以甘蓝型油菜(Brassica napus L.)豫油2号和6257的子叶和下胚轴为材料,在不同激素配比的MS培养基上诱导出了愈伤组织。将经过继代的部分愈伤转入分化培养基,分化结果表明:除基因型、外植体和分化培养基的激素配比对分化率有影响外,诱导愈伤培养基的激素配比对分化率也至关重要。豫油2号的子叶和下胚轴在最适诱导培养基(ZT 1+NAA0.5+2,4-D 0.2 mg/L)和最适分化培养基(ZT4+IAA 0.2 mg/L)组合中的愈伤分化率分别为12.5%和75%;6257的子叶和下胚轴在其最适诱导培养基(KT 2+NAA1+2,4-D 0.2 mg/L)和最适分化培养基(6-BA 4+IAA 0.02 mg/L)组合中的愈伤分化率分别为50%和37.5%。将其最适诱导培养基中的愈伤组织继代达8个月以上,建立了不同继代愈伤的再生植株无性系。 相似文献

The obligate biotrophic fungus Puccinia striiformis f. sp. tritici (Pst) employs virulence effectors to disturb host immunity and causes devastating stripe rust disease. However, our understanding of how Pst effectors regulate host defense responses remains limited. In this study, we determined that the Pst effector Hasp98, which is highly expressed in Pst haustoria, inhibits plant immune responses triggered by flg22 or nonpathogenic bacteria. Overexpression of Hasp98 in wheat (Triticum aestivum) suppressed avirulent Pst-triggered immunity, leading to decreased H2O2 accumulation and promoting P. striiformis infection, whereas stable silencing of Hasp98 impaired P. striiformis pathogenicity. Hasp98 interacts with the wheat mitogen-activated protein kinase TaMAPK4, a positive regulator of plant resistance to stripe rust. The conserved TEY motif of TaMAPK4 is important for its kinase activity, which is required for the resistance function. We demonstrate that Hasp98 inhibits the kinase activity of TaMAPK4 and that the stable silencing of TaMAPK4 compromises wheat resistance against P. striiformis. These results suggest that Hasp98 acts as a virulence effector to interfere with the MAPK signaling pathway in wheat, thereby promoting P. striiformis infection. 相似文献

Protein nanowires are critical electroactive components for electron transfer of Geobacter sulfurreducens biofilm. To determine the applicability of the nanowire proteins in improving bioelectricity production, their genes including pilA, omcZ, omcS and omcT were overexpressed in G. sulfurreducens. The voltage outputs of the constructed strains were higher than that of the control strain with the empty vector (0.470–0.578 vs. 0.355 V) in microbial fuel cells (MFCs). As a result, the power density of the constructed strains (i.e. 1.39–1.58 W m−2) also increased by 2.62- to 2.97-fold as compared to that of the control strain. Overexpression of nanowire proteins also improved biofilm formation on electrodes with increased protein amount and thickness of biofilms. The normalized power outputs of the constructed strains were 0.18–0.20 W g−1 that increased by 74% to 93% from that of the control strain. Bioelectrochemical analyses further revealed that the biofilms and MFCs with the constructed strains had stronger electroactivity and smaller internal resistance, respectively. Collectively, these results demonstrate for the first time that overexpression of nanowire proteins increases the biomass and electroactivity of anode-attached microbial biofilms. The relationship between head pteridine fluorescence (HPF) levels and age in adult females and males of a common necrophagous fly, Chrysomya megacephala, and effects of temperature and fly sex on the relationship were studied by pteridine fluorescence spectrophotometry. Factors affecting HPF levels in flies were found to include fly age, temperature and fly sex, among which the fly age was the most dominant one. There were significant linear relationships between HPF levels and age both for female and male adult flies at five constant temperatures, i. e. 16°C, 20°C, 24°C, 28°C and 32°C. The relationship between mean rate of pteridine accumulation (FV or MV) and temperature (t) could be well described by a modified exponential equation of FV=0.01288 e(0.2241t‐3.127)+0.3649 (r2= 0.9987) for females and a linear regression equation of MV= 0.0574 t ‐ 0.3637 (r2= 0.9557) for males. Using the information from the experiments at five constant temperatures, three calculated methods as the candidates were developed for accurately determining the age of the fly by HPF levels at ambient temperature. The results revealed that these three methods were suitable for estimating the age only for male flies, but not for female flies. The smallest average error of the predicated age was 2.55 days for males. In addition, how to employ which of these three developed methods for determining ages of male flies in practical was also discussed. 相似文献

大鼠颈动脉注射1%FlNa,荧光显微镜下活体观察肠系膜微血管血流状态及FlNa的渗出情况,并在不同时间点经股动脉采血,测定血浆内FlNa浓度随时间的变化,利用组织匀浆测定不同脏器中FlNa的分布,再辅以冰冻切片进行观察。活体观察发现,FlNa注入体内后,经微血管迅速向周围组织渗出,最后汇集于淋巴管,血浆及组织匀浆FlNa浓度的测定表明,FlNa浓度随时间的变化呈指数衰减,各脏器FlNa的分布极不相同。冰冻切片也显示了同样的分布差别。这些结果表明,我们所建立的方法可直观、定量地反映FlNa在微血管的通透情况。 相似文献
