为了探求玉米(Zea mays)光合作用和生长对重复干旱的响应机制, 采用盆栽试验, 分别测定了不同程度土壤干旱处理3周时、随后复水1周时以及再次不同程度干旱处理3周时玉米幼苗光合参数和生长的变化。第一次土壤干旱处理后, 重度干旱处理显著降低玉米株高、单株总叶面积、地上部分及根系生物量以及叶片的蒸腾速率(Tr)、气孔导度(Gs)、胞间CO2浓度(Ci)、净光合速率(Pn)和最大净光合速率(Amax), 但显著提高光补偿点和暗呼吸速率; 中度干旱处理同样显著降低玉米株高、叶面积和地上部分生物量, 但对根系生物量无影响, 因而根冠比增大, 对上述光合参数的负效应也不具有显著性。复水可使前期经受中度和重度干旱处理的玉米植株的光合能力和生长速率恢复到正常水分条件下生长的植株的水平, 但株高和叶面积没有恢复到对照水平。当玉米再次经受水分亏缺处理时, 与只遭受第二次中度或重度干旱处理的植株相比, 经历过前期中度干旱处理的植株的株高、生物量和光合参数没有显著变化, 但叶面积显著下降; 经历过前期重度干旱处理植株的TrGsCiPnAmax和表观量子效率显著升高, 而株高、叶面积和生物量显著降低。综上所述, 第一次重度干旱处理显著降低玉米叶片的光合能力和生长, 复水可使光合能力和生长速率恢复到正常水分条件下生长植株的水平, 但不能消除前期干旱对生长产生的不利影响。前期中度干旱可以刺激玉米根系的生长和显著提高根冠比, 有利于提高对二次干旱的抵抗能力, 并使总的生物量保持在对照水平, 而前期重度干旱处理虽然在光合作用上能提高植株对二次干旱的抵御能力, 但不能弥补前期干旱处理对生长的不利影响。因此, 在生产实践中, 如果进行抗旱锻炼, 应限制在中度干旱水平, 避免重度干旱。  相似文献

在50只麻醉的大鼠孤离双侧颈动脉窦区,将不同窦内压(ISP)与其对应的平均动脉压(MAP)值进行Logistic五参数曲线拟合,根据所得ISP-MAP关系曲线及其特征参数,观察侧脑室注射(i.c.v)组胺(HA)对颈动脉窦压力感受性反射(CSR)的影响,并对其作用机制进行了初步探讨。结果如下:(1)i.c.v.HA(100ng)导致ISP-MAP关系曲线显著上移,ISP和增益(Gain)关系曲线中部明显下移,反射参数中阈压(TP),饱和压(SP)和最大增益时的窦内压(ISPGmax)值增大,MAP反射变动范围(MAPrange)及反射最大增益(Gmax)减小。(2)预先i.c.v.H1受体拮抗剂氯苯吡胺(CHL,5μg)或H2受体拮抗剂西咪替丁(CIM,15μg),可明显减弱HA的上述效应,CIM的这种减弱作用不如CHL的显著。(3)预先同时i.c.v.CHL和CIM(分别为5和15μg),能完全取消HA的效应。(4)预先向孤束核(NTS)内注射CHL(0.5μg)或CIM(1.5μg),对HA效应的影响与i.c.v.CHL或CIM后的相类似,但NTS内注射CHL或CIM后i.c.v.HA所致的TP变化表现明显下降。以上结果提示,侧脑室给HA使CSR产生快速重调定,反射敏感性下降,功能受抑;其机制是通过中枢HA受体介导,H1受体的作用比H2受体更为明显;下丘脑-NTS的HA能通路可能是HA调节CSR的下行通路之一。NTS处的HA受体在i.c.v.HA抑制CSR的机制中可能发挥重要的作用。  相似文献

A homology model of the extracellular domain of the mGlu3 subtype of metabotropic glutamate (mGlu) receptor was generated and tested using site-directed mutagenesis, a radioligand-binding assay using the Group II selective agonist (2S,2'R,3'R)-2-(2',3'-[3H]dicarboxycyclopropyl) glycine ([3H]DCG-IV), and in a fluorescence-based functional assay in live transiently transfected human embryonic kidney cells. Ten of the 12 mGlu3 mutants (R64A, R68A, Y150A, S151A, T174A, D194A, Y222A, R277A, D301A and K389) showed either no binding or a 90% or greater loss of specific [3H]DCG-IV binding. Several analogous mutations in mGlu2 supported the results obtained with mGlu3. These results demonstrate that the binding of [3H]DCG-IV to mGlu3 is exceptionally sensitive to mutagenesis-induced perturbations. In silico docking of DCG-IV into the agonist binding pocket of mGlu3 facilitated the interpretation the mutagenesis results. Tyrosines 150 and 222, and arginine 277 show close contacts with the third carboxylic acid group in DCG-IV, which is not present in glutamate or (2S,1'S,2'S)-2-(carboxycyclopropyl)glycine (L-CCG-I). Mutation of these three amino acids to alanine resulted in a near complete loss of receptor activation by DCG-IV and retention of near wild-type affinity for L-CCG-I. It is proposed that hydrogen bonding between this carboxylate and tyrosines 150 and 222 and arginine 277 provide a partial explanation for the high affinity and Group II selectivity of DCG-IV. These findings define the essential features of the ligand-binding pocket of mGlu3 and, together with other recent studies on mGlu receptors, provide new opportunities for structure-based drug design.  相似文献

Rationale

The sacred lotus (Nelumbo nucifera) contains many phytochemicals and has a history of human use. To determine which compounds may be responsible for reported psychotropic effects, we used in silico predictions of the identified phytochemicals. Nuciferine, an alkaloid component of Nelumbo nucifera and Nymphaea caerulea, had a predicted molecular profile similar to antipsychotic compounds. Our study characterizes nuciferine using in vitro and in vivo pharmacological assays.

Methods

Nuciferine was first characterized in silico using the similarity ensemble approach, and was followed by further characterization and validation using the Psychoactive Drug Screening Program of the National Institute of Mental Health. Nuciferine was then tested in vivo in the head-twitch response, pre-pulse inhibition, hyperlocomotor activity, and drug discrimination paradigms.

Results

Nuciferine shares a receptor profile similar to aripiprazole-like antipsychotic drugs. Nuciferine was an antagonist at 5-HT2A, 5-HT2C, and 5-HT2B, an inverse agonist at 5-HT7, a partial agonist at D2, D5 and 5-HT6, an agonist at 5-HT1A and D4 receptors, and inhibited the dopamine transporter. In rodent models relevant to antipsychotic drug action, nuciferine blocked head-twitch responses and discriminative stimulus effects of a 5-HT2A agonist, substituted for clozapine discriminative stimulus, enhanced amphetamine induced locomotor activity, inhibited phencyclidine (PCP)-induced locomotor activity, and rescued PCP-induced disruption of prepulse inhibition without induction of catalepsy.

Conclusions

The molecular profile of nuciferine was similar but not identical to that shared with several approved antipsychotic drugs suggesting that nuciferine has atypical antipsychotic-like actions.  相似文献

以‘日本晴’水稻为试验材料,研究铝胁迫对水稻幼苗生长的影响,及铝胁迫下其根系和功能叶片中脂质含量及组分的变化。结果表明Al处理显著抑制了水稻根尖的伸长和生长,降低了植株的生物量以及叶绿素含量。铝胁迫后,半乳糖脂含量变化显著,根和叶中的单半乳糖甘油二酯(MGDG)和双半乳糖甘油二酯(DGDG)含量均降低;而磷脂(PL)及硫代异鼠李糖甘油二酯(SQDG)的含量在根中表现为上升,在叶中则显著降低。此外,根和叶中的脂肪酸不饱和度均降低。由此可见,铝胁迫引起水稻膜脂组分和含量的变化,这种变化可能是铝抑制植物生长的重要原因之一。  相似文献

Objective

Numerous studies have investigated the associations between herpesviruses and chronic periodontitis; however, the results remain controversial. To derive a more precise estimation, a meta-analysis on all available studies was performed to identify the association between herpesviruses and chronic periodontitis.

Methods

A computerized literature search was conducted in December 2014 to identify eligible case-control studies from the PUBMED and EMBASE databases according to inclusion and exclusion criteria. Data were extracted and pooled odds ratios (OR) with 95% confidence intervals (CI) were used to assess the association between herpesviruses and risk of chronic periodontitis. A fixed or random effects model was determined based on a heterogeneity test. Sensitivity analysis was conducted to investigate stability and reliability. Publication bias was investigated using the Begg rank correlation test and Egger''s funnel plot.

Results

Ten eligible studies were included to investigate the association between Epstein–Barr virus (EBV) and chronic periodontitis. The results showed that EBV has a significant association with chronic periodontitis compared with periodontally healthy group (OR = 5.74, 95% CI = 2.53–13.00, P<0.001). The association between human cytomegalovirus (HCMV) and chronic periodontitis was analyzed in 10 studies. The pooled result showed that HCMV also has a significant association with chronic periodontitis (OR = 3.59, 95% CI = 1.41–9.16, P = 0.007). Similar results were found in the sensitivity analyses. No significant publication bias was observed. Two eligible studies were included to investigate the association between herpes simplex virus (HSV) and chronic periodontitis risk. The association between HSV and chronic periodontitis was inconclusive (OR = 2.81 95% CI = 0.95–8.27, P = 0.06). Only one included study investigated the association between human herpesvirus 7 (HHV-7) and chronic periodontitis risk (OR = 1.00, 95% CI = 0.21–4.86).

Conclusion

The findings of this meta-analysis suggest that two members of the herpesvirus family, EBV and HCMV, are significantly associated with chronic periodontitis. There is insufficient evidence to support associations between HSV, HHV-7 and chronic periodontitis.  相似文献

紫薇梨象Pseudorobitis gibbus Redtenbacher是一种严重危害紫薇的新害虫。紫薇梨象在泰安1年发生1代,以幼虫或成虫在被害果实内休眠越冬。越冬幼虫在翌年5月前后化蛹,5月中下旬羽化,6月上中旬进入成虫盛发期,产卵集中在7月下旬—8月中旬,并且虫态交错现象严重。入冬后的蛀果率高达60%以上,对紫薇的观赏价值和种实繁殖构成很大的威胁。紫薇梨象成虫具有假死性和较强的趋光性,耐饥能力一般为36h。成虫对类似于植物蜜源的蜂蜜具有显著趋向性;对类似于食用醋的酸性挥发物质有较强的忌避性。紫薇在园林绿化中的大量应用,将加剧紫薇梨象的地理扩散,对山东周边省份乃至全国的紫薇构成潜在威胁。建议采用物理防治,同时利用天敌及性引诱剂等生物措施来综合防治紫薇梨象。  相似文献

目的:探讨蓝斑α1和α2受体在脑室注射(ICV)组胺(HA)对颈动脉窦反射(CBR)重调定中的作用.方法:孤离麻醉SD大鼠的双侧颈动脉窦区,将不同窦内压(ISP)与其对应的平均动脉压(MAP)值进行Logistic五参数曲线拟合,求得ISP-MAP关系曲线及其特征参数,观察ICV HA以及预先在蓝斑(LC)微量注射α1或α2受体拮抗剂对CBR的影响.结果:ICV HA(60 μmol·L,5μl)导致ISP-MAP关系曲线后半程显著上移(P<0.05),反射参数MAP反射变动范围及反射最大增益减小(P<0.05);预先向LC注射选择性的α1受体拮抗剂酚苄明(PBZ,3μmol·L,500nl)或α2受体拮抗剂育亨宾(YOH,2.5μmol·L-1,500 nl),均能明显加强HA的上述效应,PBZ的这种加强作用不如YOH的显著(P<0.05).结论:脑室给HA使CBR产生快速重调定,反射敏感性下降;LC的α1、α2受体作用可减弱ICV HA对CBR的抑制性重调定;α2受体在调制这种重调定的过程中可能发挥更为重要的作用.  相似文献

在PEG-6000胁迫条件下,以微管蛋白基因为内参基因、水通道蛋白基因ZmPIP1-1和ZmPIP1-2为检测基因,采用半定量逆转录聚合酶链式反应(RT-PCR)体系检测它们在玉米根系中的表达情况。实验结果是:胁迫条件下,ZmPIP1-1的表达量在杂交F,代‘户单4号’(抗旱)和母本‘天四’(抗旱)根系中增多,它的表达量与品种的抗旱性呈正相关,并且胁迫不同时间段它的表达量有差异;而ZmPIP1-2在3个玉米品种的不同水分处理条件下,表达量均没有明显变化。这提示,水分胁迫条件下根系中某些种类的水通道蛋白基因的表达量增多,并且与品种的抗旱性有关;而另一些水通道蛋白基因的表达不受水分亏缺的影响。  相似文献
