In the human body, arsenic is metabolized by methylation. Understanding this process is important and provides insight into the relationship between arsenic and its related diseases. We used the rapid equilibrium kinetic model to study the reaction sequence of arsenite methylation. The results suggest that the mechanism for arsenite methylation is a completely ordered mechanism that is also of general interest in reaction systems with different reductants, such as tris(2-carboxyethyl)phosphine, cysteine, and glutathione. In the reaction, cysteine residues of recombinant human arsenic (+3 oxidation state) methyltransferase (hAS3MT) coordinate with arsenicals and involve the methyl transfer step. S-Adenosyl-l-methionine (AdoMet) is the first-order reactant, which modulates the conformation of hAS3MT to a best matched state by hydrophobic interaction. As the second-order reactant, reductant reduces the disulfide bond, most likely between Cys-250 and another cysteine residue of hAS3MT, and exposes the active site cysteine residues for binding trivalent inorganic arsenic (iAs3+) to give monomethylarsonic dicysteine (MADC3+). In addition, the reaction can be extended to further methylate MADC3+ to dimethylarsinic cysteine (DAMC3+). In the methylation reaction, the β-pleated sheet content of hAS3MT is increased, and the hydrophobicity of the microenvironment around the active sites is decreased. Similarly, we confirm that both the high β-pleated sheet content of hAS3MT and the high dissociation ability of the enzyme-AdoMet-reductant improve the yield of dimethylated arsenicals.  相似文献

为了探索艾叶挥发油β-环糊精包合的最佳生产工艺,提高挥发油在制剂中的稳定性.本文采用正交实验法,通过测定油利用率、包合物收得率及含油率考察包合工艺.结果表明:最佳生产工艺条件:A3 B2 C3(β-环糊精和艾叶挥发油的比例为8:1,油和乙醇的比例为1:1.5,包合温度为60℃,时间为2 h).  相似文献

The molecular structure of amylopectin and its varphi,beta-limit dextrins from starch of 13 amaranth cultivars was determined by HPAEC-PAD after debranching. Chain length profiles of amylopectins showed bimodal distributions. The molar-based ratios of the average chain lengths of amylopectins (CLap) ranged from 17.41 to 18.22. The molar-based average chain lengths (CLld) and average B-chain lengths (BCLld) of varphi,beta-limit dextrins varied from 7.68 to 8.05, and from 14.10 to 14.73, respectively. Correlation analysis indicated that most structural parameters were positively correlated with thermal properties with few exceptions, whereas the content of fraction fa' ("'" stands for molar-based chain length ratio) was negatively correlated with the thermal properties. Pasting properties of cold paste viscosity (CPV) and setback were also correlated with amylopectin structural parameters.  相似文献

We investigated the role of large-conductance Ca(2+)-activated K(+) (BK) channels in beta3-adrenoceptor (beta3-AR)-induced relaxation in rat urinary bladder smooth muscle (UBSM). BRL 37344, a specific beta3-AR agonist, inhibits spontaneous contractions of isolated UBSM strips. SR59230A, a specific beta3-AR antagonist, and H89, a PKA inhibitor, reduced the inhibitory effect of BRL 37344. Iberiotoxin, a specific BK channel inhibitor, shifts the BRL 37344 concentration response curves for contraction amplitude, net muscle force, and tone to the right. Freshly dispersed UBSM cells and the perforated mode of the patch-clamp technique were used to determine further the role of beta3-AR stimulation by BRL 37344 on BK channel activity. BRL 37344 increased spontaneous, transient, outward BK current (STOC) frequency by 46.0 +/- 20.1%. In whole cell mode at a holding potential of V(h) = 0 mV, the single BK channel amplitude was 5.17 +/- 0.28 pA, whereas in the presence of BRL 37344, it was 5.55 +/- 0.41 pA. The BK channel open probability was also unchanged. In the presence of ryanodine and nifedipine, the current-voltage relationship in response to depolarization steps in the presence and absence of BRL 37344 was identical. In current-clamp mode, BRL 37344 caused membrane potential hyperpolarization from -26.1 +/- 2.1 mV (control) to -29.0 +/- 2.2 mV. The BRL 37344-induced hyperpolarization was eliminated by application of iberiotoxin, tetraethylammonium or ryanodine. The data indicate that stimulation of beta3-AR relaxes rat UBSM by increasing the BK channel STOC frequency, which causes membrane hyperpolarization and thus relaxation.  相似文献

The effects of temperature changes on oxygen consumption of Chinese shrimp (Fenneropenaeus chinensis Osbeck) were studied. The response of oxygen consumption to a temperature rise was conformed to partial metabolic compensation. No compensatory response was observed at lower temperature. A sudden temperature increase by 12 °C resulted an overshoot in oxygen consumption in shrimp adapted to 19 °C, while a sudden decrease by 12 °C in shrimp adapted to 19 °C resulted in an undershoot in oxygen consumption. The shrimp adapted to 31 °C responded with an undershoot in oxygen consumption when a sudden temperature drop by 12 °C occurred. But overshoot in oxygen consumption did not occur when the shrimps were transferred back from 19 to 31 °C. The amplitude of oxygen consumption was reduced in shrimp during the process of acclimation to the temperature diel fluctuation. After the shrimp had adapted to the temperature fluctuation, the daily mean oxygen consumption of shrimp at diel temperature fluctuation from 24 to 30 °C was significantly lower than those adapted to the constant temperature at 27 °C (P<0.05). The decrease in metabolic rate may account for the increase in the growth rate of shrimp at a diel fluctuating thermal regime.  相似文献
