A new hadrosaurid dinosaur, Arenysaurus ardevoli gen. et sp. nov., from the Late Maastrichtian of Aren (Huesca, South-central Pyrenees) is described on the basis of a partial, articulated skull, mandibular remains and postcranial elements, including vertebrae, girdle and limb bones. Arenysaurus is characterized by having a very prominent frontal dome; nearly vertical prequadratic (squamosal) and jugal (postorbital) processes, and deltopectoral crest of the humerus oriented anteriorly. Moreover, it possesses a unique combination of characters: short frontal (length/width approximately 0.5); midline ridge of parietal at level of the postorbital-squamosal bar; parietal excluded from the occiput; squamosal low above the cotyloid cavity. A phylogenetical analysis indicates that Arenysaurus is a rather basal member of Lambeosaurinae and the sister-taxon to Amurosaurus and the Corythosaurini-Parasaurolophini clade. The phylogenetic and biogeographical relationships of Arenysaurus and other lambeosaurines suggest a palaeogeographical connection between Asia and Europe during the Late Cretaceous.  相似文献

Background

Aberrant promoter DNA methylation has been shown to play a role in acute myeloid leukemia (AML) pathophysiology. However, further studies to discuss the prognostic value and the relationship of the epigenetic signatures with defined genomic rearrangements in acute myeloid leukemia are required.

Methodology/Principal Findings

We carried out high-throughput methylation profiling on 116 de novo AML cases and we validated the significant biomarkers in an independent cohort of 244 AML cases. Methylation signatures were associated with the presence of a specific cytogenetic status. In normal karyotype cases, aberrant methylation of the promoter of DBC1 was validated as a predictor of the disease-free and overall survival. Furthermore, DBC1 expression was significantly silenced in the aberrantly methylated samples. Patients with chromosome rearrangements showed distinct methylation signatures. To establish the role of fusion proteins in the epigenetic profiles, 20 additional samples of human hematopoietic stem/progenitor cells (HSPC) transduced with common fusion genes were studied and compared with patient samples carrying the same rearrangements. The presence of MLL rearrangements in HSPC induced the methylation profile observed in the MLL-positive primary samples. In contrast, fusion genes such as AML1/ETO or CBFB/MYH11 failed to reproduce the epigenetic signature observed in the patients.

Conclusions/Significance

Our study provides a comprehensive epigenetic profiling of AML, identifies new clinical markers for cases with a normal karyotype, and reveals relevant biological information related to the role of fusion proteins on the methylation signature.  相似文献

Egg and faecal pellet production rates, and their functional response to food and temperature, were measured for the Antarctic copepod Metridia gerlachei during January 1996. The study area comprised the Gerlache and Bransfield Straits and Drake Passage. The highest rates of “in situ” egg and faecal pellet production were observed in Gerlache stations, coinciding with chlorophyll a concentrations approaching food saturation levels. In the Bransfield and Drake stations, with very low chlorophyll concentrations, the rates of egg production were either very low, or no eggs were produced. Egg production rates, although well correlated with “in situ” chlorophyll values, appeared to be independent of food concentration on a short time-scale (24-h incubations), while the production of faecal pellets was closely related to food abundance in the same experiments. In general, the rates of egg production were low, even at food saturation, with a very high individual variability. Although in the majority of Gerlache stations about 50% of total chlorophyll a corresponded to the phytoplankton size-fraction >10 μm, M. gerlachei feeds preferentially on the <10-μm fraction. Temperature had no clear effects on egg production rate but had a significant effect on pellet production rates, with maximum values at 2.5°C. These features appear to agree with the reproductive strategy attributed to the species, based on a relatively low rate of egg production extended over a long reproductive period. Recent fieldwork in the Late Eocene of Zambrana (Álava, Basque-Cantabrian Region) has yielded a new species of Pachynolophus: P. zambranensis nov. sp. According to the phylogenetic analysis made in this study, the Zambrana species is part of the monophyletic Pachynolophus, which is defined at least by three synapomorphies: significant progressive increase of the size in the M1/1-M3/3 series, being M3/3 outstandingly bigger than M2/2; moderate increase of the relative surface of M3/ with respect to M2/ surface, and weak labial cingula in the lower series. Pachynolophus includes the species P. duvali, P. livinierensis, P. garimondi, P. lavocati, P. zambranensis, P. boixedatensis and P. cesserasicus. P. molipotensis and P. cayluxi are not represented by sufficient material to attribute them to Pachynolophus. P. bretovensis does not share a synapomorphy of Pachynolophus and P. hookeri should not be referred to this genus. P. zambranensis shares several synapomorphies with P. garimondi and P. lavocati, which are: the posprotocrista of the P3-4/ series is a well-defined lophe; the P4/ has a subquadrangular outline and shows a strong posterolingual cingulum, forming a posterior widening; and there is an “incipient mesostyle” (pseudomesostyle?) in the upper molars. The species P. zambranensis is mainly characterized by lophodont dentition, with the postprotocrista obliquely oriented to the ectoloph in the P3-4/ series, and a “pseudomesostyle” also present in the P3-4/ series. It represents the first mention of the genus Pachynolophus described in the Late Eocene of the Iberian Peninsula.  相似文献

The ligand binding module five (LA5) of the low density lipoprotein receptor is a small, single-domain protein of 40 residues and three disulfide bonds with a calcium binding motif that is essential for its structure and function. Several mutations in LA5 have been reported to cause familial hypercholesterolemia by impairing a proper folding of the module. The current study reports the oxidative folding and reductive unfolding pathways of wild type and mutant LA5 modules through kinetic and structural analysis of the trapped intermediates. Wild type LA5 folding involves an initial phase of nonspecific packing where the sequential oxidation of its cysteines gives rise to complex equilibrated populations of intermediates. In the presence of calcium, the attainment of a coordination-competent conformation becomes the rate-limiting step of folding while binding of the ion funnels both thermodynamically and kinetically the folding reaction toward the native state. In the absence of calcium, a scrambled isomer (termed Xa) constitutes the global free energy minimum of the folding process. Xa and the native form are stable, inter-convertible species whose relative populations at equilibrium appear displaced in disease-linked mutants toward the scrambled form. Because stable scrambled isomers such as Xa avoid the exposition of reactive cysteines in misfolded modules, they might constitute a strategy to prevent wrong interactions with other domains during folding of the receptor. Comparison of the folding pathways of wild type and mutant LA5 provides the molecular basis to understand how LA modules fold into a functional conformation or upon mutation misfold and lead to disease.  相似文献

Polychlorinated biphenyls (PCBs) are persistent environmental contaminants that can induce inflammatory processes in the vascular endothelium. We hypothesize that the plasma membrane microdomains called caveolae are critical in endothelial activation and toxicity induced by PCBs. Caveolae are particularly abundant in endothelial cells and play a major role in endothelial trafficking and the regulation of signaling pathways associated with the pathology of vascular diseases. We focused on the role of caveolae and their major protein component, caveolin-1 (Cav-1), on aryl hydrocarbon receptor (AhR)-mediated induction of cytochrome P450 1A1 (CYP1A1) by coplanar PCBs. Endothelial cell exposure to PCB77 increased both caveolin-1 and CYP1A1 levels in a time-dependent manner in total cell lysates, with a maximum increase at 6h. Furthermore, PCB77 accumulated mainly in the caveolae-rich fraction, as determined by gas chromatograph-mass spectrometry. Immunoprecipitation analysis revealed that PCB77 increased AhR binding to caveolin-1. Silencing of caveolin-1 significantly attenuated PCB77-mediated induction of CYP1A1 and oxidative stress. Similar effects were observed in caveolin-1 null mice treated with PCB77. These data suggest that caveolae may play a role in regulating vascular toxicity induced by persistent environmental pollutants such as coplanar PCBs. This may have implications in understanding mechanisms of inflammatory diseases induced by environmental pollutants.  相似文献

This work is focused on the evaluation of a beta-cyclodextrin polymer as a carrier medium in a fluidized bed bioreactor treating aqueous phenol as a model pollutant. The insoluble polymer support was obtained in the shape of spherical beads by crosslinking beta-cyclodextrin with epichlorohydrin. A batch of swollen polymer particles was loaded into the reactor and inoculated with a mixed bacterial culture. Bacterial growth on the polymer beads was initially stimulated by glucose addition to the medium, and then gradually replaced with phenol. The operational variables studied after the acclimation period included phenol load, hydraulic residence time and recirculation flow rate. Low hydraulic residence times and moderate phenol loads were applied. The elimination capacity was usually about 1.0 kg-phenol/m(3)d, although a maximum of 2.8 kg-phenol/m(3)d was achieved with a retention time of only 0.55 h. The depuration efficiency was not affected by the recirculation flow rate in the range studied. Neither operational nor support stability problems were detected during the operation. A high degree of expansion was achieved in the bioreactor due to the hydrogel nature of the cyclodextrin polymer and, consequently, a low energy requirement was necessary to fluidize the bed.  相似文献

BACKGROUND AND AIMS: Broomrapes (Orobanche spp.) are holoparasitic weeds that cause devastating losses in many economically important crops. The molecular mechanisms that control early stages of host infection in Orobanche are poorly understood, partly due to the lack of experimentally tractable in vitro systems that allow the efficient application of molecular tools. Here an improved axenic system for the analysis of pre-infection stages in O. ramosa in the absence of the host plant is described. METHODS: An optimized protocol for seed disinfection, based on formaldehyde, was developed. Orobanche ramosa seeds were conditioned in Petri dishes with filter paper, stimulated by addition of the synthetic strigol analogue GR24, and the percentage of germination as well as attachment-organ formation was determined. KEY RESULTS: Treatment of O. ramosa seeds with tobacco-root exudate or with GR24 resulted in highly reproducible germination rates around 70 %. A conditioning period of 8 d was both necessary and sufficient to allow optimal germination in response to GR24. Conditioned seeds that were dehydrated for several months remained fully responsive to GR24 without the need of a new conditioning period. Treatments as short as 5 min with GR24 were sufficient to fully and irreversibly induce the seed germination response. Approximately half of the germinated seeds initiated attachment-organ development. Similar rates of attachment organ induction were also detected in the rare cases of seeds that had germinated spontaneously on water. CONCLUSIONS: The results suggest that the conditioning period produces persistent changes in the seeds required for responsiveness to external stimulants. The rapid action of GR24 suggests that it may act via a receptor-mediated signalling mechanism. While germination in O. ramosa is induced by exogenous stimuli, attachment organ differentiation appears to be triggered by unknown endogenous signals. The new in vitro culture system will have useful applications for the molecular analysis of early stages of parasitic development in Orobanche.  相似文献

The Lower Miocene deposits of the Bardenas Reales of Navarre (NW Ebro Basin, northern Iberian Peninsula) have yielded a diverse vertebrate fauna, including remains of amphibians and reptiles. These remains occur in several localities in the Tudela Formation. The fossiliferous levels belong to the Biozones MN2b-3 (Biozones Z-A of the Ramblian, i.e., Late Aquitanian to Early Burdigalian in age). The amphibians and reptiles represent at least 13 out of 37 vertebrate species. Amphibians consist of a salamandrid urodele and two or three anurans. All the turtles are cryptodirans and consist of the chelydrid Chelydropsis apellanizi, the testudinids Ptychogaster (Temnoclemmys) bardenensis and Ptychogaster ronheimensis, and a Trionychinae indet. Squamates are represented by the anguid lizard Ophisaurus sp., a non-anguid lacertilian, an amphisbaenian, the erycine boid? Eryx sp., and indeterminate colubrids. Crocodilian remains are assigned to the basal alligatoroid Diplocynodon sp. The fossil associations of the Bardenas Reales of Navarre suggest that the vertebrates lived in the centre of an endoreic basin with stretches of water under intertropical to subtropical climatic conditions.  相似文献

Behaviour has direct links to wing morphology in bird species. Many studies have postulated migration to be one of the most important forces of selection acting on wing morphology, particularly in relation to wing pointedness. Studies in passerines have found that adults have longer and more pointed wings than juveniles, especially in migratory species. We analysed differences in wing morphology between age groups of the European Turtle Dove, a non‐passerine migratory species that benefits from rounded wings during their daily activity, due to its ground‐feeding behaviour and acrobatic flight style. Our results show that adults of this species have longer but more rounded wings than juveniles. This suggests that in this species wing morphology in juveniles is selected to facilitate the first migration, whereas other selection forces (e.g. flight manoeuvrability) are more important after the first moult. These data also explain why juveniles are not as adept at escaping from predators or hunters as adults.  相似文献
