宋内氏痢疾菌侵袭相关基因的克隆与表达

用粘粒PJB8为载体,构建了宋内氏痢疾菌I相大质粒基因库。用菌落原位杂交,Western blot方法从文库中筛选到1株表达IpaA,B,C,D 4种蛋白的重组子。经重组质粒酶切和Southern blot等方法对宋内氏菌和福氏2a痢疾菌的侵袭相关基因片段作了初步比较分析。

Studies on atrophic change of soleus muscle and its countermeasures in suspended rat

In the environment of microgravity, the disused atrophy of skeletal muscle, especially leg's muscle, would occur. The three purposes of this study were: 1. To observe the dynamic changes of disused atrophy of skeletal muscle under simulated weightlessness; 2. To approach the mechanism of disused atrophy of muscle; 3. To approach the countermeasures for reducing the degree of atrophy of muscle.     

Variation of the rfb gene clusters in Salmonella enterica.

In order to explore the genetic variation of O antigens of Salmonella enterica, we surveyed 164 strains (132 serovars) belonging to 45 serogroups, using 25 mostly single-gene rfb DNA probes for colony hybridization. The results revealed that strains within a serogroup have very similar or identical rfb genes. At least three of the four rhamnose genes were detected in all 17 serogroups reported to contain rhamnose, and one or more were detected in three others. The likelihood of being detected decreased in the order rfbB, rfbC, rfbA, and rfbD, which is the map order, suggesting a gradient of divergence. Mannose pathway genes were much less conserved, and of 27 groups reported to contain mannose or mannose derivatives colitose or fucose, only 9 hybridized to the rfbM and rfbK probes. Dideoxyhexose genes were found only in groups reported to contain dideoxyhexoses. Group D2, which had not been studied previously, appears to resemble group D1, with the substitution of one gene from group E1 to give a change in one linkage. In contrast to sugar pathway genes, sugar transferase genes did not in general hybridize to strains of other groups outside the closely related groups A, B, and D, with the exception of the galactose transferase gene also shared by groups C2, C3, and all E groups.     

Strategies for the computation of infrared CD and absorption spectra of biological molecules: ribonucleic acids.

We report observed and computed infrared (vibrational) circular dichroism spectra of a number of polyribonucleic acids in aqueous solutions in the 1600-1750 cm-1 spectral region, in which C = O and some nucleotide base ring stretching vibrations occur. The experimental data are compared with results calculated using different levels of sophistication within the exciton approach. We find that observed band shapes are generally well reproduced by these models, particularly if care is taken to determine the direction of the vibrational dipole transition moments accurately.     

嗜盐耐盐微生物抗盐胁迫相关离子转运蛋白研究进展

离子转运蛋白在维持细胞内pH稳态、离子动态平衡等方面发挥着重要作用。钠离子转运体和钾离子转运体在嗜盐耐盐微生物中广泛存在,其"保钾排钠"机制是微生物抗盐胁迫的两大策略之一。近年来,嗜盐耐盐微生物中许多新型钠、钾离子转运体被陆续发现,如RDD蛋白、UPF0118蛋白、DUF蛋白和KimA蛋白等;Fe³⁺、Mg²⁺等其他金属离子的转运蛋白也被证实可通过影响微生物胞内相容性溶质的合成起到渗透调节的作用。本文综述了嗜盐耐盐微生物中抗盐胁迫相关的各类离子转运蛋白,分析其分子结构和工作机理,并对这些蛋白在农业方面的应用进行了展望。继续发现新的离子转运蛋白,探究抗盐胁迫相关离子转运蛋白的结构和机理,解析各转运系统的协同作用及分子调控机制,将进一步加深对嗜盐耐盐微生物抗盐胁迫调控的认识,并为盐碱地农作物的改良等提供新的思路。     

论受控于X连锁基因性状的变异性

本文应用数量遗传学方法,结合概率运算,从理论上分析了由X连锁基因控制的性状在雌雄群体中的变异情况。表明雄（男）性群体的这类性状遗传变异范围有大于雌（女）性群体的倾向。对这一问题的探讨,将有助于对X连锁基因及所决定性状的遗传特性、进化特点及基因定位等的研究提供新的思路。     

Conformation and activity ofPhaseolus coccineus var. rubronanus lectin

The conformation of native and denaturedPhaseolus coccineus var. rubronanus lectin was studied by circular dichroism (CD) and correlated to the hemagglutinating activity. The far-UV CD spectrum at 25°C showed a broad, negative band around 223 nm and a positive one at 196 nm. CD data analysis of the lectin indicated a -sheet-rich protein. At high temperatures, the spectrum was blue-shifted with increasing magnitude; these changes correlated well with the loss of the activity. The conformation of lectin betweenpH 2 and 10 remained essentially unchanged. AtpH 13 the CD spectrum resembled that of unordered form with a negative band near 200 nm and the activity was completely lost. The denatured lectin in 6 M guanidine hydrochloride would be renatured upon diluting the denaturant to 0.75 M; the changes in CD spectrum again correlated well with the loss of the activity. The effect of sodium dodecyl sulfate on the lectin was drastic; it sharply increased thea-helix at the expense of the -sheet and reduced the activity; the changes reached a plateau above 20 mM surfactant.     

He—Ne激光对君子兰幼果和胡萝卜根的愈伤组织形成及生长的影响

利用6mv的He—Ne激光器,每天对君子兰幼果、胡萝卜根的外植体辐照5分钟,连续辐照20天,对愈伤组织的形成和生长有一定的促进作用。但同样条件下,辐照10分钟,对君子兰幼果,胡萝卜根的愈伤组织形成和生长都有明显的抑制作用。     

激光辐照真菌的酯酶,淀粉酶同工酶研究

本文报导了用He—Ne激光辐照真菌米曲酶的酯酶同工酶和淀粉同工酶的分析研究结果,实验表明激光对米曲霉同工酶有一定的影响。
