Pancreatic cancer is one of the most lethal malignancies, with a prominent desmoplastic reaction as its defining hallmark. The past several decades have seen dramatic progress in understanding of pancreatic cancer pathogenesis, including identification of precursor lesions, sequential transformation from normal pancreatic tissue to invasive pancreatic cancer and corresponding signature genetic events, and the biological impact of these events on malignant behavior. However, the currently used therapeutic strategies for epithelial tumor cells, which have exhibited potent antitumor activity in cell culture and animal models, have failed to produce significant effects in the clinic. The desmoplastic stroma surrounding pancreatic cancer cells, which accounts for about 90% of a tumor's mass, clearly is not a passive scaffold for cancer cells but an active contributor to carcinogenesis. Improved understanding of the dynamic interaction between cancer cells and the stroma will be important to designing effective therapeutic strategies for pancreatic cancer. This review focuses on the origin of stromal molecular and cellular components in pancreatic tumors, their biological effects on pancreatic cancer cells, and the orchestration of these two components.  相似文献

为了探讨芋(Colocasia esculenta(L.)Schott)试管球茎膨大期间糖类物质积累特点,以红香芋无菌试管苗为材料,研究了高浓度蔗糖诱导条件下,红香芋试管球茎形成及膨大过程中主要碳水化合物的变化规律,以及与相关酶活性的关系。结果表明:(1)在红香芋试管球茎膨大过程中,果糖、葡萄糖和总可溶性糖含量均呈先升高后降低的变化趋势,果糖含量在诱导至第27天时达到最大值,而总可溶性糖和葡萄糖含量均在第34天达到峰值;蔗糖含量呈现先上升、后下降、再上升的变化趋势,在培养第48天时积累量达到最大值。(2)红香芋试管球茎总淀粉含量、直链和支链淀粉含量均随培养时间的延长而增加,至膨大后期总淀粉含量达到最大值,淀粉总含量约占干重的76%,并以支链淀粉含量为主。(3)解剖学观察发现,随着试管球茎的形成与膨大,贮藏组织中淀粉粒密度不断增大,至球茎膨大后期,淀粉粒布满薄壁细胞,并且处于比较稳定的水平。(4)诱导培养至第41天时,试管球茎的ADPG焦磷酸化酶和Q-酶活性均达到最大值,分别为1.22和2.39μmol·g~(-1)·min~(-1)。相关性分析发现,从茎基部开始膨大(20d)至ADPG焦磷酸化酶和Q-酶活性达峰值(41d)时,ADPG焦磷酸化酶活性与总淀粉含量、Q-酶活性与支链淀粉含量的相关系数分别为0.819和0.738,二者均呈极显著正相关。研究认为,淀粉的积累以及可溶性糖类含量的变化与红香芋试管球茎的膨大发育密切相关,并受到相关酶的调控。  相似文献

A rapid, sensitive and specific liquid chromatography-tandem mass spectrometry (LC-MS-MS) method has been developed and validated for the simultaneous analysis of hydrocodone (HYC) and its metabolite hydromorphone (HYM) in human plasma. A robotic liquid handler and a 96-channel liquid handling workstation were used to aliquot samples, to add internal standard (I.S.), and to extract analytes of interest. A 96-well mixed-mode solid-phase cartridge plate was used to extract the analytes and I.S. The chromatographic separation was on a silica column (50 x 3 mm, 5-microm) with a mobile phase consisting of acetonitrile, water and trifluoroacetic acid (TFA) (92:8:0.01, v/v). The run time for each injection was 2.5 min with the retention times of approximately 2.1 and 2.2 min for HYC and HYM, respectively. The tandem mass spectrometric detection was by monitoring singly charged precursor-->product ion transition 300-->199 (m/z) for HYC, and 28-->185 (m/z) for HYM. The validated calibration curve range was 0.100-100 ng/ml, based on a plasma volume of 0.3 ml. The correlation coefficients were greater than or equal to 0.9996 for both HYC and HYM. The low limit of quantitation (LLOQ) was 0.100 ng/ml for both HYC and HYM with signal-to-noise ratio (S/N) of 50 and 10. respectively. The deuterated analytes, used as internal standards, were monitored at mass transitions 303-->199 (m/z) for HYC-d3 and 289-->185 (m/z) for HYM-d3. The inter-day (n= 17) precision of the quality control (QC) samples were < or = 3.5% RSD (relative standard deviation) for HYC and < or = 4.7% RSD for HYM, respectively. The inter-day accuracy of the QC samples were < or = 2.1% RE (relative error) for HYC and < or = 1.8% RE for HYM. The intra-day (n=6) precision and accuracy of the QC samples were < or = 2.6% RSD and < or = 3.0% RE for HYC, and < or = 4.7% RSD and < or = 2.4% RE for HYM. There was no significant deviation from the nominal values after a 5-fold dilution of high concentration QC samples by blank matrix. The QC samples were stable when kept at room temperature for 24-h or experienced three freeze-thaw cycles. The extraction recoveries were 86% for HYC and 78% for HYM. No detectable carryover was observed when a blank sample was injected immediately after a 2500 ng/ml sample that was 25-fold more concentrated than the upper limit of quantitation (ULOQ).  相似文献

A field survey of metal concentrations and mycorrhizal status of plants growing on copper mine tailings was conducted in Anhui Province, China. Available phosphorus and organic matter in the tailings were very low. High concentrations of Pb, Zn, As and Cd as well as Cu were observed on some sites. The dominant plants growing on mine tailings belonged to the families Gramineae and Compositae, and the most widely distributed plant species wereImperata cylindrica, Cynodon dactylon andPaspalum distichum. Coreopsis drummondii also grew well on the arid sites but not on wet sites. Very low or zero arbuscular mycorrhizal (AM) fungal colonization was observed in most of the plants, but extensive mycorrhizal colonization was recorded in the roots ofC. drummondii andC. dactylon. Metal concentrations in plant tissues indicated that /.cylindrica andP. distichum utilized avoidance mechanisms to survive at high metal concentrations. The investigation suggests that remediation and revegetation of heavy metal contaminated sites might be facilitated by selection of tolerant plant species. Isolation of tolerant AM fungi may also be warranted.  相似文献

Esophageal squamous cell carcinoma (ESCC), a malignant neoplasm with high incidence, is a severe global public health threat. The current modalities used for treating ESCC include surgery, chemotherapy, and radiotherapy. Although ESCC management and treatment strategies have improved over the last decade, the overall 5-year survival rate remains <20%. Therefore, the identification of novel therapeutic strategies that can increase ESCC patient survival rates is urgently needed. Oxethazaine, an amino-amide anesthetic agent, is mainly prescribed in combination with antacids to relieve esophagitis, dyspepsia, and other gastric disorders. In the present study, we found that oxethazaine inhibited the proliferation and migration of esophageal cancer cells. According to the results of in vitro screening and binding assays, oxethazaine binds directly to AURKA, suppresses AURKA activity, and inhibits the downstream effectors of AURKA. Notably, we found that oxethazaine suppressed tumor growth in three patient-derived esophageal xenograft mouse models and tumor metastasis in vivo. Our findings suggest that oxethazaine can inhibit ESCC proliferation and metastasis in vitro and in vivo by targeting AURKA.Subject terms: Cancer prevention, Cell growth  相似文献

Understanding the molecular and cellular mechanisms of human primordial germ cells (hPGCs) is essential in studying infertility and germ cell tumorigenesis. Many RNA-binding proteins (RBPs) and non-coding RNAs are specifically expressed and functional during hPGC developments. However, the roles and regulatory mechanisms of these RBPs and non-coding RNAs, such as microRNAs (miRNAs), in hPGCs remain elusive. In this study, we reported a new regulatory function of DAZL, a germ cell-specific RBP, in miRNA biogenesis and cell proliferation. First, DAZL co-localized with miRNA let-7a in human PGCs and up-regulated the levels of >100 mature miRNAs, including eight out of nine let-7 family, miR21, miR22, miR125, miR10 and miR199. Purified DAZL directly bound to the loops of precursor miRNAs with sequence specificity of GUU. The binding of DAZL to the precursor miRNA increased the maturation of miRNA by enhancing the cleavage activity of DICER. Furthermore, cell proliferation assay and cell cycle analysis confirmed that DAZL inhibited the proliferation of in vitro PGCs by promoting the maturation of these miRNAs. Evidently, the mature miRNAs up-regulated by DAZL silenced cell proliferation regulators including TRIM71. Moreover, DAZL inhibited germline tumor cell proliferation and teratoma formation. These results demonstrate that DAZL regulates hPGC proliferation by enhancing miRNA processing.  相似文献

Introduction  

Osteoarthritis is characterized by the progressive destruction of cartilage in the articular joints. Novel therapies that promote resurfacing of exposed bone in focal areas are of interest in osteoarthritis because they may delay the progression of this disabling disease in patients who develop focal lesions. Recently, the addition of 80% deacetylated chitosan to cartilage microfractures was shown to promote the regeneration of hyaline cartilage. The molecular mechanisms by which chitosan promotes cartilage regeneration remain unknown. Because neutrophils are transiently recruited to the microfracture site, the effect of 80% deacetylated chitosan on the function of neutrophils was investigated. Most studies on neutrophils use preparations of chitosan with an uncertain degree of deacetylation. For therapeutic purposes, it is of interest to determine whether the degree of deacetylation influences the response of neutrophils to chitosan. The effect of 95% deacetylated chitosan on the function of neutrophils was therefore also investigated and compared with that of 80% deacetylated chitosan.  相似文献
