本实验选择具有优良肉质的本地金华猪与肉质较差的引进品种皮特兰猪杂交所产的金皮F2代为研究对象,利用PCR-RFLP的方法检测金皮F2代猪CAST基因型的频率,并以肌球蛋白重链(MHC)免疫组织化学、过碘酸希夫反应和苏木精-伊红等多种组织学方法,研究猪背腰最长肌的肌肉组织学特性,并对其进行图像分析,在此基础上采用SAS分析其相互关系。结果发现.PAS染色后肌纤维可区分为三种类型。PAS阴性(Ⅰ)、PAS反应强阳性(Ⅱ-a)和PAS反应强度中间型(Ⅱ-b)。利用肌球蛋白重链单克隆抗体MHCs和MHCf染色结果表明.金皮F2代杂交猪背腰最长肌MHCs阳性(慢肌)纤维的比例为7.62%,快肌纤维为92.38%。CAST基因的扩增产物具有524bp和632bp两个Hinfl多态性片段,定义等位基因为A(114+192+400+632bp),B(114+192+400+524bp)。AA、BB和AB基因型频率分别为0.1795、0.5897和0.2308,A和B的基因型频率分别为0.4743和0.5256。AA、BB和AB单型对眼肌面积.系水率、pH值、导电率等参数均无显著的影响。CAST基因的HinfI多态性对肌纤维的轴比有显著影响,AA单型有产生较多轴比较大(近似椭圆形)肌纤维的倾向.而BB则控制形成更多的轴比更小(近似于圆形)的肌纤维。具有AA单型的个体具有更多的肌间结缔组织.而具有BB单型的个体的肌间结缔组织较少.AB单型的个体介于两者之间。 相似文献

Fluorescence properties of Early Cambrian acritarchs were investigated using Leica das Mikroskop (DM)microscopy with a mercury lamp.Well-preserved autoflurescence properties show a correlation between acritarchs morphology and the intensity of emitted fluorescence.In accordance with the fluorescence intensity of organic cell walls,two groups ofmicrofossils were distinguished.Results of observation in this study,which are consistent with those of the previous foreign studies,are in good agreement with regular difference in autofluorescence intensity among palynomorphs reported by McPhilemy (1998).Spores and algae,including Botryococcus,have very bright fluorescence while acritarchs often show less intense fluorescence.Dark brown microfossils have been reworked,and have little or no fluorescence. 相似文献

This paper investigated the correlation between polymorphisms and haplotypes in the apolipoprotein B (apoB) gene (SP-I/D, XbaI-RFLP, VNTR) and natural longevity persons among the Uygur people in Xin-jiang. For this purpose, 191 healthy Uygur individuals aged above 90 from Hetian area of Xinjiang were recruited, and another 53 persons aged 65—70 from the same nationality, the same region and with the same gender ratio, served as the control group. Genotyping was performed by PCR-SSP, PCR-RFLP and PCR-sequencing methods. Logistic regression analyses revealed that the frequencies of X X genotype, M and L alleles and the genetypes composed of M and L were significantly higher in the longevity group than in the control group. In haplotype analyses, we found that, in the long-lived people, the frequency of haplotypes composed of the X and M alleles was significantly higher whereas the frequency of haplotypes composed of the X- and S alleles was significantly lower (both P<0.05) I than those of their controls. These results indicated that the S allele, SS genotype and X -S, D-S, D-X -S haplotypes were the possible adverse factors, whereas the M, L alleles, X X , MM, ML, LL genotypes and I-X -M, X -M haplotypes were the possibe protective factors for the naturally long-lived Uygur people in China. 相似文献

IgG hinge region peptide bonds are susceptible to degradation by hydrolysis. To study the effect of Fab and Fc on hinge region peptide bond hydrolysis, a recombinant humanized monoclonal IgG1 antibody, its F(ab')2 fragment, and a model peptide with amino acid sequence corresponding to the hinge region were incubated at 40 degrees C in formulation buffer including complete protease inhibitor and EDTA for 0, 2, 4, 6 and 8 weeks. Two major cleavage sites were identified in the hinge region of the intact recombinant humanized monoclonal antibody and its F(ab')2 fragment, but only one major cleavage site of the model peptide was identified. Hinge region peptide bond hydrolysis of the intact antibody and its F(ab')2 fragment degraded at comparable rates, while the model peptide degraded much faster. It was concluded that Fab region of the IgG, but not Fc portion had significant effect on preventing peptide bond cleavage by direct hydrolysis. Hydrolysis of hinge region peptide bonds was accelerated under both acidic and basic conditions. 相似文献

Fishmeal wastewater, a seafood processing waste, was utilized for production of lactic acid and fungal biomass by Rhizopus oryzae AS 3.254 with the addition of sugars. The 30 g/l exogenous glucose in fishmeal wastewater was superior to starch in view
of productivities of lactic acid and fungal biomass, and COD reduction. Fishmeal wastewater can be a replacement for peptone
which was the most suitable nitrogen source for lactic acid production among the tested organic or inorganic nitrogen sources.
Exogenous NaCl (12 g/l) completely inhibited the production of lactic acid and fungal growth. In the medium of COD 5,000 mg/l
fishmeal wastewater with the addition of 30 g/l glucose, the maximum productivity of lactic acid was 0.723 g/l h corresponding
to productivity of fungal biomass 0.0925 g/l h, COD reduction 84.9% and total nitrogen removal 50.3% at a fermentation time

用含有不同浓度(0~400μmol/L)Cd(NO3)2的Hoagland营养液处理砂培的菊芋。处理50d后,测定植物体内镉积累量以及过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,并对POD同工酶进行电泳分析。发现在Cd50~100μmol/L浓度内,随着镉浓度的升高,菊芋根和叶中镉的积累量显著增加,而随后积累量的增加有所减少。根和叶中MDA含量显著上升,说明镉引起了膜脂过氧化。0~100μmol/LCd处理,根和叶中POD活性随Cd浓度增加而增强,而在200~400μmol/LCd处理下有所减弱。根和叶SOD活性在50~200μmol/LCd处理下随Cd浓度增加而增强,而在400μmol/LCd处理下SOD活性明显受到抑制。根和叶CAT活性随Cd浓度升高而增强。电泳结果显示,POD同工酶变化明显,镉诱导出一条新酶带LP10。菊芋POD同工酶可以作为镉污染的土壤的生物指示剂。 相似文献

摘 要 探讨人羊膜间充质细胞(human amniotic mesenchymal cells,hAMCs)向心肌细胞分化的能力。采用胶原酶消化法分离hAMCs,用流式细胞仪进行表型鉴定;用5-氮杂胞苷和碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)诱导hAMCs向心肌细胞分化,免疫荧光染色法检测诱导后细胞中特异蛋白结蛋白和α-辅肌动蛋白的表达,RT-PCR检测心肌特异性转录因子Nkx2.5 、GATA-4和心肌特异性收缩蛋白α-肌球蛋白重链(α-myosin heavy chain,α-MHC)mRNA的表达。结果显示:①hAMCs原代培养至第6 d,贴壁细胞汇合度可达80%,呈漩涡状生长。传代后hAMCs增殖迅速,3~4 d细胞汇合度可达100%,细胞呈梭形或多角形。②hAMCs表达CD44和波形蛋白,不表达CK19。③hAMCs经诱导分化8~10 d后细胞排列紧密,多为长梭形。③hAMCs诱导2 w和4 w表达α-辅肌动蛋白和心肌特异性转录因子Nkx2.5。④诱导前后的hAMCs均表达结蛋白和GATA-4,但均未见α-MHC表达。说明hAMCs具有向心肌样细胞分化的能力,可望成为细胞心肌成形术(cellular cardiomyoplasty,CCM)的候选细胞。 相似文献

经RT-PCR扩增了H5N1亚型禽流感病毒血凝素基因(HA)片断,限制性内切酶酶切后将其克隆到pFastBacHTA杆状病毒转座载体,经酶切鉴定及测序,筛选出阳性重组转座载体pFastBac-H5。将pFastBacH5转化含有杆状病毒穿梭载体(bacmid)的DH10Bac感受态细胞,通过蓝白斑筛选和PCR鉴定获得重组杆状病毒穿梭载体rBacmid-H5。rBacmid-H5在脂质体介导下转染sf9昆虫细胞,SDS-PAGE蛋白电泳、Western blot、血凝试验和血凝抑制试验分析表明:分子量约63Kd重组血凝素蛋白(rH5)在sf9昆虫细胞中实现了高效表达。rH5具有血凝活性,而且其血凝活性能够被H5N1禽流感病毒高免血清所抑制;rH5免疫鸡诱导产生针对H5N1禽流感病毒亚型特异的血凝抑制抗体,说明表达的重组蛋白具有与天然蛋白相似的生物活性。 相似文献
