate filaments in the cytoskeleton exhibit completely different patterns of organization, suggesting that NG2 may use microfilamentous stress fibers as a means of cytoskeletal anchorage. Consistent with this is the observation that cytochalasin D disrupts the organization of both stress fibers in the cytoskeleton and NG2 on the cell surface. Very similar linear cell surface arrays are also seen with three other cell surface molecules thought to interact with the actin cytoskeleton: the α5β1 integrin, the CD44 proteoglycan, and the L1 neuronal cell adhesion molecule. Since the cytoplasmic domains of these four molecules are dissimilar, it seems possible that cytoskeletal anchorage in each case may occur via different mechanisms. One indication of such differences can be seen in colchicine-treated cells which have lost their flattened morphology but still retain long actin-positive tendrils as remnants of the actin cytoskeleton. NG2 and α5β1 are associated with these tendrils while CD44 and L1 are not, suggesting that at least two subclasses of cell surface molecules exist which can interact with different subdomains of the actin cytoskeleton. © 1996 Wiley-Liss, Inc.  相似文献

Background

Preterm birth, the birth of an infant prior to 37 completed weeks of gestation, is the leading cause of perinatal morbidity and mortality. Preterm infants are at greater risk of respiratory, gastrointestinal and neurological diseases. Despite significant research in developed countries, little is known about the causes of preterm birth in many developing countries, especially China. This study investigates the association between sciodemographic data, obstetric risk factor, and preterm birth in five Maternal and Child Health hospitals in Beijing, China.

Methods and Findings

A case-control study was conducted on 1391 women with preterm birth (case group) and 1391 women with term delivery (control group), who were interviewed within 48 hours of delivery. Sixteen potential factors were investigated and statistical analysis was performed by univariate analysis and logistic regression analysis. Univariate analysis showed that 14 of the 16 factors were associated with preterm birth. Inter-pregnancy interval and inherited diseases were not risk factors. Logistic regression analysis showed that obesity (odds ratio (OR) = 3.030, 95% confidence interval (CI) 1.166–7.869), stressful life events (OR = 5.535, 95%CI 2.315–13.231), sexual activity (OR = 1.674, 95%CI 1.279–2.191), placenta previa (OR 13.577, 95%CI 2.563–71.912), gestational diabetes mellitus (OR = 3.441, 95%CI1.694–6.991), hypertensive disorder complicating pregnancy (OR = 6.034, 95%CI = 3.401–10.704), history of preterm birth (OR = 20.888, 95%CI 2.519–173.218) and reproductive abnormalities (OR = 3.049, 95%CI 1.010–9.206) were independent risk factors. Women who lived in towns and cities (OR = 0.603, 95%CI 0.430–0.846), had a balanced diet (OR = 0.533, 95%CI 0.421–0.675) and had a record of prenatal care (OR = 0.261, 95%CI 0.134–0.510) were less likely to have preterm birth.

Conclusions

Obesity, stressful life events, sexual activity, placenta previa, gestational diabetes mellitus, hypertensive disorder complicating pregnancy, history of preterm birth and reproductive abnormalities are independent risk factors to preterm birth. Identification of remedial factors may inform local health and education policy.  相似文献

以江西省50种珍稀濒危植物为定量研究对象,采用以濒危系数、遗传价值系数、物种价值系数和生物学特征系数4个指标构成的珍稀濒危植物等级综合评价值的方法对上述植物优先保护顺序进行了定量研究。对50种珍稀植物进行排序的结果基本符合《中国珍稀濒危植物》和《中国植物红皮书》第1册的分析,但也有不同:属一级保护的有长柄双花木、天目木姜子和长序榆等共10种,占研究物种总数的20.00%,其中有浙江楠和华东黄杉等2渐危种;二级保护的有胡豆莲、连香树、伯乐树、银钟花等共26种,占52.00%,其中胡豆莲属于濒危种;三级的有小花木兰、观光木等12种,占24.00%,其中有4种属稀有种,8种属渐危种;此外,八角莲、野大豆等在江西可以暂不列入保护,这两者均属渐危种,占4.00%。  相似文献
