生长素是最重要的植物激素之一,参与了植物生长发育的各个方面。植物体内游离的IAA是生长素的主要活性形式,在IAA甲基转移酶1(IAMT1)的作用下,IAA可以转变为IAA甲酯(MelAA)。MelAA本身没有活性,在植物体内的MelAA酯解酶作用下可以重新转变为IAA。MelAA是非极性分子,能够在植物体内自由扩散。利用MelAA的这种特殊性质筛选突变体,可以分离到MelAA代谢途径或者IAA途径中新的成分。我们对拟南芥种子进行EMS诱变,通过观察黑暗下下胚轴的生长情况,筛选MelAA的抗性突变体。我们成功分离到了8株可能的抗性突变体,并对其中的一个Methyl-JAAresistant1(mir1)突变体进行了深入分析。MelAA抗性突变体的筛选将为进一步了解MelAA的代谢、IAA稳态调控和响应机理提供新的材料。  相似文献

Chemical communication plays an important role in density‐dependent phase change in locusts. However, the volatile components and emission patterns of the migratory locust, Locusta migratoria, are largely unknown. In this study, we identified the chemical compositions and emission dynamics of locust volatiles from the body and feces and associated them with developmental stages, sexes and phase changes. The migratory locust shares a number of volatile components with the desert locust (Schistocerca gregaria), but the emission dynamics of the two locust species are significantly different. The body odors of the gregarious nymphs in the migratory locust consisted of phenylacetonitrile (PAN), benzaldehyde, guaiacol, phenol, aliphatic acids and 2,3‐butanediol, and PAN was the dominant volatile. Volatiles from the fecal pellets of the nymphs primarily consist of guaiacol and phenol. Principal component analysis (PCA) showed significant differences in the volatile profiles between gregarious and solitary locusts. PAN and 4‐vinylanisole concentrations were significantly higher in gregarious individuals than in solitary locusts. Gregarious mature males released significantly higher amounts of PAN and 4‐vinylanisole during adulthood than mature females and immature adults of both sexes. Furthermore, PAN and 4‐vinylanisole were completely lost in gregarious nymphs during the solitarization process, but were obtained by solitary nymphs during gregarization. The amounts of benzaldehyde, guaiacol and phenol only unidirectionally decreased from solitary to crowded treatment. Aliphatic aldehydes (C7 to C10), which were previously reported as locust volatiles, are now identified as environmental contaminants. Therefore, our results illustrate the precise odor profiles of migratory locusts during developmental stages, sexes and phase change. However, the function and role of PAN and other aromatic compounds during phase transition need further investigation.  相似文献

胭脂花是分布于中国北部的报春花科报春花属花柱二型植物,为了阐明胭脂花花粉形态的多样性,对胭脂花分布范围内的7个野生居群的花粉形态进行了测定。结果表明,胭脂花花粉属N3P4C5型,极面观多为三角形,具三孔沟,少数为四边形,具4孔沟;短花柱植株的花粉粒较大(16.53×17.58μm),花粉粒外壁粗糙,表面纹饰多,网眼大;长花柱植株的花粉粒较小(12.95×13.72μm),花粉粒外壁较光滑,表面纹饰少,网眼小。不同居群间花粉粒在大小、网眼直径、外壁纹饰等方面存在显著差异,花粉形态变异系数在4.88%~35.48%之间;短花柱类型各居群的平均变异系数为16.18%,居群间差异较大,其中庞泉沟居群变异系数最大(24.05%),海坨山居群最小(10.13%);长花柱类型各居群的平均变异系数为14.89%,庞泉沟居群的变异系数最大(19.75%),五台山与海坨山居群最小(12.50%),长花柱类型的花粉性状比短花柱植株更为稳定。UPGMA聚类分析发现,7个居群聚为两大类,除庞泉沟居群外,其他居群聚为一组,地理距离相近的居群一般聚在一起。研究结果为胭脂花种质资源遗传多样性及育种研究提供了依据。  相似文献

Major histocompatibility complex (MHC) class I information is vital for understanding variance of immune responses in HIV vaccination and biomedical models. In this study, 9 Mamu-A and 13 Mamu-B alleles were identified from the cDNA products of 10 Chinese-origin rhesus macaques. Except for two alleles that had been reported by others, eight were novel and twelve extended the partial sequences that are available in GenBank. The additional information of MHC class I antigens might be beneficial to the availability of Chinese macaques in human disease studies. Furthermore, the polymorphism of leading peptides and the natural killer receptor recognition motifs in alpha1 domain both implies that Mamu-A and Mamu-B molecules might play key roles in innate immune responses of natural killer cells.  相似文献

MHC四聚体技术是研究抗原特异性淋巴细胞应答的关键技术之一。为研究H-2Db基因型(如C57BL/6)小鼠的特异性CD8+ T细胞免疫应答, 需要建立H-2Db四聚体制备技术平台。首先以RT-PCR方法克隆H-2Db重链基因的cDNA, 进而构建H-2Db胞外域与生物素化酶BirA底物肽(BSP)融合蛋白的表达载体, 并在大肠杆菌中获得表达。在LCMV GP33-41抗原肽(KAVYNFATC, KAV)和人β2-微球蛋白存在时, 通过稀释法复性获得H-2Db/KAV单体。该单体经生物素化并纯化后与PE-链亲和素按4: 1的比例混合, 即形成四聚体。通过流式细胞术检测经KAV肽免疫的C57BL/6小鼠体内的LCMV特异性CD8+ T细胞的频率, 结果表明在外周血、引流淋巴结和脾脏中均可检测到一定频率的LCMV特异性CD8+ T细胞, 其中以对外周血标本染色的效果最佳。成功建立了小鼠H-2Db四聚体制备技术平台, 为监测及分析基于H-2Db基因型小鼠的实验性免疫治疗创造了条件。  相似文献

The RACE technique was used to clone the full‐length vitellogenin (VTG) cDNA sequence of Asian arowana (Scleropages formosus). The full‐length sequence was 5,550 bp with an open reading frame of 5,238 bp, encoding 1,745 amino acids, and 5′ and 3′ UTRs (untranslated regions) of 45 bp and 267 bp, respectively. Phylogenetic analysis showed that S. formosus and silver arowana (Osteoglossum bicirrhosum) share a close evolutionary relationship (bootstrap 100%). The quantitative real‐time PCR results showed that vtg expression was significantly higher in liver and gonads of male and female fish compared with its expression in the other tissues tested (p < 0.01). The relative expression levels of vtg in liver, gland, kidney, heart, head kidney, and brain of female fish were significantly higher than in the corresponding tissues of male fish (p < 0.05).  相似文献

Background

Polymorphisms of the brain-derived neurotrophic factor (BDNF) have been investigated as candidate genes for post-stroke depression (PSD), and its receptor, neurotrophic tyrosine kinase receptor B (TrkB), has been associated with depression. However, no further data have yet reported the association between PSD and polymorphisms in TrkB. This study aims to investigate whether a relationship exists between TrkB polymorphisms and PSD.

Methods

A total of 312 depression patients (PSD patients) and 472 non-depression patient controls (NPSD patients) were recruited. All patients were evaluated using the Hamilton Rating Scale for Depression (HAMD) to determine depression severity, and PSD patients were diagnosed in accordance with DSM-V criteria. Three single-nucleotide polymorphisms (SNPs), namely, rs1187323, rs1212171, and rs1778929, in the TrkB gene were genotyped by high-resolution melt analysis.

Results

The SNP rs1778929 was significantly more associated with incident PSD in participants with the TT genotype than in those with CC (OR 0.482, 95% CI: 0.313–0.744). In terms of rs1187323, stroke was significantly more associated with incident depression in participants with the AC genotype than in those with AA (OR 0.500, 95% CI: 0.368–0.680). The minor allele (T) of rs1778929 (P = 0.024, OR = 0.725, 95% CI = 0.590–0.890) and the minor allele (C) of rs1187323 (P = 0.000, OR = 0.598, 95% CI = 0.466–0.767) were found to be significantly associated with PSD. Neither genotype nor allele frequencies of rs1212171 showed statistically significant differences between PSD and NPSD patients.

Conclusions

The results suggest that rs1778929 and rs1187323 in the TrkB gene are significantly associated with post-stroke depression in the Chinese population. Further studies are necessary to confirm our findings.  相似文献

氮添加对高寒草甸土壤微生物呼吸及其温度敏感性的影响   总被引:3,自引:0,他引:3  
土壤氮素的可利用性是控制土壤微生物呼吸的重要因素之一,大量研究已经表明增加土壤活性氮的含量可以降低微生物呼吸,但是土壤氮输入对土壤微生物呼吸温度敏感性的影响还不清楚。以青藏高原高寒草甸为研究对象,通过野外施氮试验和室内控制试验相结合的方式,在5℃、15℃和25℃条件下对3种施氮水平的土壤(对照,0g N m~(-2)a~(-1);低氮,5g N m~(-2)a~(-1);高氮,15g N m~(-2)a~(-1))进行培养,探讨土壤微生物呼吸及其温度敏感性对不同氮添加水平的响应情况。结果表明:(1)3个温度培养下的土壤微生物呼吸速率和累积碳释放量均随施氮量的增加而显著降低(P0.05);(2)氮添加对5℃和15℃培养条件下的微生物呼吸温度敏感性没有显著影响,但显著地增加了15℃和25℃培养条件下的微生物呼吸温度敏感性(P0.05);(3)线性相关分析表明,土壤累积碳释放量与土壤有机碳的难降解性显著负相关(P0.05),而15℃和25℃培养条件下的微生物呼吸温度敏感性与土壤有机碳的难降解性显著正相关(P0.05)。结果表明,在全球气候变暖的背景下,土壤氮输入将增加预测青藏高原高寒草甸地区土壤碳排放的不确定性。  相似文献
