Myelin-derived proteins, such as tenascin-R (TN-R), myelin associate glycoprotein (MAG), oligodendrocyte-myelin glycoprotein
(OMgp), and Nogo-A, inhibit the central nervous system regeneration. In this study, the DNA vaccine encoding for oligodendrocyte
and myelin-related antigens was employed to attenuate the axonal growth inhibitory properties of myelin in the setting of
spinal cord injury. Using a rat spinal cord dorsal hemisection model, the vaccine directed against the inhibitory epitopes
of Nogo-A, MAG, OMgp, and TN-R was administered intramuscularly once a week following spinal cord injury, supplemented with
local application of specific anti-sera against the four antigens. Anterograde labeling of dorsal column fibers showed active
axonal regeneration through the lesion site at the eighth week following the treatment in experimental group but not in control
groups. Light microscopic and ultrastructural analysis revealed that vaccination with these myelin-related antigens did not
lead to demyelinating disease. OMgp and TN-R levels were down-regulated at the lesion site together with a parallel increase
in growth-associated protein 43 levels in the treatment groups. This study reveals the effective approach of a DNA vaccine
strategy by attaining the special antibody to direct neutralization of the myelin inhibitors during spinal cord injury. 相似文献

目的:建立一种新的电泳活性染色方法,以快速地对活性电泳中γ-谷氨酰转肽酶(GGT)进行显色,从而准确鉴定和定位该酶,并可用于该酶的电泳法纯化制备。方法:低温下,样品活性电泳结束后立即将浸有γ-L-谷氨酰-α-萘氨和双甘肽混合底物溶液的滤纸贴在胶面上反应5min,然后再用浸有对氨基苯磺酸和亚硝酸钠混合显色液的滤纸覆盖在基质滤纸上显色。结果:在滤纸上很快显示出一条红色条带,经切胶酶促反应检验确定为GGT,经电泳法和高效液相色谱法确定GGT达到了电泳纯。结论:该法快速、灵敏、简单、直观,为GGT的鉴定和纯化提供了一条新思路。 相似文献

One of the several factors that contribute to the low efficiency of mammalian somatic cloning is poor fusion between the small somatic donor cell and the large recipient oocyte. This study was designed to test phytohemagglutinin (PHA) agglutination activity on fusion rate, and subsequent developmental potential of cloned bovine embryos. The toxicity of PHA was established by examining its effects on the development of parthenogenetic bovine oocytes treated with different doses (Experiment 1), and for different durations (Experiment 2). The effective dose and duration of PHA treatment (150 microg/mL, 20 min incubation) was selected and used to compare membrane fusion efficiency and embryo development following somatic cell nuclear transfer (Experiment 3). Cloning with somatic donor fibroblasts versus cumulus cells was also compared, both with and without PHA treatment (150 microg/mL, 20 min). Fusion rate of nuclear donor fibroblasts, after phytohemagglutinin treatment, was increased from 33 to 61% (P < 0.05), and from 59 to 88% (P < 0.05) with cumulus cell nuclear donors. The nuclear transfer (NT) efficiency per oocyte used was improved following PHA treatment, for both fibroblast (13% versus 22%) as well as cumulus cells (17% versus 34%; P < 0.05). The cloned embryos, both with and without PHA treatment, were subjected to vitrification and embryo transfer testing, and resulted in similar survival (approximately 90% hatching) and pregnancy rates (17-25%). Three calves were born following vitrification and embryo transfer of these embryos; two from the PHA-treated group, and one from non-PHA control group. We concluded that PHA treatment significantly improved the fusion efficiency of somatic NT in cattle, and therefore, increased the development of cloned blastocysts. Furthermore, within a determined range of dose and duration, PHA had no detrimental effect on embryo survival post-vitrification, nor on pregnancy or calving rates following embryo transfer. 相似文献

目的 采用活体成像技术比较四种剂量荧光素酶标记肿瘤细胞在小鼠体内生长及肺转移情况,为光学标记肿瘤模型的药物筛选或机制研究提供参考资料.方法 以荧光素酶作为报告基因导人小鼠乳腺癌细胞4T1中,经G418筛选获得稳定表达荧光素酶的细胞克隆并扩大培养.标记细胞稀释成1×107细胞/mL,2×107细胞/mL,5×107细胞/mL和1×108细胞/mL四种剂量,取0.1 mL接种子BALB/c小鼠右侧第二对乳腺脂肪垫内,制作小鼠原位乳腺癌模型,比较肿瘤细胞在小鼠体内生长及肺转移情况.结果获得稳定表达荧光素酶基因的细胞克隆,在致瘤性方面和亲代细胞无明显差别,四种剂量细胞接种BALB/c小鼠后,均有肿瘤生长,接种第28天时,四种剂量接种的原位移植瘤大小没有明显差别,但接种两个高剂量肿瘤细胞的小鼠组各有2只小鼠死亡;接种后31 d,发现四种剂量接种的原位移植瘤均发生不同程度的转移,随着观察天数的增加,转移程度逐渐严重,接种后42 d,小鼠陆续发生死亡.结论 根据转移和死亡情况,确定接种1×106个细胞/只不仅肺转移明显,而且存活时间一般超过45 d,比高剂量接种存活时间长,为最佳肺转移剂量. 相似文献

目的:分析糖尿病急性并发症患者合并横纹肌溶解的临床特征及预后,为临床及时诊断和治疗提供依据。方法:对我院2003年1月~2009年5月住院患者查阅病例资料,糖尿病酮症酸中毒及高血糖高渗状态患者根据肌酸激酶升高与否,分为血清肌酸激酶升高组(A组)和血清肌酸激酶正常组(B组),比较两组临床特征及预后。结果:A、B两组比较,A组较B组血清尿素氮、肌酐、肌红蛋白明显升高,住院时间明显延长,但出院时A组较B组每天每公斤胰岛素剂量减少,差异有统计学意义(P<0.05)。A组中3例患者血清肌酸激酶大于1000U/L,符合横纹肌溶解综合征诊断标准。结论:糖尿病酮酸中毒及高血糖高渗状态患者应重视肌酶检查,早期诊断横纹肌溶解综合征,保护肾脏和胰岛β细胞功能,缩短住院时间。 相似文献

Brain-derived neurotrophic factor (BDNF) binds to its cell surface receptor TrkB to regulate differentiation, development, synaptic plasticity, and functional maintenance of neuronal cells. Binding of BDNF triggers TrkB dimerization and autophosphorylation, which provides docking sites for adaptor proteins to recruit and activate downstream signaling molecules. The molecular mechanisms underlying BDNF-TrkB endocytic trafficking crucial for spatiotemporal control of signaling pathways remain to be elucidated. Here we show that retrolinkin, a transmembrane protein, interacts with endophilin A1 and mediates BDNF-activated TrkB (pTrk) trafficking and signaling in CNS neurons. We find that activated TrkB colocalizes and interacts with the early endosome marker APPL1. Both retrolinkin and endophilin A1 are required for BDNF-induced dendrite development and acute extracellular signal-regulated kinase activation from early endosomes. Suppression of retrolinkin expression not only blocks BDNF-triggered TrkB internalization, but also prevents recruitment of endophilin A1 to pTrk vesicles trafficking through APPL1-positive endosomes. These findings reveal a novel mechanism for BDNF-TrkB to regulate signaling both in time and space through a specific membrane trafficking pathway. 相似文献

目的:观察外加稳恒直流电场对兔腹主动脉周围电势差的影响.方法:在兔腹主动脉两侧腰大肌埋置刺激电极,在血管外膜、血管内外膜间安置测量电极,给予稳恒直流电场刺激,电场强度采用3V或4V,记录电场刺激前后血管周围电势差的变化情况.结果:①通电后即刻,血管周围电势差与通电前相比无变化(P>0.05).②通电后30min,血管外膜间和血管内外膜间电势差均明显增加,4V组增加更为明显.③断电后30min,两外膜间电势差基本恢复至通电前水平,而血管内外膜间电势差仍高于通电前,两刺激电极间仍有较高电势差存在.结论:将铂电极置入兔腰大肌,连接稳恒直流电源后构成的是一个RC电路,应用3V电压或者4V电压刺激都可以引起血管周围电势的变化. 相似文献

Peptide identification of tandem mass spectra by a variety of available search algorithms forms the foundation for much of modern day mass spectrometry-based proteomics. Despite the critical importance of proper evaluation and interpretation of the results generated by these algorithms there is still little consistency in their application or understanding of their similarities and differences. A survey was conducted of four tandem mass spectrometry peptide identification search algorithms, including Mascot, Open Mass Spectrometry Search Algorithm, Sequest, and X! Tandem. The same input data, search parameters, and sequence library were used for the searches. Comparisons were based on commonly used scoring methodologies for each algorithm and on the results of a target-decoy approach to sequence library searching. The results indicated that there is little difference in the output of the algorithms so long as consistent scoring procedures are applied. The results showed that some commonly used scoring procedures may lead to excessive false discovery rates. Finally an alternative method for the determination of an optimal cutoff threshold is proposed. 相似文献

carried out with agarose gel electrophoresis, flow cytometry and transmission electron microscopy. The results indicated that
there were visible apoptosis bodies and typical DNA ladders by genomic DNA fragmentation. The quantitative analysis of apoptosis
by flow cytometry indicated that rHco-gal-m/f peptides induced apoptosis was time and dose dependent. Ultrastructural studies of the PBLCs revealed that a large
number of apoptotic cells were present in galectin-treated cells, which had the typical morphologic changes of apoptosis such
as reduction of the cytoplasmic volume, loss of cell surface microvilli, chromatin condensation and fragmentation of the apoptotic
