Lomentospora (Scedosporium) prolificans is an opportunistic pathogen capable of causing invasive infections in immunocompromised patients. The fungus is able to disseminate via the bloodstream finally arriving at the central nervous system producing neurological symptoms and, in many cases, patient death. In this context, microglial cells, which are the resident immune cells in the central nervous system, may play an important role in these infections. However, this aspect of anti‐L. prolificans immunity has been poorly researched to date. Thus, the interactions and activity of microglial cells against L. prolificans were analysed, and the results show that there was a remarkable impairment in their performance regarding phagocytosis, the development of oxidative burst, and in the production of pro‐inflammatory cytokines, compared with macrophages. Interestingly, L. prolificans displays great growth also when challenged with immune cells, even when inside them. We also proved that microglial phagocytosis of the fungus is highly dependent on mannose receptor and especially on dectin‐1. Taken together, these data provide evidence for an impaired microglial response against L. prolificans and contribute to understanding the pathobiology of its neurotropism.  相似文献

Although the Bothremydidae are the most abundant and diverse group of turtles in the Upper Cretaceous fauna of Europe, the record attributed to the genus Elochelys was, until now, quite scarce. However, the revision of the specimens previously attributed to this taxon and the analysis of new material from the Spanish record significantly increase our knowledge of these turtles. Evaluation of the morphological variability, affecting many characters that have been considered diagnostic until now and comparisons with other members of the node Bothremydini, allow us to amend the diagnosis of the genus Elochelys, recognising Elochelys perfecta as a valid single species and reassigning its other representative, “Elochelys” convenarum, to the new genus Iberoccitanemys.  相似文献

The measurement of copepod adult female fecundity as an estimateof adult growth (production) rates has been an approach widelyused in both laboratory and field studies. Here we demonstratethat the basic assumption of this approach, i.e. that estimatedegg production during the incubation reflects the ‘insitu’ conditions, is not always legitimate and that itis temperature dependent. The enhancement of metabolic ratesinduced by high temperatures induces a more accented bias ofthe method. We also provide guidelines on how to conduct theincubations in order to obtain more reliable estimates.  相似文献

The use of 1,4-disubstituted 1,2,3-triazoles as trans-amide bond surrogates has become an important tool for the synthesis of metabolically stabilized peptidomimetics. These heterocyclic bioisosters are generally incorporated into the peptide backbone by applying a diazo-transfer reaction followed by CuAAC (click chemistry) with an α-amino alkyne. Even though the manual synthesis of backbone-modified triazolo-peptidomimetics has been reported by us and others, no procedure has yet been described for an automated synthesis using peptide synthesizers. In order to efficiently adapt these reactions to an automated setup, different conditions were explored, putting special emphasis on the required long-term stability of both the diazo-transfer reagent and the Cu(I) catalyst in solution. ISA·HCl is the reagent of choice to accomplish the diazo-transfer reaction; however, it was found instable in DMF, the most commonly used solvent for SPPS. Thus, an aqueous solution of ISA·HCl was used to prevent its degradation over time, and the composition in the final diazo-transfer reaction was adjusted to preserve suitable swelling conditions of the resins applied. The CuAAC reaction was performed without difficulties using [Cu (CH3CN)4]PF6 as a catalyst and TBTA as a stabilizer to prevent oxidation to Cu(II). The optimized automated two-step procedure was applied to the synthesis of structurally diverse triazolo-peptidomimetics to demonstrate the versatility of the developed methodology. Under the optimized conditions, five triazolo-peptidomimetics (8–5 amino acid residues) were synthesized efficiently using two different resins. Analysis of the crude products by HPLC-MS revealed moderate to good purities of the desired triazolo-peptidomimetics (70–85%). The synthesis time ranged between 9 and 12.5 h.  相似文献

The spring phytoplankton composition has been investigated ata 50 m deep station off Plymouth in the English Channel for6 years (1993–1999). The percentage of diatoms duringthe spring bloom was significantly correlated with the NorthAtlantic Oscillation index. A similar relationship between phytoplanktonand North Atlantic Oscillation has also been found in a Swedishlake, suggesting a possible link between atmospheric forcingand phytoplankton composition.  相似文献

Background  

Horizontal transfer (HT) could play an important role in the long-term persistence of transposable elements (TEs) because it provides them with the possibility to avoid the checking effects of host-silencing mechanisms and natural selection, which would eventually drive their elimination from the genome. However, despite the increasing evidence for HT of TEs, its rate of occurrence among the TE pools of model eukaryotic organisms is still unknown.  相似文献

A conceptual model of the possible role of lipids in the lifecycle of Calanus finmarchicus is proposed. Lipid storage couldplay a key role at various levels: (i) by triggering diapausethrough variations in cholesterol and fatty acid derived hormonelevels; (ii) by determining the overwintering depth in relationto the convective mixed layer; and (iii) by playing an importantrole in the population adaptation to the hydrological conditionsof the basin. The protein family known as synucleins is composed of α-, β- and γ-synuclein. The most widely studied is the α-synuclein protein due to its participation in essential processes of the central nervous system. Neurotoxicity of this protein is related to the presence of multiplications (duplications and triplications) and point mutations in the gene sequence of the α-synuclein gene (SNCA), differential expression of its isoforms and variations in post-transductional modifications. Neurotoxicity is also related to cytoplasmic inclusions known as Lewy bodies (LBs) and Lewy neurites (LNs), which are also present in α-synucleinopathies. In general, the β-synuclein protein, codified by the SNCB gene, acts as a regulator of processes triggered by α-synuclein and its function is altered by variations in the gene sequence, while γ-synuclein, codified by the SNCG gene, seems to play a major role in certain tumoral processes.  相似文献
