Considering the current low level of mechanization for domestic green onion planting and the high labor intensity of artificial planting, a 2ZYX-2 green onion ditching and transplanting machine, which can complete ditching, ridging, transplanting, repression, soil covering and other operations, is designed in this study. The Central Composite test design method was carried out with the speed of the transplanting machine, the depth of the opener and the horizontal position of the opener as the experimental factors and with the qualification ratio of perpendicularity, the variation coefficient of the plant spacing and the qualification ratio of the planting depth as the test index. Through the analysis of the model interaction and response surface, the change laws that the influence the machine’s forward speed, the depth of the opener and the horizontal position of the opener were studied. The regression model was optimized by Design-Expert 8.0.6 software, and the accuracy of the predicted results was verified by experiments. The optimal working parameters showed that the forward speed of the machine was 0.06 m/s, the depth of the opener was 102 mm, and the horizontal position of the opener was 29 mm. Under conditions of optimal working parameters, the qualification rate of the verticality was 86.83%, the coefficient of variation for the plant spacing was 2.77, and the pass rate of planting depth was 88.26%. The research related to the thesis can provide a reference for the mechanized planting of green onion, which is of great significance to the cost-effectiveness of the green onion industry.  相似文献

突变p53 (mutant p53, Mut-53)聚集体的形成是p53突变后使原本包裹在其疏水核心内部的黏附序列暴露,黏附序列迅速成核组装,形成无定形的原纤维. Mut-p53聚集体不仅可以以显性负效应(dominant-negative effect,DN)的方式使野生型p53 (wild type p53,Wt-p53)失活,还表现出功能获得(gain-of-function,GOF)特性,促进肿瘤的发生和发展.在卵巢癌、结肠癌、前列腺癌等多种肿瘤细胞中均发现了Mut-p53的异常聚集,其与肿瘤的转移、耐药和预后不良具有显著的相关性.因此,p53聚集是逆转化疗耐药及肿瘤治疗的潜在靶点.设计和发现靶向Mut-p53聚集体的小分子化合物,抑制p53疏水核心内部黏附序列的暴露,恢复p53的功能从而发挥抗肿瘤作用成为了当今研究热点.本文就p53聚集体对肿瘤发生发展的影响及目前靶向Mut-p53聚集体的研究策略进行了综述.  相似文献

Asthma is a common chronic inflammatory disease characterized by the infiltration and accumulation of memory-like Th2 cells and eosinophils. Viral infection has emerged as the most common cause of severe episodes of asthma. For the treatment of bronchial asthma, the root of liquorice (Glycyrrhiza glabra) has been used as a traditional medicine in the East and West. Licochalcone A is the predominant, characteristic chalcone in liquorice root. To determine whether licochalcone A possesses an anti-inflammatory effect, we tested its effect on the expression and production of thymic stromal lymphopoietin (TSLP) in BEAS 2B cells and primary bronchial epithelial cells. We found that polyinosinic-polycytidylic acid (poly-IC)-induced TSLP expression was suppressed by treatment with licochalcone A in a dose- and time-dependent manner. We also found that poly-IC-induced mRNA expression of other proinflammatory mediators such as MCP-1, RANTES, and IL-8 was suppressed by licochalcone A. Furthermore, licochalcone A suppressed poly-IC-induced nuclear factor kappa B (NF-κB) nuclear translocation and DNA-binding activity by suppressing the Iκβ kinase (IKK) activity but not by direct phosphorylation of p65 at serine 276. Collectively, our findings suggest that licochalcone A suppresses poly-IC-induced TSLP expression and production by inhibiting the IKK/NF-κB signaling pathway, which might be involved in the pathogenesis of virus-exacerbated asthma. Further elucidation of the mechanisms underlying these observations can help develop therapeutic strategies for virally induced asthma.  相似文献

Tissue engineering involves ex vivo seeding of anchorage-dependent mammalian cells onto scaffolds, or transplanting cells in vivo. The cell expansion currently requires repeated cell detachment from solid substrata by enzymatic, chemical or mechanical means. The report here presents a high yield three-dimensional culture and harvest system circumventing the conventional detachment requirements. Cells mixed with dilute cationic collagen were microencapsulated within an ultra-thin shell of synthetic polymers. The cationic collagen could rapidly form a conformal layer of collagen fibers around cells to support cell proliferation and functions. The collagen could be readily removed from cells with a buffer rinse after harvesting from the fragile microcapsules. The cells harvested from this system demonstrate improved attachment, morphology and functions over conventionally cultured cells, upon binding to ligand-conjugated polymer surfaces. The harvested cells can be re-encapsulated and allowed to proliferate again, or used immediately in applications.  相似文献

Predicting the levels of chlorophyll-a (Chl-a) is a vital component of water quality management, which ensures that urban drinking water is safe from harmful algal blooms. This study developed a model to predict Chl-a levels in the Yuqiao Reservoir (Tianjin, China) biweekly using water quality and meteorological data from 1999-2012. First, six artificial neural networks (ANNs) and two non-ANN methods (principal component analysis and the support vector regression model) were compared to determine the appropriate training principle. Subsequently, three predictors with different input variables were developed to examine the feasibility of incorporating meteorological factors into Chl-a prediction, which usually only uses water quality data. Finally, a sensitivity analysis was performed to examine how the Chl-a predictor reacts to changes in input variables. The results were as follows: first, ANN is a powerful predictive alternative to the traditional modeling techniques used for Chl-a prediction. The back program (BP) model yields slightly better results than all other ANNs, with the normalized mean square error (NMSE), the correlation coefficient (Corr), and the Nash-Sutcliffe coefficient of efficiency (NSE) at 0.003 mg/l, 0.880 and 0.754, respectively, in the testing period. Second, the incorporation of meteorological data greatly improved Chl-a prediction compared to models solely using water quality factors or meteorological data; the correlation coefficient increased from 0.574-0.686 to 0.880 when meteorological data were included. Finally, the Chl-a predictor is more sensitive to air pressure and pH compared to other water quality and meteorological variables.  相似文献
